Rapid spread of a densovirus in a major crop pest following wide-scale adoption of Bt-cotton in China.

ABSTRACT: Bacillus thuringiensis (Bt) crops have been widely planted and the effects of Bt-crops on populations of the target and non-target insect pests have been well studied. However, the effects of Bt-crops exposure on microorganisms that interact with crop pests have not previously been quantified. Here, we use laboratory and field data to show that infection of Helicoverpa armigera with a densovirus (HaDV2) is associated with its enhanced growth and tolerance to Bt-cotton. Moreover, field monitoring showed a much higher incidence of cotton bollworm infection with HaDV2 in regions cultivated with Bt-cotton than in regions without it, with the rate of densovirus infection increasing with increasing use of Bt-cotton. RNA-seq suggested tolerance to both baculovirus and Cry1Ac were enhanced via the immune-related pathways. These findings suggest that exposure to Bt-crops has selected for beneficial interactions between the target pest and a mutualistic microorganism that enhances its performance on Bt-crops under field conditions.

Project description:The current study was directed to investigate the effect of temperature and humidity on insect pest complex of cotton. This study was carried out on the farmer field of a farmer of MouzaMehraywala tehsil and district Rajanpur southern part of the Punjab, Pakistan (29.1044° N, 70.3301° E) in the month of May, 2019 to November 2019. The climatic conditions of the study site were; a Rajanpur lie on 96m above sea level Rajanpur has a desert climate. During the year, there is virtually no rainfall. The average temperature in Rajanpur is 26.0°C | 78.8°F. Precipitation here is about 205 mm / 8.1 inch per year. Our results indicate that's the correlation of temperature (minimum and maximum), humidity (minimum and maximum), rainfall and weather condition (clear or cloud), it is clear that except temperature there is a positive correlation of jassid population. While among the relation of jassid with different factor there is very weak relation with jassid population regarding temperature (-0.001), the relation of humidity, rainfall and sky condition with jassid population is positive and moderate (0.520, 0.668 & 0.575 respectively), while the relation of jassid population among these factor is significant except temperature. The results indicate that's the correlation of temperature, humidity, rainfall and weather condition, it is clear that there is positive correlation of thrips population. While among the relation of thrips with different factor there is very weak relation of thrips population with temperature (.103), the relation of humidity, rainfall and sky condition with thrips population is positive and moderate (.515.751& .577 respectively), while the relation thrips population among these factor is significant except temperature. The results indicate that's correlation of temperature, humidity, rainfall and weather condition, it is clear that there is positive correlation of Whitefly population. While among the relation of Whitefly with different factors there is very moderate relation of Whitefly population with temperature (.076), the relation of humidity, rainfall and sky condition with Whitefly population is negative and moderate (-.051.-.368 & -.559 respectively), while the relation Whitefly population among these factor is significant except temperature.

Project description:Drought stress impacts cotton plant growth and productivity across countries. Plants can initiate morphological, cellular, and proteomic changes to adapt to unfavorable conditions. However, our knowledge of how cotton plants respond to drought stress at the proteome level is limited. Herein, we elucidated the molecular coordination underlining the drought tolerance of two inbred cotton varieties, Bacillus thuringiensis-cotton [Bt-cotton + Cry1 Ac gene and Cry 2 Ab gene; NCS BG II BT (BTCS/BTDS)] and Hybrid cotton variety [Non-Bt-cotton; (HCS/HDS)]. Our morphological observations and biochemical experiments showed a different tolerance level between two inbred lines to drought stress. Our proteomic analysis using 2D-DIGE revealed that the changes among them were not obviously in respect to their controls apart from under drought stress, illustrating the differential expression of 509 and 337 proteins in BTDS and HDS compared to their controls. Among these, we identified eight sets of differentially expressed proteins (DEPs) and characterized them using MALDI-TOF/TOF mass spectrometry. Furthermore, the quantitative real-time PCR analysis was carried out with the identified drought-related proteins and confirmed differential expressions. In silico analysis of DEPs using Cytoscape network finds ATPB, NAT9, ERD, LEA, and EMB2001 to be functionally correlative to various drought-responsive genes LEA, AP2/ERF, WRKY, and NAC. These proteins play a vital role in transcriptomic regulation under stress conditions. The higher drought response in Bt cotton (BTCS/BTDS) attributed to the overexpression of photosynthetic proteins enhanced lipid metabolism, increased cellular detoxification and activation chaperones, and reduced synthesis of unwanted proteins. Thus, the Bt variety had enhanced photosynthesis, elevated water retention potential, balanced leaf stomata ultrastructure, and substantially increased antioxidant activity than the Non-Bt cotton. Our results may aid breeders and provide further insights into developing new drought-tolerant and high-yielding cotton hybrid varieties.
