Project description:Oscillatory neural activities are prevalent in the brain with their phase realignment contributing to the coordination of neural communication. Phase realignments may have especially strong (or weak) impact when neural activities are strongly synchronized (or desynchronized) within the interacting populations. We report that the spatiotemporal dynamics of strong regional synchronization measured as maximal EEG spectral power-referred to as activation-and strong regional desynchronization measured as minimal EEG spectral power-referred to as suppression-are characterized by the spatial segregation of small-scale and large-scale networks. Specifically, small-scale spectral-power activations and suppressions involving only 2-7% (1-4 of 60) of EEG scalp sites were prolonged (relative to stochastic dynamics) and consistently co-localized in a frequency specific manner. For example, the small-scale networks for θ, α, β1, and β2 bands (4-30 Hz) consistently included frontal sites when the eyes were closed, whereas the small-scale network for γ band (31-55 Hz) consistently clustered in medial-central-posterior sites whether the eyes were open or closed. Large-scale activations and suppressions involving over 17-30% (10-18 of 60) of EEG sites were also prolonged and generally clustered in regions complementary to where small-scale activations and suppressions clustered. In contrast, intermediate-scale activations and suppressions (involving 7-17% of EEG sites) tended to follow stochastic dynamics and were less consistently localized. These results suggest that strong synchronizations and desynchronizations tend to occur in small-scale and large-scale networks that are spatially segregated and frequency specific. These synchronization networks may broadly segregate the relatively independent and highly cooperative oscillatory processes while phase realignments fine-tune the network configurations based on behavioral demands.

Project description:Adverse listening conditions increase the demand on cognitive resources needed for speech comprehension. In an exploratory study, we aimed to identify independent power spectral features in the EEG useful for studying the cognitive processes involved in this effortful listening. Listeners performed the coordinate response measure task with a single-talker masker at a 0-dB signal-to-noise ratio. Sounds were left unfiltered or degraded with low-pass filtering. Independent component analysis (ICA) was used to identify independent components (ICs) in the EEG data, the power spectral dynamics of which were then analyzed. Frontal midline theta, left frontal, right frontal, left mu, right mu, left temporal, parietal, left occipital, central occipital, and right occipital clusters of ICs were identified. All IC clusters showed some significant listening-related changes in their power spectrum. This included sustained theta enhancements, gamma enhancements, alpha enhancements, alpha suppression, beta enhancements, and mu rhythm suppression. Several of these effects were absent or negligible using traditional channel analyses. Comparison of filtered to unfiltered speech revealed a stronger alpha suppression in the parietal and central occipital clusters of ICs for the filtered speech condition. This not only replicates recent findings showing greater alpha suppression as listening difficulty increases but also suggests that such alpha-band effects can stem from multiple cortical sources. We lay out the advantages of the ICA approach over the restrictive analyses that have been used as of late in the study of listening effort. We also make suggestions for moving into hypothesis-driven studies regarding the power spectral features that were revealed.

Project description:There has been a growing interest in computational electroencephalogram (EEG) signal processing in a diverse set of domains, such as cortical excitability analysis, event-related synchronization, or desynchronization analysis. In recent years, several inconsistencies were found across different EEG studies, which authors often attributed to methodological differences. However, the assessment of such discrepancies is deeply underexplored. It is currently unknown if methodological differences can fully explain emerging differences and the nature of these differences. This study aims to contrast widely used methodological approaches in EEG processing and compare their effects on the outcome variables. To this end, two publicly available datasets were collected, each having unique traits so as to validate the results in two different EEG territories. The first dataset included signals with event-related potentials (visual stimulation) from 45 subjects. The second dataset included resting state EEG signals from 16 subjects. Five EEG processing steps, involved in the computation of power and phase quantities of EEG frequency bands, were explored in this study: artifact removal choices (with and without artifact removal), EEG signal transformation choices (raw EEG channels, Hjorth transformed channels, and averaged channels across primary motor cortex), filtering algorithms (Butterworth filter and Blackman-Harris window), EEG time window choices (-750 ms to 0 ms and -250 ms to 0 ms), and power spectral density (PSD) estimation algorithms (Welch's method, Fast Fourier Transform, and Burg's method). Powers and phases estimated by carrying out variations of these five methods were analyzed statistically for all subjects. The results indicated that the choices in EEG transformation and time-window can strongly affect the PSD quantities in a variety of ways. Additionally, EEG transformation and filter choices can influence phase quantities significantly. These results raise the need for a consistent and standard EEG processing pipeline for computational EEG studies. Consistency of signal processing methods cannot only help produce comparable results and reproducible research, but also pave the way for federated machine learning methods, e.g., where model parameters rather than data are shared.

Project description:IntroductionElectroencephalographic (EEG) data quality is severely compromised when recorded inside the magnetic resonance (MR) environment. Here we characterized the impact of the ballistocardiographic (BCG) artifact on resting-state EEG spectral properties and compared the effectiveness of seven common BCG correction methods to preserve EEG spectral features. We also assessed if these methods retained posterior alpha power reactivity to an eyes closure-opening (EC-EO) task and compared the results from EEG-informed fMRI analysis using different BCG correction approaches.MethodElectroencephalographic data from 20 healthy young adults were recorded outside the MR environment and during simultaneous fMRI acquisition. The gradient artifact was effectively removed from EEG-fMRI acquisitions using Average Artifact Subtraction (AAS). The BCG artifact was corrected with seven methods: AAS, Optimal Basis Set (OBS), Independent Component Analysis (ICA), OBS followed by ICA, AAS followed by ICA, PROJIC-AAS and PROJIC-OBS. EEG signal preservation was assessed by comparing the spectral power of traditional frequency bands from the corrected rs-EEG-fMRI data with the data recorded outside the scanner. We then assessed the preservation of posterior alpha functional reactivity by computing the ratio between the EC and EO conditions during the EC-EO task. EEG-informed fMRI analysis of the EC-EO task was performed using alpha power-derived BOLD signal predictors obtained from the EEG signals corrected with different methods.ResultsThe BCG artifact caused significant distortions (increased absolute power, altered relative power) across all frequency bands. Artifact residuals/signal losses were present after applying all correction methods. The EEG reactivity to the EC-EO task was better preserved with ICA-based correction approaches, particularly when using ICA feature extraction to isolate alpha power fluctuations, which allowed to accurately predict hemodynamic signal fluctuations during the EEG-informed fMRI analysis.DiscussionCurrent software solutions for the BCG artifact problem offer limited efficiency to preserve the EEG spectral power properties using this particular EEG setup. The state-of-the-art approaches tested here can be further refined and should be combined with hardware implementations to better preserve EEG signal properties during simultaneous EEG-fMRI. Existing and novel BCG artifact correction methods should be validated by evaluating signal preservation of both ERPs and spontaneous EEG spectral power.

Project description:BackgroundTypical electroencephalogram (EEG) recordings often contain substantial artifact. These artifacts, often large and intermittent, can interfere with quantification of the EEG via its power spectrum. To reduce the impact of artifact, EEG records are typically cleaned by a preprocessing stage that removes individual segments or components of the recording. However, such preprocessing can introduce bias, discard available signal, and be labor-intensive. With this motivation, we present a method that uses robust statistics to reduce dependence on preprocessing by minimizing the effect of large intermittent outliers on the spectral estimates.New methodUsing the multitaper method (Thomson, 1982) as a starting point, we replaced the final step of the standard power spectrum calculation with a quantile-based estimator, and the Jackknife approach to confidence intervals with a Bayesian approach. The method is implemented in provided MATLAB modules, which extend the widely used Chronux toolbox.ResultsUsing both simulated and human data, we show that in the presence of large intermittent outliers, the robust method produces improved estimates of the power spectrum, and that the Bayesian confidence intervals yield close-to-veridical coverage factors.Comparison to existing methodThe robust method, as compared to the standard method, is less affected by artifact: inclusion of outliers produces fewer changes in the shape of the power spectrum as well as in the coverage factor.ConclusionIn the presence of large intermittent outliers, the robust method can reduce dependence on data preprocessing as compared to standard methods of spectral estimation.

Project description:Electroencephalography (EEG) data possess a complex structure that includes regional, functional, and longitudinal dimensions. Our motivating example is a word segmentation paradigm in which typically developing (TD) children, and children with autism spectrum disorder (ASD) were exposed to a continuous speech stream. For each subject, continuous EEG signals recorded at each electrode were divided into one-second segments and projected into the frequency domain via fast Fourier transform. Following a spectral principal components analysis, the resulting data consist of region-referenced principal power indexed regionally by scalp location, functionally across frequencies, and longitudinally by one-second segments. Standard EEG power analyses often collapse information across the longitudinal and functional dimensions by averaging power across segments and concentrating on specific frequency bands. We propose a hybrid principal components analysis for region-referenced longitudinal functional EEG data, which utilizes both vector and functional principal components analyses and does not collapse information along any of the three dimensions of the data. The proposed decomposition only assumes weak separability of the higher-dimensional covariance process and utilizes a product of one dimensional eigenvectors and eigenfunctions, obtained from the regional, functional, and longitudinal marginal covariances, to represent the observed data, providing a computationally feasible non-parametric approach. A mixed effects framework is proposed to estimate the model components coupled with a bootstrap test for group level inference, both geared towards sparse data applications. Analysis of the data from the word segmentation paradigm leads to valuable insights about group-region differences among the TD and verbal and minimally verbal children with ASD. Finite sample properties of the proposed estimation framework and bootstrap inference procedure are further studied via extensive simulations.

Project description:Electroencephalography (EEG) studies produce region-referenced functional data via EEG signals recorded across scalp electrodes. The high-dimensional data can be used to contrast neurodevelopmental trajectories between diagnostic groups, for example between typically developing (TD) children and children with autism spectrum disorder (ASD). Valid inference requires characterization of the complex EEG dependency structure as well as covariate-dependent heteroscedasticity, such as changes in variation over developmental age. In our motivating study, EEG data is collected on TD and ASD children aged two to twelve years old. The peak alpha frequency, a prominent peak in the alpha spectrum, is a biomarker linked to neurodevelopment that shifts as children age. To retain information, we model patterns of alpha spectral variation, rather than just the peak location, regionally across the scalp and chronologically across development. We propose a covariate-adjusted hybrid principal components analysis (CA-HPCA) for EEG data, which utilizes both vector and functional principal components analysis while simultaneously adjusting for covariate-dependent heteroscedasticity. CA-HPCA assumes the covariance process is weakly separable conditional on observed covariates, allowing for covariate-adjustments to be made on the marginal covariances rather than the full covariance leading to stable and computationally efficient estimation. The proposed methodology provides novel insights into neurodevelopmental differences between TD and ASD children.

Project description:Autism spectrum disorder (ASD) is a neurodevelopmental condition that affects the brain's function. Electroencephalography (EEG) is a non-invasive technique that measures the electrical activity of the brain and can reveal its dynamics and information processing. This study explores an eyes-opened resting state quantitative EEG analysis of 49 children with ASD and 39 typically developing (TD or Control) children, using various features of entropy and complexity. Time and frequency domain features were applied for all EEG channels, such as the power spectra, brain rate, sample entropy, permutation entropy, spectral entropy, Tsallis entropy, Rényi entropy, Lempel-Ziv complexity, and Higuchi fractal dimension. The features were compared between the ASD and TD groups and tested for statistical significance. The results showed that the ASD group had a lower brain rate, higher Tsallis entropy and Rényi entropy, and lower Lempel-Ziv complexity than the TD group. The entropy results show impaired neural synchronization, increased randomness, and noise in ASD. The Lempel-Ziv complexity results showed that it is a potential indicator of the existence of focal spikes in the EEG signals of ASD. The brain-rate results show a low level of arousal in ASD. The findings suggest that entropy and complexity measures can be useful tools for characterizing the EEG features of ASD and provide insights into the neurophysiological mechanisms of the disorder.

Project description:Study objectivesMyotonic dystrophy type 1 (DM1) is a multisystemic disorder that involves the central nervous system (CNS). Individuals with DM1 commonly present with sleep dysregulation, including excessive daytime sleepiness and sleep-disordered breathing. We aim to characterize electroencephalogram (EEG) power spectra from nocturnal polysomnography (PSG) in patients with DM1 compared to matched controls to better understand the potential CNS sleep dysfunction in DM1.MethodsA retrospective, case-control (1:2) chart review of patients with DM1 (n = 18) and matched controls (n = 36) referred for clinical PSG at the Stanford Sleep Center was performed. Controls were matched based on age, sex, apnea-hypopnea index (AHI), body mass index (BMI), and Epworth Sleepiness Scale (ESS). Sleep stage and respiratory metrics for the two groups were compared. Power spectral analysis of the EEG C3-M2 signal was performed using the fast Fourier transformation.ResultsPatients with DM1 had significantly increased theta percent power in stage N2 sleep compared to matched controls. Theta/beta and theta/alpha percent power spectral ratios were found to be significantly increased in stage N2, N3, all sleep stages combined, and all wake periods combined in patients with DM1 compared to controls. A significantly lower nadir O2 saturation was also found in patients with DM1 versus controls.ConclusionsCompared to matched controls, patients with DM1 had increased EEG theta spectral power. Increased theta/beta and theta/alpha power spectral ratios in nocturnal PSG may reflect DM1 pathology in the CNS.

Project description:Slow wave oscillations in the electroencephalogram (EEG) during sleep may reflect both sleep need and intensity, which are implied in homeostatic regulation. Adenosine is strongly implicated in sleep homeostasis, and a single nucleotide polymorphism in the adenosine deaminase gene (ADA G22A) has been associated with deeper and more efficient sleep. The present study verified the association between the ADA G22A polymorphism and changes in sleep EEG spectral power (from C3-A2, C4-A1, O1-A2, and O2-A1 derivations) in the Epidemiologic Sleep Study (EPISONO) sample from São Paulo, Brazil. Eight-hundred individuals were subjected to full-night polysomnography and ADA G22A genotyping. Spectral analysis of the EEG was carried out in all individuals using fast Fourier transformation of the signals from each EEG electrode. The genotype groups were compared in the whole sample and in a subsample of 120 individuals matched according to ADA genotype for age, gender, body mass index, caffeine intake status, presence of sleep disturbance, and sleep-disturbing medication. When compared with homozygous GG genotype carriers, A allele carriers showed higher delta spectral power in Stage 1 and Stages 3+4 of sleep, and increased theta spectral power in Stages 1, 2 and REM sleep. These changes were seen both in the whole sample and in the matched subset. The higher EEG spectral power indicates that the sleep of individuals carrying the A allele may be more intense. Therefore, this polymorphism may be an important source of variation in sleep homeostasis in humans, through modulation of specific components of the sleep EEG.
