Electrochemical Investigation of Curcumin-DNA Interaction by Using Hydroxyapatite Nanoparticles-Ionic Liquids Based Composite Electrodes.

ABSTRACT: Hydroxyapatite nanoparticles (HaP) and ionic liquid (IL) modified pencil graphite electrodes (PGEs) are newly developed in this assay. Electrochemical impedance spectroscopy (EIS), scanning electron microscopy (SEM), energy-dispersive X-ray spectroscopy (EDX), and cyclic voltammetry (CV) were applied to examine the microscopic and electrochemical characterization of HaP and IL-modified biosensors. The interaction of curcumin with nucleic acids and polymerase chain reaction (PCR) samples was investigated by measuring the changes at the oxidation signals of both curcumin and guanine by differential pulse voltammetry (DPV) technique. The optimization of curcumin concentration, DNA concentration, and the interaction time was performed. The interaction of curcumin with PCR samples was also investigated by gel electrophoresis.

Project description:Quaternary ammonium theophylline-based ionic liquids and imidazolium-based ionic liquids, magnesium oxide and silica nanoparticles were used in order to investigate the interaction with Gram negative Escherichia coli and Gram positive Bacillus cereus. The changes of bacterial sensitivity to both nanoparticles (NPs) and ionic liquids (ILs) were examined. In order to assess the impact of ILs on the interaction of nanoparticles with bacteria, respirometric analysis, activity of dehydrogenases, peroxidase analyses as well as scanning and fluorescence microscopy examinations were conducted. The interactions of ILs with nanoparticles based on adsorption and sedimentation tests were also investigated in order to assess how the ILs affect the agglomeration of NPs. It was assumed, as the main hypothesis of the present studies, that the differences in sensitivity of bacteria to combined ILs and NPs can be observed, even if the concentration of both compounds are below the minimum inhibitory concentration (MIC). The results indicated that ILs strongly affected the sensitivity of bacteria to nanoparticles however, the changes of sensitivity depended on the surface characteristics of the nanoparticles. The presence of ILs at non-lethal concentrations caused an increase of bacterial sensitivity to MgO nanoparticles. Notably, the sensitivity of Gram positive bacteria increased significantly when ILs were present. This was an important observation because the toxicity of nanoparticles toward Gram positive bacteria is usually lower than their toxicity toward Gram negative bacteria. Using silica nanoparticles, the presence of ionic liquids caused the adsorption of bacteria onto the surface of nanoparticle agglomerates. In conclusion, two opposing effects have been observed. On the one hand, the toxicity of MgO NPs in the presence of ILs has increased. On the other hand, the presence of silica nanoparticles caused a decreased sensitivity of both types of bacteria toward ILs. Our studies indicate potentially useful processes in many environmantal protection technologies like water treatment where flocculation and disinfection are extremely needed.

Project description:This paper describes an electropolymerization-based on-chip valving system, accomplished by electrosynthesis of conductive polymeric ionic liquid (CPIL) films at selected points within an array of bipolar electrodes (BPEs), in which each of these wireless electrodes spans an IL-aqueous phase boundary. The low viscosity and high hydrophobicity of the CPIL precursor allow it to be patterned by established microfluidic methods. This advancement has the potential to impact microscale analysis because it allows on-demand creation of solid CPIL microstructures at locations specified by microfluidics, phase boundaries, and electrode potentials. To achieve this outcome, an imidazolium-based IL was functionalized with a pyrrole moiety, and the viscosity was tuned by choosing the appropriate counterion to form a CPIL with the desired viscosity, hydrophobicity, and oxidation potential. This monomer species was then introduced into a microfluidic device, which was prefilled with an aqueous buffer solution. The device comprised many parallel microchannels lined with nanoliter-scale chambers. BPEs interconnected the channels such that the BPE tips were each aligned to a chamber opening. The electrodes contacting the outermost channels were connected directly to a power supply and functioned as driving electrodes. The CPIL displaced the buffer in the channels and established a phase boundary at the opening of each chamber, thereby digitizing the aqueous phase. Finally, an alternating square waveform (under mode 1) was applied for 5 min to yield immobilized polymer films at a location defined by the BPE poles. In total, three modes were developed and three corresponding polymer film patterns were formed. Under mode 2, a DC power supply was used to achieve a dissymmetrical polymer film pattern, and, under mode 3, a regional polymer film pattern was formed under an AC potential with a DC offset. Our preliminary results demonstrate that the generated polymer films are immobile and sufficiently thick to seal the chambers at room temperature over the duration of our observation window (50 min), and this seal is maintained even at elevated temperatures that induce partial evaporation of the chamber contents. A key point is that this method is compatible with a preceding step─dielectrophoretic capture of single melanoma cells within the nanoliter-scale chambers.
