Rethinking temperature effects on leaf growth, gene expression and metabolism: Diel variation matters.

ABSTRACT: Plants have evolved to grow under prominently fluctuating environmental conditions. In experiments under controlled conditions, temperature is often set to artificial, binary regimes with constant values at day and at night. This study investigated how such a diel (24 hr) temperature regime affects leaf growth, carbohydrate metabolism and gene expression, compared to a temperature regime with a field-like gradual increase and decline throughout 24 hr. Soybean (Glycine max) was grown under two contrasting diel temperature treatments. Leaf growth was measured in high temporal resolution. Periodical measurements were performed of carbohydrate concentrations, carbon isotopes as well as the transcriptome by RNA sequencing. Leaf growth activity peaked at different times under the two treatments, which cannot be explained intuitively. Under field-like temperature conditions, leaf growth followed temperature and peaked in the afternoon, whereas in the binary temperature regime, growth increased at night and decreased during daytime. Differential gene expression data suggest that a synchronization of cell division activity seems to be evoked in the binary temperature regime. Overall, the results show that the coordination of a wide range of metabolic processes is markedly affected by the diel variation of temperature, which emphasizes the importance of realistic environmental settings in controlled condition experiments.

Project description:BACKGROUND: Numerous biochemical and physiological parameters of living organisms follow a circadian rhythm. Although such rhythmic behavior is particularly pronounced in plants, which are strictly dependent on the daily photoperiod, data on the molecular aspects of the diurnal cycle in plants is scarce and mostly concerns the model species Arabidopsis thaliana. Here we studied the leaf transcriptome in seedlings of maize, an important C4 crop only distantly related to A. thaliana, throughout a cycle of 10 h darkness and 14 h light to look for rhythmic patterns of gene expression. RESULTS: Using DNA microarrays comprising ca. 43,000 maize-specific probes we found that ca. 12% of all genes showed clear-cut diel rhythms of expression. Cluster analysis identified 35 groups containing from four to ca. 1,000 genes, each comprising genes of similar expression patterns. Perhaps unexpectedly, the most pronounced and most common (concerning the highest number of genes) expression maxima were observed towards and during the dark phase. Using Gene Ontology classification several meaningful functional associations were found among genes showing similar diel expression patterns, including massive induction of expression of genes related to gene expression, translation, protein modification and folding at dusk and night. Additionally, we found a clear-cut tendency among genes belonging to individual clusters to share defined transcription factor-binding sequences. CONCLUSIONS: Co-expressed genes belonging to individual clusters are likely to be regulated by common mechanisms. The nocturnal phase of the diurnal cycle involves gross induction of fundamental biochemical processes and should be studied more thoroughly than was appreciated in most earlier physiological studies. Although some general mechanisms responsible for the diel regulation of gene expression might be shared among plants, details of the diurnal regulation of gene expression seem to differ between taxa.

Project description:Body size, and, by extension, growth has impacts on physiology, survival, attainment of sexual maturity, fecundity, generation time, and population dynamics, especially in ectotherm animals that often exhibit extensive growth following attainment of sexual maturity. Frequently, growth is analyzed at the population level, providing useful population mean growth parameters but ignoring individual variation that is also of ecological and evolutionary significance. Our long-term study of Lake Erie Watersnakes, Nerodia sipedon insularum, provides data sufficient for a detailed analysis of population and individual growth. We describe population mean growth separately for males and females based on size of known age individuals (847 captures of 769 males, 748 captures of 684 females) and annual growth increments of individuals of unknown age (1,152 males, 730 females). We characterize individual variation in asymptotic size based on repeated measurements of 69 males and 71 females that were each captured in five to nine different years. The most striking result of our analyses is that asymptotic size varies dramatically among individuals, ranging from 631-820 mm snout-vent length in males and from 835-1125 mm in females. Because female fecundity increases with increasing body size, we explore the impact of individual variation in asymptotic size on lifetime reproductive success using a range of realistic estimates of annual survival. When all females commence reproduction at the same age, lifetime reproductive success is greatest for females with greater asymptotic size regardless of annual survival. But when reproduction is delayed in females with greater asymptotic size, lifetime reproductive success is greatest for females with lower asymptotic size when annual survival is low. Possible causes of individual variation in asymptotic size, including individual- and cohort-specific variation in size at birth and early growth, warrant further investigation.

Project description:Lipids and carbohydrates are the two primary energy sources for both animals and insects. Energy homeostasis is under strict control by the neuroendocrine system, and disruption of energy homeostasis leads to the development of various disorders, such as obesity, diabetes, fatty liver syndrome, and cardiac dysfunction. One critical factor in this respect is feeding habits and diet composition. Insects are good models to study the physiological and biochemical background of the effect of diet on energy homeostasis and related disorders; however, most studies are based on a single model species, Drosophila melanogaster. In the current study, we examined the effects of four different diets, high fat (HFD), high sugar (HSD), calcium-rich (CRD), and a plant-based (PBD) on energy homeostasis in younger (third instar) and older (fifth instar) larvae of the Egyptian cotton leafworm, Spodoptera littoralis (Lepidoptera: Noctuidae) in comparison to a regular artificial bean diet. Both HSD and HFD led to weight gain, while CRD had the opposite effect and PBD had no effect in fifth instar larvae and pupae. The pattern was the same for HSD and CRD in third instar larvae while a reduction in weight was detected with HFD and PBD. Larval development was shortest with the HSD, while HFD, CRD, and PBD led to retardation compared to the control. Triglyceride (TG) levels were higher with HFD, HSD, and PBD, with larger lipid droplet sizes, while CRD led to a reduction of TG levels and lipid droplet size. Trehalose levels were highest with HSD, while CRD led to a reduction at third instar larvae, and HFD and PBD had no effect. Fifth instar larvae had similar levels of trehalose with all diets. There was no difference in the expression of the genes encoding neuropeptides SpoliAKH and SpoliILP1-2 with different diets in third instar larvae, while all three genes were expressed primarily with HSD, and SpolisNPF was primarily expressed with HFD in fifth instar larvae. In summary, different diet treatments alter the development of insects, and energy and metabolic pathways through the regulation of peptide hormones.
