A study on selective transformation of norbornadiene into fluorinated cyclopentane-fused isoxazolines.

ABSTRACT: This work presents an examination of the selective functionalization of norbornadiene through nitrile oxide 1,3-dipolar cycloaddition/ring-opening metathesis (ROM)/cross-metathesis (CM) protocols. Functionalization of commercially available norbornadiene provided novel bicyclic scaffolds with multiple stereogenic centers. The synthesis involved selective cycloadditions, with subsequent ROM of the formed cycloalkene-fused isoxazoline scaffolds and selective CM by chemodifferentiation of the olefin bonds of the resulting alkenylated derivatives. Various experimental conditions were applied for the CM transformations with the goal of exploring substrate and steric effects, catalyst influence and chemodifferentiation of the olefin bonds furnishing the corresponding functionalized, fluorine-containing isoxazoline derivatives.

Project description:Transition metal-mediated N-O bond cleavage reactions of heterobicycloalkene-fused 3-methyl-2-isoxazolines were investigated. Optimal cleavage conditions were found with Raney nickel/AlCl3 mediation in aqueous methanol. The reaction provided a diverse collection of novel heterobicycle-fused β-hydroxyketones with good to excellent yields (66-95%) and without the need for chromatographic purification.

The selective hydrosilylation of norbornadiene-2,5 by monohydrosiloxanes.

Project description:A simple one-step approach for the selective synthesis of exo-norbornenes with organosilicon substituents is suggested through the direct hydrosilylation of norbornadiene-2,5 with chlorine-free silanes. Using the example of norbornadiene-2,5 hydrosilylation with pentamethyldisiloxane and 1,1,1,3,5,5,5-heptamethyltrisiloxane, the possibility of obtaining exo-isomers of norbornenes with 100 exo-/endo-selectivity is shown. The investigation of Pt-, Rh-, and Pd-complexes in combination with various ligands as catalysts was performed. The hydrosilylation of norbornadiene-2,5 in the presence of Pt- or Rh-catalysts was not selective and led to a mixture consisting of three isomers (exo-/endo-norbornenes and substituted nortricyclane). In the case of the Pd-salt/ligand catalytic system, the formation of an endo-isomer was not observed at all and only two isomers were formed (exo-norbornene and nortricyclane). The selectivity of exo-norbornene/nortricyclane formation strongly depended on the nature of the ligand in the Pd-catalyst. The best selectivity was revealed when R-MOP was the ligand, while the highest catalytic activity was reached with a dioxalane-containing ligand.

Palladium-catalyzed synthesis of cyclopentane-fused benzocyclobutenes via tandem directed carbopalladation/C-H bond functionalization.

Project description:A new Pd-catalyzed reaction for the stereoselective synthesis of cyclopentane-fused benzocyclobutenes is described. These transformations likely proceed via carbamate-directed carbopalladation followed by intramolecular C-H activation of an alkylpalladium intermediate. The mechanistic relationship between these transformations and Pd-catalyzed reactions of gamma-(n-Boc-amino)alkenes with aryl bromides that afford pyrrolidines is discussed. Differences in reactivity between Pd-amino and Pd-amido complexes appear to play a key role in the outcome of these transformations.

Electrophilic aromatic substitution using fluorinated isoxazolines at the C5 position <i>via</i> C-F bond cleavage.

Project description:Electrophilic aromatic substitution at the C5 position of isoxazolines and construction of a new quaternary carbon center were achieved in this paper. This is the first report of carbon-carbon (C-C) bond formation onto isoxazoline without compromising the ring structure. Various aromatics including heteroaromatics gave the desired products in good yields, especially aromatics bearing electron-donating groups. The reaction proceeds via the SEAr reaction mechanism, in which carbocation intermediates generated from the fluorinated isoxazolines via C-F bond cleavage reacted with aromatics.

Copolymerization of Norbornene and Norbornadiene Using a cis-Selective Bimetallic W-Based Catalytic System.

Project description:The bimetallic cluster Na[W₂(μ-Cl)₃Cl₄(THF)₂]·(THF)₃ ({W₂}, {W 3 W}6+, a'²e'⁴), which features a triple metal-metal bond, is a highly efficient room-temperature initiator for ring opening metathesis polymerization (ROMP) of norbornene (NBE) and norbornadiene (NBD), providing high-cis polymers. In this work, {W₂} was used for the copolymerization of the aforementioned monomers, yielding statistical poly(norbornene)/poly(norbornadiene) PNBE/PNBD copolymers of high molecular weight and high-cis content. The composition of the polymer chain was estimated by 13C CPMAS NMR data and it was found that the ratio of PNBE/PNBD segments in the polymer chain was relative to the monomer molar ratio in the reaction mixture. The thermal properties of all copolymers were similar, resembled the properties of PNBD homopolymer and indicated a high degree of cross-linking. The morphology of all materials in this study was smooth and non-porous; copolymers with higher PNBE content featured a corrugated morphology. Glass transition temperatures were lower for the copolymers than for the homopolymers, providing a strong indication that those materials featured a branched-shaped structure. This conclusion was further supported by viscosity measurements of copolymers solutions in THF. The molecular structure of those materials can be controlled, potentially leading to well-defined star polymers via the "core-first" synthesis method. Therefore, {W₂} is not only a cost-efficient, practical, highly active, and cis-stereoselective ROMP-initiator, but it can also be used for the synthesis of more complex macromolecular structures.

Gas-Phase Transformation of Fluorinated Benzoporphyrins to Porphyrin-Embedded Conical Nanocarbons.

Project description:Geodesic nitrogen-containing graphene fragments are interesting candidates for various material applications, but the available synthetic protocols, which need to overcome intrinsic strain energy during the formation of the bowl-shaped skeletons, are often incompatible with heteroatom-embedded structures. Through this mass spectrometry-based gas-phase study, we show by means of collision-induced dissociation experiments and supported by density functional theory calculations, the first evidence for the formation of a porphyrin-embedded conical nanocarbon. The influences of metalation and functionalization of the used tetrabenzoporphyrins have been investigated, which revealed different cyclization efficiencies, different ionization possibilities, and a variation of the dissociation pathway. Our results suggest a stepwise process for HF elimination from the fjord region, which supports a selective pathway towards bent nitrogen-containing graphene fragments.

Novel fluorinated ring-fused chlorins as promising PDT agents against melanoma and esophagus cancer.

Project description:Investigation of novel 4,5,6,7-tetrahydropyrazolo[1,5-a]pyridine-fused chlorins, derived from 5,10,15,20-tetrakis(pentafluorophenyl)porphyrin, as PDT agents against melanoma and esophagus cancer is disclosed. Diol and diester fluorinated ring-fused chlorins, including derivatives with 2-(2-hydroxyethoxy)ethanamino groups at the phenyl rings, were obtained via a two-step methodology, combining SNAr and [8π + 2π] cycloaddition reactions. The short-chain PEG groups at the para-position of the phenyl rings together with the diol moiety at the fused pyrazole ring promote a red-shift of the Soret band, a decrease of the fluorescence quantum yield and an increase of the singlet oxygen formation quantum yield, improving the photophysical characteristics required to act as a photosensitizer. Introduction of these hydrophilic groups also improves the incorporation of the sensitizers by the cells reaching cellular uptake values of nearly 50% of the initial dose. The rational design led to a photosensitizer with impressive IC50 values, 13 and 27 nM against human melanoma and esophageal carcinoma cell lines, respectively.

Mechanochemical transformation of planar polyarenes to curved fused-ring systems.

Project description:The transformation of planar aromatic molecules into π-extended non-planar structures is a challenging task and has not been realized by mechanochemistry before. Here we report that mechanochemical forces can successfully transform a planar polyarene into a curved geometry by creating new C-C bonds along the rim of the molecular structure. In doing so, mechanochemistry does not require inert conditions or organic solvents and provide better yields within shorter reaction times. This is illustrated in a 20-minute synthesis of corannulene, a fragment of fullerene C60, in 66% yield through ball milling of planar tetrabromomethylfluoranthene precursor under ambient conditions. Traditional solution and gas-phase synthetic pathways do not compete with the practicality and efficiency offered by the mechanochemical synthesis, which now opens up a new reaction space for inducing curvature at a molecular level.

Exploiting Partial Solubility in Partially Fluorinated Thermoplastic Blends to Improve Adhesion during Fused Deposition Modeling.

Project description:This work studies the effect of interlayer adhesion on mechanical performance of fluorinated thermoplastics produced by fused deposition modeling (FDM). Here, we study the anisotropic mechanical response of 3D-printed binary blends of poly (vinylidene fluoride) (PVDF) and poly (methyl methacrylate) (PMMA) with the isotropic mechanical response of these blends fabricated via injection molding. Various PVDF/PMMA filament compositions were produced by twin-screw extrusion and, subsequently, injection-molded or 3D printed into dog-bone shapes. Specimen mechanical and thermal properties were evaluated by mode I tensile testing and differential scanning calorimetry, respectively. Results show that higher PMMA concentration not only improved the tensile strength and decreased ductility but reduced PVDF crystallization. As expected, injection-molded samples revealed better mechanical properties compared to 3D printed specimens. Interestingly, 3D printed blends with lower PMMA content demonstrated better diffusion (adhesion) across interfaces than those with a higher amount of PMMA. The present study provides new findings that may be used to tune mechanical response in 3D printed fluorinated thermoplastics, particularly for energy applications.
