Project description:Cyanobacteria are prokaryotic phototrophs capable of achieving high cellular densities with minimal inputs. These prokaryotic organisms can grow using sunlight as energy source and carbon dioxide as carbon source what makes them promising candidates as microbial cell factories for the production of numerous compounds such as chemicals, fuels, or biocatalysts. In this study, we have successfully designed and constructed using synthetic biology approach two recombinant strains of Synechococcus elongatus PCC7942 for heterologous expression of the industrially relevant Bacillus subtilis CotA laccase. One of the strains (PCC7942-NSI-CotA) was constructed through integration of the laccase gene into neutral site I of the cyanobacterial genome whilst the other (PCC7942-NSII-CotA) targeted neutral site II of the genome. Of the two strains the one with CotA laccase integrated in neutral site II (PCC7942-NSII-CotA) was superior in terms of growth rate and enzymatic activity toward typical laccase substrates: ABTS [2,2-azino-bis (3-ethylbenzothiazoline-6-sulfonate)] and syringaldazine. That may suggest that two of the traditionally used neutral sites of S. elongatus PCC7942 are not equally suitable for the expression of certain transgenes. The PCC7942-NSII-CotA produced protein was capable of decolourising three classes of dyes namely: anthraquinonic-, azo-, and indigoid-type over 7 days of incubation making the strain a potentially useful microbial cell factory for the production of broad-spectrum biodegradation agent. Interestingly, presence of additional synthetic redox mediator ABTS had no effect on the degradation of these dyes.

Project description:Bacillus amyloliquefaciens HB-26, a Gram-positive bacterium was isolated from soil in China. SDS-PAGE analysis showed this strain secreted six major protein bands of 65, 60, 55, 34, 25 and 20 kDa. A bioassay of this strain reveals that it shows specific activity against P. brassicae and nematode. Here we describe the features of this organism, together with the draft genome sequence and annotation. The 3,989,358 bp long genome (39 contigs) contains 4,001 protein-coding genes and 80 RNA genes.

Project description:Textile waste contributes to the pollution of both terrestrial and aquatic ecosystems. While natural textile fibres are known to be biodegraded by microbes, the vast majority of textiles now contain a mixture of processed plant-derived polymers and synthetic materials generated from petroleum and are commonly dyed with azo dyes. This presents a complex recycling problem as the separation of threads and removal of dye are challenging and costly. As a result, the majority of textile waste is sent to landfill or incinerated. This project sought to assess the potential of fungal bioremediation of textile-based dye as a step towards sustainable and environmentally-friendly means of disposal of textile waste. Successful development of an agar-independent microcosm enabled the assessment of the ability of two fungal species to grow on a range of textiles containing an increasing percentage of elastane. The white rot fungus Hypholoma fasciculare was shown to grow well on semi-synthetic textiles, and for the first time, bioremediation of dye from textiles was demonstrated. Volatile analysis enabled preliminary assessment of the safety profile of this process and showed that industrial scale-up may require consideration of volatile capture in the design process. This study is the first to address the potential of fungi as bioremediation agents for solid textile waste, and the results suggest this is an avenue worthy of further exploration.

Project description:This study proposes the treatment and valorization of denim textile effluents through a fermentative hydrogen production process. Also, the study presents the decolorizing capabilities of bacterial and fungal isolates obtained from the fermented textile effluents. The maximum hydrogen production rate was 0.23 L H2/L-d, achieving at the same time color removal. A total of thirty-five bacteria and one fungal isolate were obtained from the fermented effluents and screened for their abilities to decolorize indigo dye, used as a model molecule. From them, isolates identified as Bacillus BT5, Bacillus BT9, Lactobacillus BT20, Lysinibacillus BT32, and Aspergillus H1T showed notable decolorizing capacities. Lactobacillus BT20 reached 90% of decolorization using glucose as co-substrate after 11 days of incubation producing colorless metabolites. Bacillus BT9 was able to utilize the indigo dye as the sole carbon source achieving a maximum decolorization of 60% after 9 days of incubation and producing a red-colored metabolite. In contrast, Bacillus BT5 and Lysinibacillus BT32 exhibited the lowest percentages of decolorization, barely 33% after 16 and 11 days of incubation, respectively. When Aspergillus H1T was grown in indigo dye supplemented with glucose, 96% of decolorization was reached after 2 days. This study demonstrates the valorization of denim textile effluents for the production of hydrogen via dark fermentation with concomitant color removal.

Project description:Dye-decolorizing peroxidase (DyP)-type peroxidases are a family of heme-containing peroxidases. Because DyP-type peroxidases can degrade recalcitrant anthraquinone dyes and lignin, their potential applications in the treatment of wastewater containing dyes and lignin degradation are expected. Although many DyP-type peroxidases have been characterized experimentally, most of the reported DyP-type peroxidases are from basidiomycetous fungi and bacteria. Therefore, the taxonomic distribution of the DyP-type peroxidases remains unclear. In this study, we analyzed the phylogenetic tree using all DyP-type peroxidase sequences available in the InterPro database. The findings mainly divided this family into three classes. Metazoa and Archaea also have the genes coding for DyP-type peroxidases, and the sequences belonging to two subclasses have the pyruvate formate lyase or cytochrome P450 domain in addition to the DyP domain. This study reveals differences in the conservation of important residues among classes. The findings will accelerate research on the DyP-type peroxidase family. Highlights • DyP-type peroxidase family is mainly divided into three classes P, I, and V.• Obvious difference of three classes is the amino acid sequence lengths.• Metazoa and Archaea also have the genes coding for DyP-type peroxidases.• Subclasses with domains other than DyP also exist.

Project description:Based on the well-known excellent adsorbent ability of chicken eggshells, the adsorptive capacity and mechanism of Remazol Brilliant Violet-5R (RBV-5R) dye by eggshell was investigated. Exploiting the high surface-area-to-volume ratio and porous structure of this natural adsorbent, the developed procedure showed to be useful for the efficient adsorption of RBV-5R dye from contaminated water. The protocol was thoroughly optimized by investigating the effect of the dye concentration, biomass-contaminated water ratio, particle size of the adsorbent, pH and temperature, as they are key factors in the efficiency of the dye removal process. The eggshell material was characterized by different types of microscopy techniques (stereo, polarization, SEM) as well as elemental analysis (element distribution mapping, EDX), Raman spectroscopy and BET-surface density measurements. EDX, FTIR and Raman spectroscopy proved the presence of the adsorbed dye on the surface of the biomaterial. It was shown that under optimal conditions, the environmentally friendly and inexpensive eggshell could be a reliable adsorbent for Remazol dye removal from wastewater.

Project description:In the biorefinery using lignocellulosic biomass as feedstock, pretreatment to breakdown or loosen lignin is important step and various approaches have been conducted. For biological pretreatment, we screened Bacillus subtilis KCTC2023 as a potential lignin-degrading bacterium based on veratryl alcohol (VA) oxidation test and the putative heme-containing dye-decolorizing peroxidase was found in the genome of B. subtilis KCTC2023. The peroxidase from B. subtilis KCTC2023 (BsDyP) was capable of oxidizing various substrates and atypically exhibits substrate-dependent optimum temperature: 30°C for dyes (Reactive Blue19 and Reactive Black5) and 50°C for high redox potential substrates (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid [ABTS], VA, and veratryl glycerol-β-guaiacyl ether [VGE]) over +1.0 V vs. normal hydrogen electrode. At 50°C, optimum temperature for high redox potential substrates, BsDyP not only showed the highest VA oxidation activity (0.13 Umg(-1)) among the previously reported bacterial peroxidases but also successfully achieved VGE decomposition by cleaving Cα-Cβ bond in the absence of any oxidative mediator with a specific activity of 0.086 Umg(-1) and a conversion rate of 53.5%. Based on our results, BsDyP was identified as the first bacterial peroxidase capable of oxidizing high redox potential lignin-related model compounds, especially VGE, revealing a previously unknown versatility of lignin degrading biocatalyst in nature.
