Comparative Mitogenomic Analysis of Five Awl Skippers (Lepidoptera: Hesperiidae: Coeliadinae) and Their Phylogenetic Implications.

ABSTRACT: To determine the significance of mitochondrial genome characteristics in revealing phylogenetic relationships and to shed light on the molecular evolution of the Coeliadinae species, the complete mitochondrial genomes (mitogenomes) of five Coeliadinae species were newly sequenced and analyzed, including Hasora schoenherr, Burara miracula, B. oedipodea, B. harisa, and Badamia exclamationis. The results show that all five mitogenomes are double-strand circular DNA molecules, with lengths of 15,340 bp, 15,295 bp, 15,304 bp, 15,295 bp, and 15,289 bp, respectively, and contain the typical 37 genes and a control region. Most protein-coding genes (PCGs) begin with ATN, with 3 types of stop codons including TAA, TAG, and an incomplete codon T-; most of the genes terminate with TAA. All of the transfer RNA genes (tRNAs) present the typical cloverleaf secondary structure except for the trnS1. Several conserved structural elements are found in the AT-rich region. Phylogenetic analyses based on three datasets (PCGs, PRT, and 12PRT) and using maximum likelihood (ML) and Bayesian inference (BI) methods show strong support for the monophyly of Coeliadinae, and the relationships of the five species are (B. exclamationis + ((B. harisa + (B. oedipodea + B. miracula)) + H. schoenherr)).

Project description:BackgroundInsect mitochondrial genomes (mitogenomes) are the most extensively used genetic marker for evolutionary and population genetics studies of insects. The Pentatomoidea superfamily is economically important and the largest superfamily within Pentatomomorpha with over 7,000 species. To better understand the diversity and evolution of pentatomoid species, we sequenced and annotated the mitogenomes of Eurydema gebleri and Rubiconia intermedia, and present the first comparative analysis of the 11 pentatomoid mitogenomes that have been sequenced to date.ResultsWe obtained the complete mitogenome of Eurydema gebleri (16,005 bp) and a nearly complete mitogenome of Rubiconia intermedia (14,967 bp). Our results show that gene content, gene arrangement, base composition, codon usage, and mitochondrial transcription termination factor sequences are highly conserved in pentatomoid species, especially for species in the same family. Evolutionary rate analyses of protein-coding genes reveal that the highest and lowest rates are found in atp8 and cox1 and distinctive evolutionary patterns are significantly correlated with the G + C content of genes. We inferred the secondary structures for two rRNA genes for eleven pentatomoid species, and identify some conserved motifs of RNA structures in Pentatomidea. All tRNA genes in pentatomoid mitogenomes have a canonical cloverleaf secondary structure, except for two tRNAs (trnS1 and trnV) which appear to lack the dihydrouridine arm. Regions that are A + T-rich have several distinct characteristics (e.g. size variation and abundant tandem repeats), and have potential as species or population level molecular markers. Phylogenetic analyses based on mitogenomic data strongly support the monophyly of Pentatomoidea, and the estimated phylogenetic relationships are: (Urostylididae + (Plataspidae + (Pentatomidae + (Cydnidae + (Dinidoridae + Tessaratomidae))))).ConclusionsThis comparative mitogenomic analysis sheds light on the architecture and evolution of mitogenomes in the superfamily Pentatomoidea. Mitogenomes can be effectively used to resolve phylogenetic relationships of pentatomomorphan insects at various taxonomic levels. Sequencing more mitogenomes at various taxonomic levels, particularly from closely related species, will improve the annotation accuracy of mitochondrial genes, as well as greatly enhance our understanding of mitogenomic evolution and phylogenetic relationships in pentatomoids.

Project description:We present an analysis of the names proposed by Carl Plötz in 1884 for the New World species in the genus Pyrgus Hübner, [1819] facilitated by the genomic sequencing of extant primary type specimens comparatively with a larger sample of more recently collected specimens of these species and their relatives. The changes to nomenclature suggested here are only caused by the identity of primary type specimens as revealed by their phenotypes or through genomic sequencing. All neotypes are designated to stabilize nomenclature in agreement with the current usage of these names, which in unison agrees best with the information available about them. Lectotypes are designated for the following 5 taxa: Pyrgus (Scelothrix [sic]) bellatrix Plötz, 1884 (type locality Argentina: Buenos Aires), Pyrgus (Pyrgus) willi Plötz, 1884 (type locality in Brazil: Minas Gerais), Pyrgus (Pyrgus) albescens Plötz, 1884 (type locality in Mexico), Pyrgus (Syrichthus [sic]) lycurgus Plötz, 1884 (type locality in "Central America", likely southern Mexico), and Pyrgus occidentalis Skinner, 1906 (type locality USA: Texas, San Antonio). Neotypes are designated for the following 4 taxa: Pyrgus (Pyrgus) adepta Plötz, 1884 (Herrich-Schäffer in litt.) (type locality Colombia: Bogota), Pyrgus (Scelothrix [sic]) dion Plötz, 1884 (type locality Colombia: Bogota), Pyrgus (Scelothrix [sic]) adjutrix Plötz, 1884 (Herrich-Schäffer in litt.) A new name Burnsius albezens Grishin sp. n. (type locality USA: Arizona, Cochise Co., Portal) is proposed for the species misidentified as B. albescens. Furthermore, genomic comparisons reveal two other new species and one new subspecies of Burnsius Grishin, 2019: B. burnsi Grishin sp. n. (type locality Mexico: Veracruz, Huatusco), B. adepta inepta Grishin ssp. n. (type locality Ecuador: Pichincha, Tandapi), and B. orcynus Grishin sp. n. (type locality Curaçao: Hato Field) that are cryptic and can be confidently identified only by their genotype.
