Drivers of parasite β-diversity among anuran hosts depend on scale, realm and parasite group.

ABSTRACT: A robust understanding of what drives parasite β-diversity is an essential step towards explaining what limits pathogens' geographical spread. We used a novel global dataset (latitude -39.8 to 61.05 and longitude -117.84 to 151.49) on helminths of anurans to investigate how the relative roles of climate, host composition and spatial distance to parasite β-diversity vary with spatial scale (global, Nearctic and Neotropical), parasite group (nematodes and trematodes) and host taxonomic subset (family). We found that spatial distance is the most important driver of parasite β-diversity at the global scale. Additionally, we showed that the relative effects of climate concerning distance increase at the regional scale when compared with the global scale and that trematodes are generally more responsive to climate than nematodes. Unlike previous studies done at the regional scale, we did not find an effect of host composition on parasite β-diversity. Our study presents a new contribution to parasite macroecological theory, evidencing spatial and taxonomic contingencies of parasite β-diversity patterns, which are related to the zoogeographical realm and host taxonomic subset, respectively. Project description:We partitioned the total beta diversity in the species composition of anuran tadpoles to evaluate if species replacement and nestedness components are congruent at different spatial resolutions in the Brazilian Atlantic Forest. We alternated the sampling grain and extent of the study area (among ponds at a site, among ponds within regions, among sites within regions, and among sites within regions pooled together) to assess the importance of anuran beta diversity components. We then performed variation partitioning to evaluate the congruence of environmental descriptors and geographical distance in explaining the spatial distribution of the species replacement and nestedness components. We found that species replacement was the main component of beta diversity, independent of the sampling grain and extent. Furthermore, when considering the same sampling grain and increasing the extent, the values of species replacement increased. On the other hand, when considering the same extent and increasing the sampling grain, the values of species replacement decreased. At the smallest sampling grain and extent, the environmental descriptors and geographic distance were not congruent and alternated in the percentage of variation explaining the spatial distribution of species replacement and nestedness. At the largest spatial scales (SSs), the biogeographical regions showed higher values of the percentage explaining the variation in the beta diversity components. We found high values of species replacement independently of the spatial resolution, but the processes driving community assembly seem to be dependent on the SS. At small scales, both stochastic and deterministic factors might be important processes structuring anuran tadpole assemblages. On the other hand, at a large spatial grain and extent, the processes restricting species distributions might be more effective for drawing inferences regarding the variation in anuran beta diversity in different regions of the Brazilian Atlantic Forest.

Project description:In the search for novel drug candidates, diverse environmental microbiomes have been surveyed for their secondary metabolite biosynthesis potential, yet little is known about the biosynthetic diversity encoded by divergent microbiomes from different ecosystems, and the environmental parameters driving this diversity. Here, we used targeted amplicon sequencing of adenylation (AD) and ketosynthase (KS) domains along with 16S sequencing to delineate the unique biosynthetic potential of microbiomes from three separate habitats (soil, water, and sediments) exhibiting unique small spatial scale physicochemical gradients. The estimated richness of AD domains was highest in marine sediments with 656 ± 58 operational biosynthetic units (OBUs), while the KS domain richness was highest in soil microbiomes with 388 ± 67 OBUs. Microbiomes with rich and diverse bacterial communities displayed the highest PK potential across all ecosystems, and on a small spatial scale, pH and salinity were significantly, positively correlated to KS domain richness in soil and aquatic systems, respectively. Integrating our findings, we were able to predict the KS domain richness with a RMSE of 31 OBUs and a R2 of 0.91, and by the use of publicly available information on bacterial richness and diversity, we identified grassland biomes as being particularly promising sites for the discovery of novel polyketides. Furthermore, a focus on acidobacterial taxa is likely to be fruitful, as these were responsible for most of the variation in biosynthetic diversity. Overall, our results highlight the importance of sampling diverse environments with high taxonomic diversity in the pursuit for novel secondary metabolites. IMPORTANCE To counteract the antibiotic resistance crisis, novel anti-infective agents need to be discovered and brought to market. Microbial secondary metabolites have been important sources of inspiration for small-molecule therapeutics. However, the isolation of novel antibiotics is difficult, and the risk of rediscovery is high. With the overarching purpose of identifying promising microbiomes for discovery of novel bioactivity, we mapped out the most significant drivers of biosynthetic diversity across divergent microbiomes. We found the biosynthetic potential to be unique to individual ecosystems, and to depend on bacterial taxonomic diversity. Within systems, and on small spatial scales, pH and salinity correlated positively to the biosynthetic richness of the microbiomes, Acidobacteria representing the taxa most highly associated with biosynthetic diversity. Ultimately, understanding the key drivers of the biosynthesis potential of environmental microbiomes will allow us to focus bioprospecting efforts and facilitate the discovery of novel therapeutics.
