Comparison of Design Approaches for Low-Cost Sampling Mechanisms in Open-Source Chemical Instrumentation.

ABSTRACT: Robotic positioning systems are used in a variety of chemical instruments, primarily for liquid handling purposes, such as autosamplers from vials or well plates. Here, two approaches to the design of open-source autosampler positioning systems for use with 96-well plates are described and compared. The first system, a 3-axis design similar to many low-cost 3D printers that are available on the market, is constructed using an aluminum design and stepper motors. The other system relies upon a series of 3D printed parts to achieve movement with a series of linker arms based on Selective Compliance Assembly Robot Arm (SCARA) design principles. Full printer design files, assembly instructions, software, and user directions are included for both samplers. The positioning precision of the 3-axis system is better than the SCARA mechanism due to finer motor control, albeit with a slightly higher cost of materials. Based on the improved precision of this approach, the 3-axis autosampler system was used to demonstrate the generation of a segmented flow droplet stream from adjacent wells within a 96-well plate.

Project description:ImportanceEndoscopes are paramount to the practice of otolaryngology. To provide physicians in low-middle-income countries with adequate tools to treat otolaryngologic problems, it is necessary to create a low-cost sustainable option.ObjectiveTo describe the design and usability of an open-source, low-cost flexible laryngoscope that addresses the lack of affordable and accessible methods for otolaryngologic visualization in resource-limited settings.Design, setting, and participantsThis quality improvement study used a mixed-methods approach, including a technical description of device design as well as quantitative and qualitative survey evaluation of device usability. Engineering involved device design, sourcing or manufacturing individual components, fabricating a prototype, and iterative testing. Key assumptions and needs for the device were identified in collaboration with otolaryngologists in Zimbabwe, and designed and simulated by biomedical engineers in a US university laboratory. Board-certified otolaryngologists at a single US university hospital trialed a completed prototype on simulated airways between May 2023 and June 2023.Main outcomes and measuresTechnical details on the design of the device are provided. Otolaryngologist gave feedback on device characteristics, maneuverability, and visualization using the System Usability Scale, a customized Likert-scale questionnaire (5-point scale), and semistructured interviews.ResultsA functional prototype meeting requirements was completed consisting of a distal-chip camera, spring bending tip, handle housing the control mechanism and electronics, and flexible polyether block amide-coated silicone sheath housing the camera and control wires; an external monitor provided real-time visualization and ability to store data. A total of 14 otolaryngologists participated in the device review. The mean (SD) System Usability Scale score was 88.93 (10.08), suggesting excellent usability. The device was rated highly for ease of set up, physical attributes, image quality, and functionality.Conclusions and relevanceThis quality improvement study described the design of a novel open-source low-cost flexible laryngoscope that external review with otolaryngologists suggests was usable and feasible in various resource-limited environments. Future work is needed to translate the model into a clinical setting.

Project description:The design-based and model-based approaches to frequentist statistical inference rest on fundamentally different foundations. In the design-based approach, inference relies on random sampling. In the model-based approach, inference relies on distributional assumptions. We compare the approaches in a finite population spatial context.We provide relevant background for the design-based and model-based approaches and then study their performance using simulated data and real data. The real data is from the United States Environmental Protection Agency's 2012 National Lakes Assessment. A variety of sample sizes, location layouts, dependence structures, and response types are considered. The population mean is the parameter of interest, and performance is measured using statistics like bias, squared error, and interval coverage.When studying the simulated and real data, we found that regardless of the strength of spatial dependence in the data, the generalized random tessellation stratified (GRTS) algorithm, which explicitly incorporates spatial locations into sampling, tends to outperform the simple random sampling (SRS) algorithm, which does not explicitly incorporate spatial locations into sampling. We also found that model-based inference tends to outperform design-based inference, even for skewed data where the model-based distributional assumptions are violated. The performance gap between design-based inference and model-based inference is small when GRTS samples are used but large when SRS samples are used, suggesting that the sampling choice (whether to use GRTS or SRS) is most important when performing design-based inference.There are many benefits and drawbacks to the design-based and model-based approaches for finite population spatial sampling and inference that practitioners must consider when choosing between them. We provide relevant background contextualizing each approach and study their properties in a variety of scenarios, making recommendations for use based on the practitioner's goals.
