Project description:BackgroundObesity, typically quantified in terms of Body Mass Index (BMI) exceeding threshold values, is considered a leading cause of premature death worldwide. For given body size (BMI), it is recognized that risk is also affected by body shape, particularly as a marker of abdominal fat deposits. Waist circumference (WC) is used as a risk indicator supplementary to BMI, but the high correlation of WC with BMI makes it hard to isolate the added value of WC.Methods and findingsWe considered a USA population sample of 14,105 non-pregnant adults (age ≥ 18) from the National Health and Nutrition Examination Survey (NHANES) 1999-2004 with follow-up for mortality averaging 5 yr (828 deaths). We developed A Body Shape Index (ABSI) based on WC adjusted for height and weight: ABSI ≡ WC/(BMI(2/3)height(1/2)). ABSI had little correlation with height, weight, or BMI. Death rates increased approximately exponentially with above average baseline ABSI (overall regression coefficient of +33% per standard deviation of ABSI [95% confidence interval: +20%-+48%), whereas elevated death rates were found for both high and low values of BMI and WC. 22% (8%-41%) of the population mortality hazard was attributable to high ABSI, compared to 15% (3%-30%) for BMI and 15% (4%-29%) for WC. The association of death rate with ABSI held even when adjusted for other known risk factors including smoking, diabetes, blood pressure, and serum cholesterol. ABSI correlation with mortality hazard held across the range of age, sex, and BMI, and for both white and black ethnicities (but not for Mexican ethnicity), and was not weakened by excluding deaths from the first 3 yr of follow-up.ConclusionsBody shape, as measured by ABSI, appears to be a substantial risk factor for premature mortality in the general population derivable from basic clinical measurements. ABSI expresses the excess risk from high WC in a convenient form that is complementary to BMI and to other known risk factors.

Project description:Weathering in the critical zone causes volumetric strain and mass loss, thereby creating subsurface porosity that is vital to overlying ecosystems. We used geochemical and geophysical measurements to quantify the relative importance of volumetric strain and mass loss---the physical and chemical components of porosity---in weathering of granitic saprolite of the southern Sierra Nevada, California, USA. Porosity and strain decrease with depth and imply that saprolite more than doubles in volume during exhumation to the surface by erosion. Chemical depletion is relatively uniform, indicating that changes in porosity are dominated by processes that cause strain with little mass loss. Strain-induced porosity production at our site may arise from root wedging, biotite weathering, frost cracking, and the opening of fractures under ambient topographic stresses. Our analysis challenges the conventional view that volumetric strain can be assumed to be negligible as a porosity-producing mechanism in saprolite.

Project description:Heat waves are often termed as the silent killer and have become even more important as recent studies suggest that the heat wave have become second most devastating extreme weather events in terms of human deaths and losses. It is also been largely realised by scientific community that it is not just the high temperatures which are responsible for the gruesome effect of heat waves but several other meteorological parameters play a vital role in aggravating the impact and causing much more damages. In view of the above the attention of scientific community, weather forecasters as well as disaster managers has shifted to also take into account the different meteorological parameters like maximum and minimum temperatures, relative humidity, wind speed, duration/spell of heat waves and its intensity which are aggravating the impact of heat stress. In this background, this study is undertaken as an attempt to quantify the effect of different meteorological parameters on heat wave on different regions of India for different summer months (March, April, May and June). In this study the impact of individual meteorological parameter as well their cumulative effect is studied based on data of 30 years (1981-2010) for 300 stations. The effect of different meteorological parameters is identified for different months for different regions of the country. Also the cumulative scores are calculated for different regions considering different meteorological parameters, as a first initiative to perform heat hazard analysis and zonation over the entire country. This could serve as initial step for planning mitigation and adaptation strategies throughout the country. These scores as thresholds for different regions may be also useful for operational forecaster's for early impact based warning services as well as for the disaster managers, for taking effective and timely actions.

Project description:ObjectivesAdult body mass (MB) empirically scales as height (Ht) squared (MB ∝ Ht(2) ), but does regional body mass and body composition as a whole also scale as Ht(2) ? This question is relevant to a wide range of biological topics, including interpretation of body mass index (BMI).MethodsDual-energy X-ray absorptiometry (DXA) was used to quantify regional body mass [head (MH), trunk, arms, and legs] and whole-body composition [fat, lean soft tissue (LST), and bone mineral content (BMC)] in non-Hispanic (NH) white, NH black, Mexican American, and Korean adults participating in the National Health and Nutrition Examination Survey (NHANES; n = 17,126) and Korean NHANES (n = 8,942). Regression models were developed to establish Ht scaling powers for each measured component with adjustments for age and adiposity.ResultsExploratory analyses revealed a consistent scaling pattern across men and women of the four population groups: regional mass powers, head (∼0.8-1) < arms and trunk (∼1.8-2.3) < legs (∼2.3-2.6); and body composition, LST (∼2.0-2.3) < BMC (∼2.1-2.4). Small sex and population differences in scaling powers were also observed. As body mass scaled uniformly across the eight sex and population groups as Ht(∼2) , tall and short subjects differed in body shape (e.g., MH/MB ∝ Ht(-∼1) ) and composition.ConclusionsAdult human body shape and relative composition are a function of body size as represented by stature, a finding that reveals a previously unrecognized phenotypic heterogeneity as defined by BMI. These observations provide new pathways for exploring mechanisms governing the interrelations between adult stature, body morphology, biomechanics, and metabolism.

Project description:Single-index models are becoming increasingly popular in many scientific applications as they offer the advantages of flexibility in regression modeling as well as interpretable covariate effects. In the context of survival analysis, the single-index hazards models are natural extensions of the Cox proportional hazards models. In this paper, we propose a novel estimation procedure for single-index hazard models under a monotone constraint of the index. We apply the profile likelihood method to obtain the semiparametric maximum likelihood estimator, where the novelty of the estimation procedure lies in estimating the unknown monotone link function by embedding the problem in isotonic regression with exponentially distributed random variables. The consistency of the proposed semiparametric maximum likelihood estimator is established under suitable regularity conditions. Numerical simulations are conducted to examine the finite-sample performance of the proposed method. An analysis of breast cancer data is presented for illustration.

Project description:'Cell Painting' is an imaging-based high-throughput phenotypic profiling (HTPP) method in which cultured cells are fluorescently labeled to visualize subcellular structures (i.e., nucleus, nucleoli, endoplasmic reticulum, cytoskeleton, Golgi apparatus / plasma membrane and mitochondria) and to quantify morphological changes in response to chemicals or other perturbagens. HTPP is a high-throughput and cost-effective bioactivity screening method that detects effects associated with many different molecular mechanisms in an untargeted manner, enabling rapid in vitro hazard assessment for thousands of chemicals. Here, 1201 chemicals from the ToxCast library were screened in concentration-response up to ∼100 μM in human U-2 OS cells using HTPP. A phenotype altering concentration (PAC) was estimated for chemicals active in the tested range. PACs tended to be higher than lower bound potency values estimated from a broad collection of targeted high-throughput assays, but lower than the threshold for cytotoxicity. In vitro to in vivo extrapolation (IVIVE) was used to estimate administered equivalent doses (AEDs) based on PACs for comparison to human exposure predictions. AEDs for 18/412 chemicals overlapped with predicted human exposures. Phenotypic profile information was also leveraged to identify putative mechanisms of action and group chemicals. Of 58 known nuclear receptor modulators, only glucocorticoids and retinoids produced characteristic profiles; and both receptor types are expressed in U-2 OS cells. Thirteen chemicals with profile similarity to glucocorticoids were tested in a secondary screen and one chemical, pyrene, was confirmed by an orthogonal gene expression assay as a novel putative GR modulating chemical. Most active chemicals demonstrated profiles not associated with a known mechanism-of-action. However, many structurally related chemicals produced similar profiles, with exceptions such as diniconazole, whose profile differed from other active conazoles. Overall, the present study demonstrates how HTPP can be applied in screening-level chemical assessments through a series of examples and brief case studies.

Project description:BackgroundTraditional animal toxicity tests can be time and resource intensive, thereby limiting the number of chemicals that can be comprehensively tested for potential hazards to humans and/or to the environment.ObjectiveWe compared several types of data to demonstrate how alternative models can be used to inform both human and ecological risk assessment.MethodsWe reviewed and compared data derived from high throughput in vitro assays to fish reproductive tests for seven chemicals. We investigated whether human-focused assays can be predictive of chemical hazards in the environment. We examined how conserved pathways enable the use of nonmammalian models, such as fathead minnow, zebrafish, and Xenopus laevis, to understand modes of action and to screen for chemical risks to humans.ResultsWe examined how dose-dependent responses of zebrafish embryos exposed to flusilazole can be extrapolated, using pathway point of departure data and reverse toxicokinetics, to obtain human oral dose hazard values that are similar to published mammalian chronic toxicity values for the chemical. We also examined how development/safety data for human health can be used to help assess potential risks of pharmaceuticals to nontarget species in the environment.DiscussionUsing several examples, we demonstrate that pathway-based analysis of chemical effects provides new opportunities to use alternative models (nonmammalian species, in vitro tests) to support decision making while reducing animal use and associated costs.ConclusionsThese analyses and examples demonstrate how alternative models can be used to reduce cost and animal use while being protective of both human and ecological health.

Project description:Perception of rock condition (RC) is a challenge in tunnel boring machine (TBM) construction due to lack of space and time to observe and detect RC. To overcome this problem, this study aims to extract a new rock fragmentation index (RFI) that can reflect RC from real-time rock fragmentation data of the TBM. First, a comprehensive review of existing rock fragmentation models is conducted, which leads to some candidate RFIs that can reflect RC. Next, these candidate RFIs are investigated using data from 12,237 samples from a well-monitored tunnel boring process of the TBM in a 20,198 m tunnel. Further, a new RFI system is recommended as the parameter involving the optimal models. Finally, a preliminary study of the relationship between these RFIs and RC is carried out, and it is shown that these RFIs can reflect RC to a large extent. In the TBM boring process, these RFIs can be extracted from real-time TBM fragmentation data and used to predict the RC in the field. Therefore, the challenge of RC perception is solved with this new RFI system. The new RFI system offers significant potential for the real-time rock classification, prediction of the surrounding rock collapse potential, and selection of control parameters or support measures during TBM construction. This will be the key to improving TBM construction performance.

Project description:The development of deep geotechnical engineering is restricted by the complex geological conditions of deep rock masses and the unknown creep mechanism of rock in water-rich environments. To study the shear creep deformation law of the anchoring rock mass under different water content conditions, marble was used as the bedrock to make anchoring specimens, and shear creep tests of the anchoring rock mass under different water contents were carried out. The influence of water content on rock rheological characteristics is explored by analysing the related mechanical properties of the anchorage rock mass. The coupling model of the anchorage rock mass can be obtained by connecting the nonlinear rheological element and the coupling model of the anchorage rock mass in series. Related studies show that (1) shear creep curves of anchorage rock masses under different water contents have typical creep characteristics, including decay, stability and acceleration stages. The creep deformation of the specimens can be improved with increasing moisture content. (2) The long-term strength of the anchorage rock mass shows an opposite change law with increasing water content. The creep rate of the curve increases gradually with increasing water content. The creep rate curve shows a U-shaped change under high stress. (3) The nonlinear rheological element can explain the creep deformation law of rock in the acceleration stage. By connecting the nonlinear rheological element with the coupled model of anchoring rock mass in series, the coupled model of water‒rock under water cut conditions can be obtained. The model can be used to study and analyse the whole process of shear creep of an anchored rock mass under different water contents. This study can provide theoretical support for the stability analysis of anchor support tunnel engineering under water cut conditions.

Project description:Concentrating cells from aqueous samples is a common requirement for the enumeration of biomass, investigations of microbial diversity and detection of relatively rare organisms in the environment. Accurately representing the initial sampled environments in the concentrated cells is of particular importance when the subsequent analyses have tangible environmental, economic and societal consequences, as is the case with environmental exposure and risk assessment of chemicals. This study investigated the potential use of four different cell concentration methods: centrifugation, membrane filtration, tangential flow filtration and column colonisation. These methods were assessed against a series of scientific and practical criteria, including: similarity of concentrated community to initial environmental sample; cell concentration achieved; biodegradation test outcome; sample throughput; and capital and maintenance costs. All methods increased cell concentration by as little as 10-fold to as much as 1000-fold. DGGE and 454 pyrosequencing analysis showed concentrated communities to have >60% similarity to each other, and the initial sample. There was a general trend for a more reliable assessment of 4-nitrophenol biodegradation in 96-well plate biodegradation assays, with increasing cell concentration. Based on the selection criteria, it is recommended that there is not one concentration method fit for all purposes, rather, the appropriate method should be selected on a case-by-case basis. Membrane filtration would be the most suitable method for low sample volumes; the increased throughput capacity of tangential flow filtration renders it most suitable for large volumes; and centrifugation is most suitable for samples with high initial biomass concentrations. The poor similarity in microbial community composition of the column colonised samples compared to the initial samples, suggested a concentration basis; this combined with its low sample throughput precluded this approach for future concentration studies of planktonic bacterial samples.
