Oxygen Absorption in Electrocatalyst Layers Detected by Scanning Electrochemical Microscopy.

ABSTRACT: Scanning electrochemical microscopy (SECM) is able to track the local electrochemical activity of an electrolyte-immersed substrate employing an ultra-micro-electrode (UME) in micrometer-scale spatial resolution. In this study, SECM is employed to investigate the presence of oxygen in the electrocatalyst layers of polymer electrolyte membrane fuel cells and electrolyzers. Approach curves on electrocatalyst layers with the tip potential set for oxygen reduction reveal that a significant amount of oxygen is absorbed in the catalyst layer. We confirm that the coexistence of Nafion ionomer and carbon black leads to oxygen confinement. It is suggested that this oxygen is confined within the hydrophobic parts of the self-assembled Nafion on the graphitic surfaces of the carbon black.

Simultaneous Scanning Electrochemical Microscopy and UV-Vis Absorption Spectroelectrochemistry.

Project description:The combination of instrumental techniques allows obtaining precise and reliable information about the reactions taking place at the electrode/solution interface. Although UV-Vis absorption spectroelectrochemistry (UV-Vis SEC) provides a molecular insight about the species involved in the electrode process, obtaining information about the redox state of the products generated in this process is not always accessible by this technique. In this sense, scanning electrochemical microscopy (SECM) has a clear advantage, since it provides additional information on the oxidation state of the intermediates/products. Therefore, the combination of these two techniques facilitates obtaining a more complete picture of the electrochemical reaction studied from two different points of view, but under exactly the same experimental conditions. In this work, the combination of UV-Vis SEC in parallel configuration and SECM is carried out for the first time. This new technique allows distinguishing between those species that are electrochemically active and, at the same time, exhibit changes in the UV-Vis absorption spectra during the electrochemical reaction. The new experimental setup is first validated using ferrocenemethanol as a standard probe, concomitantly obtaining spectroscopic and electrochemical information that accurately describes the oxidation process. Finally, the strength of this combined technique is demonstrated by studying the antioxidant activity of o-vanillin (o-HVa) in the presence of electrogenerated superoxide. The information extracted from the new UV-Vis SEC/SECM technique makes it possible to identify, beyond any doubt, not only the origin of the electrochemical signals recorded in the SECM tip but also to evaluate the antioxidant effect of o-HVa at different concentrations.

Evaluation of local oxygen flux produced by photoelectrochemical hydroxide oxidation by scanning electrochemical microscopy.

Project description:Several in-situ electrochemical approaches have been developed for performing a localized photoelectrochemical investigation of the photoanode. One of the techniques is scanning electrochemical microscopy (SECM), which probes local heterogeneous reaction kinetics and fluxes of generated species. In traditional SECM analysis of photocatalysts, evaluation of the influence of radiation on the rate of studied reaction requires an additional dark background experiment. Here, using SECM and an inverted optical microscope, we demonstrate the determination of O2 flux caused by light-driven photoelectrocatalytic water splitting. Photocatalytic signal and dark background are recorded in a single SECM image. We used an indium tin oxide electrode modified with hematite (α-Fe2O3) by electrodeposition as a model sample. The light-driven flux of oxygen is calculated by analysis of SECM image recorded in substrate generation/tip collection mode. In photoelectrochemistry, the qualitative and quantitative knowledge of oxygen evolution will open new doors for understanding the local effects of dopants and hole scavengers in a straightforward and conventional manner.

Nanoscale electrochemical charge transfer kinetics investigated by electrochemical scanning microwave microscopy.

Project description:We show how microwave microscopy can be used to probe local charge transfer reactions with unprecedented sensitivity, visualizing surface reactions with only a few hundred molecules involved. While microwaves are too fast under classical conditions to interact and sense electrochemical processes, this is different at the nanoscale, where our heterodyne microwave sensing method allows for highly sensitive local cyclic voltammetry (LCV) and local electrochemical impedance spectroscopy (LEIS). LCV and LEIS allow for precise measurement of the localized charge transfer kinetics, as illustrated in this study for a ferrocene self-assembled monolayer immersed in an electrolyte. The theoretical analysis presented here enables a consistent mapping of the faradaic kinetics and the parasitic contributions (nonfaradaic) to be spectrally resolved and subtracted. In particular, this methodology reveals an undistorted assessment of accessible redox site density of states associated with faradaic capacitance, fractional surface coverage and electron transfer kinetics at the nanoscale. The developed methodology opens a new perspective on comprehending electrochemical reactivity at the nanoscale.

Scanning Electrochemical Microscopy of Electrically Heated Wire Substrates.

Project description:We report a new configuration for enhancing the performance of scanning electrochemical microscopy (SECM) via heating of the substrate electrode. A flattened Pt microwire was employed as the substrate electrode. The substrate was heated by an alternating current (AC), resulting in an increased mass transfer between the wire surface and the bulk solution. The electrochemical response of the Pt wire during heating was investigated by means of cyclic voltammetry (CV). The open circuit potential (OCP) of the wire was recorded over time, while varied heating currents were applied to investigate the time needed for establishing steady-state conditions. Diffusion layer studies were carried out by performing probe approach curves (PACs) for various measuring modes of SECM. Finally, imaging studies of a heated substrate electrode surface, applying feedback, substrate generation/tip collection (SG/TC), and the competition mode of SECM, were performed and compared with room temperature results.

Metal halide perovskite layers studied by scanning transmission X-ray microscopy.

Project description:We describe the investigation of metal halide perovskite layers, particularly CH3NH3PbI3 used in photovoltaic applications, by soft X-ray scanning transmission X-ray microscopy (STXM). Relevant reference spectra were used to fit the experimental data using singular value decomposition. The distribution of key elements Pb, I, and O was determined throughout the layer stack of two samples prepared by wet process. One sample was chosen to undergo electrical biasing. Spectral data shows the ability of STXM to provide relevant chemical information for these samples. We found the results to be in good agreement with the sample history, both regarding the deposition sequence and the degradation of the perovskite material.

Integration of Scanning Electrochemical Microscopy and Scanning Electrochemical Cell Microscopy in a Bifunctional Nanopipette toward Simultaneous Mapping of Activity and Selectivity in Electrocatalysis.

Project description:Scanning electrochemical microscopy (SECM) and scanning electrochemical cell microscopy (SECCM) were integrated in a single bifunctional probe for simultaneous mapping of the oxygen reduction current and the oxidation current of the produced H2O2. The dual probe is fabricated from a double-barrel θ capillary, comprising one open barrel filled with the electrolyte and another filled with pyrolytic carbon. Pt is deposited with a gas injection system (GIS) at the end of the carbon barrel. The probe integrates the advantages of both SECM and SECCM by forming an electrochemical droplet cell that embeds the Pt working electrode of the carbon barrel directly into the electrolyte meniscus formed upon sample contact from the electrolyte barrel. The versatility of the dual probe is demonstrated by mapping the oxygen reduction reaction (ORR) current and the H2O2 oxidation current of a Pt microstrip on a gold substrate. This allows simultaneous localized electrochemical measurements, highlighting the potential of the dual probe for broader applications in characterizing the electrocatalytic properties of materials.

Single-Cell Migration as Studied by Scanning Electrochemical Microscopy.

Project description:Scanning electrochemical microscopy (SECM) was used to study the migration of single live head and neck cancer cells (SCC25). The newly developed graphite paste ultramicroelectrode (UME) showed significantly less fouling in comparison to a 10 μm Pt-UME and thus could be used to monitor and track the migration pattern of a single cell. We also used SECM probe scan curves to measure the morphology (height and diameter) of a single live cancer cell during cellular migration and determined these dimensions to be 11 ± 4 μm and 40 ± 10 μm, respectively. The migration study revealed that cells within the same cell line had a heterogeneous migration pattern (migration and stationary) with an estimated migration speed of 8 ± 3 μm/h. However, serum-starved synchronized cells of the same line were found to have a non-heterogeneous cellular migration pattern with a speed of 9 ± 3 μm/h. Thus, this non-invasive SECM-based technique could potentially be expanded to other cell lines to study cellular biomechanics for improved understanding of the structure-function relationship at the level of a single cell.

Scanning electrochemical microscopy of individual single-walled carbon nanotubes.

Project description:Here we report on the novel application of scanning electrochemical microscopy (SECM) to enable spatially resolved electrochemical characterization of individual single-walled carbon nanotubes (SWNTs). The feedback imaging mode of SECM was employed to detect a pristine SWNT (approximately 1.6 nm in diameter and approximately 2 mm in length) grown horizontally on a SiO(2) surface by chemical vapor deposition. The resulting image demonstrates that the individual nanotube under an unbiased condition is highly active for the redox reaction of ferrocenylmethyltrimethylammonium used as a mediator. Micrometer-scale resolution of the image is determined by the diameter of a disk-shaped SECM probe rather than by the nanotube diameter as assessed using 1.5 and 10 microm diameter probes. Interestingly, the long SWNT is readily detectable using the larger probe although the active SWNT covers only approximately 0.05% of the insulating surface just under the tip. This high sensitivity of the SECM feedback method is ascribed to efficient mass transport and facile electron transfer at the individual SWNT.

Scanning Electrochemical Cell Microscopy Investigation of Single ZIF-Derived Nanocomposite Particles as Electrocatalysts for Oxygen Evolution in Alkaline Media.

Project description:"Single entity" measurements are central for an improved understanding of the function of nanoparticle-based electrocatalysts without interference arising from mass transfer limitations and local changes of educt concentration or the pH value. We report a scanning electrochemical cell microscopy (SECCM) investigation of zeolitic imidazolate framework (ZIF-67)-derived Co-N-doped C composite particles with respect to the oxygen evolution reaction (OER). Surmounting the surface wetting issues as well as the potential drift through the use of a non-interfering Os complex as free-diffusing internal redox potential standard, SECCM could be successfully applied in alkaline media. SECCM mapping reveals activity differences relative to the number of particles in the wetted area of the droplet landing zone. The turnover frequency (TOF) is 0.25 to 1.5 s-1 at potentials between 1.7 and 1.8 V vs. RHE, respectively, based on the number of Co atoms in each particle. Consistent values at locations with varying number of particles demonstrates OER performance devoid of macroscopic film effects.

Menadione metabolism to thiodione in hepatoblastoma by scanning electrochemical microscopy.

Project description:The cytotoxicity of menadione on hepatocytes was studied by using the substrate generation/tip collection mode of scanning electrochemical microscopy by exposing the cells to menadione and detecting the menadione-S-glutathione conjugate (thiodione) that is formed during the cellular detoxication process and is exported from the cell by an ATP-dependent pump. This efflux was electrochemically detected and allowed scanning electrochemical microscopy monitoring and imaging of single cells and groups of highly confluent live cells. Based on a constant flux model, approximately 6 x 10(6) molecules of thiodione per cell per second are exported from monolayer cultures of Hep G2 cells.
