Project description:The Guayas estuary is an ecologically and economically vital, large estuarine system located on the western coast of South America. A suite of ∼400 water samples was collected and analyzed to enable investigation of the sources of water types within this estuary, as well as environmental processes active within various portions of it. These samples were obtained at sites distributed across the major areas of the inner and outer portions of the estuary at the ends of consecutive dry (December/January) and rainy (May) seasons. At each site, Van Dorn bottles were lowered into the water from a boat (or bridge) and then triggered when they reached specified depths. When the Van Dorn bottles were brought back aboard the boat, aliquots of water from them were promptly sealed in glass containers for later analysis. These were transported to onshore laboratories where their salinities, and their oxygen (δ18O) and hydrogen (δ2H) isotopic compositions were measured. Established analytical procedures and standards were employed to obtain a robust set of resultant data. The samples, analyses, and data presented here support the associated research article, "Stable isotope evidence for the origins of waters in the Guayas estuary and Gulf of Guayaquil" [1], to which readers are referred for interpretation.

Project description:Edge effects represent an inevitable and important consequence of habitat loss and fragmentation. These effects include changes in microclimate, solar radiation, or temperature. Such abiotic effects can, in turn, impact biotic factors. They can have a substantial impact on species, communities, and ecosystems. Here we examine clinal variations in stable carbon and nitrogen isotope values for trees along an edge-interior gradient in the dry deciduous forest at Ankarafantsika National Park. We predicted that soil respiration and differences in solar irradiance would result in stratified ?¹³C values where leaves collected close to the forest floor would have lower ?¹³C values than those growing higher up in the canopy. We also anticipated that plants growing at the savannah-forest boundary would have higher ?¹³C and ?¹?N values than plants growing in the forest interior. As expected, we detected a small but significant canopy effect. Leaves growing below 2 m from the forest floor exhibit ?¹³C values that are, on average, 1.1‰ lower than those growing above this threshold. We did not, however, find any relationship between foliar ?¹³C and distance from the edge. Unpredictably, we detected a striking positive relationship between foliar ?¹?N values and increasing distance into the forest interior. Variability in physiology among species, anthropogenic influence, organic input, and rooting depth cannot adequately explain this trend. Instead, this unexpected relationship most likely reflects decreasing nutrient or water availability, or a shift in N-sources with increasing distance from the savannah. Unlike most forest communities, the trees at Ampijoroa are growing in nutrient-limited sands. In addition to being nutrient poor, these well-drained soils likely decrease the amount of soil water available to forest vegetation. Continued research on plant responses to edge effects will improve our understanding of the conservation biology of forest ecosystems in Madagascar.

Project description:Stable isotope analysis (SIA) is an important tool for investigation of animal dietary habits for determination of feeding niche. Ideally, fresh samples should be used for isotopic analysis, but logistics frequently demands preservation of organisms for analysis at a later time. The goal of this study was to establish the best methodology for preserving forest litter-dwelling crickets for later SIA analysis without altering results. We collected two cricket species, Phoremia sp. and Mellopsis doucasae, from which we prepared 70 samples per species, divided among seven treatments: (i) freshly processed (control); preserved in fuel ethanol for (ii) 15 and (iii) 60 days; preserved in commercial ethanol for (iv) 15 and (v) 60 days; fresh material frozen for (vi) 15 and (vii) 60 days. After oven drying, samples were analyzed for δ15N, δ13C values, N(%), C(%) and C/N atomic values using continuous flow isotope ratio mass spectrometry. All preservation methods tested, significantly impacted δ13C and δ15N and C/N atomic values. Chemical preservatives caused δ13C enrichment as great as 1.5‰, and δ15N enrichment as great as 0.9‰; the one exception was M. doucasae stored in ethanol for 15 days, which had δ15N depletion up to 1.8‰. Freezing depleted δ13C and δ15N by up to 0.7 and 2.2‰, respectively. C/N atomic values decreased when stored in ethanol, and increased when frozen for 60 days for both cricket species. Our results indicate that all preservation methods tested in this study altered at least one of the tested isotope values when compared to fresh material (controls). We conclude that only freshly processed material provides adequate SIA results for litter-dwelling crickets.

Project description:Seafood is a valuable source of nutrients important for fetal development. However, seafood consumption is the main route of exposure to monomethyl mercury (MeHg+) for humans. MeHg+ is highly bioavailable and potentially adversely affects fetal neurodevelopment. MeHg+ exposure from fish consumption varies significantly by age and trophic level of fish consumed as well as the frequency and amount of fish consumed. This study investigates total Hg concentrations ([THg]) in hair segments of pregnant Mexican women in relation to (1) self-reported frequency of fish and shellfish consumption, (2) maternal trophic level and marine diet contributions, determined using hair carbon (C) and nitrogen (N) stable isotopes, and (3) relates [THg] to various hair advisory thresholds. We also examined whether variation in C and N isotope values is explained by self-reported frequency of fish and shellfish consumption. A significant proportion of hair samples had [THg] higher than suggested agency thresholds and, for women within the range of the various advisory thresholds (1-20 µg g-1), the specific statistic used and threshold applied are important considerations for assessing and communicating risk. Individuals enriched in 15N (δ15N values) had higher [THg] as did individuals that reported consuming fish and shellfish more frequently, suggesting that variation in [THg] can be explained by both consumer reported diet and diet as determined by C and N stable isotope assessment. However, at higher reported fish consumption levels the trophic level is maintained while [THg] is paradoxically lower. This suggests that THg exposure and assimilation are more complicated in higher fish frequency consumption categories. [THg] is more variable at the higher concentrations, possibly indicating some exposure to non-dietary Hg, heritable variations affecting Hg toxicodynamics, and BMI and tobacco exposure factors as outlined in our companion paper.

Project description:Rising concentrations of nitrous oxide (N2O) in the atmosphere are causing widespread concern because this trace gas plays a key role in the destruction of stratospheric ozone and it is a strong greenhouse gas. The successful mitigation of N2O emissions requires a solid understanding of the relative importance of all N2O sources and sinks. Stable isotope ratio measurements (δ15N-N2O and δ18O-N2O), including the intramolecular distribution of 15N (site preference), are one way to track different sources if they are isotopically distinct. 'Top-down' isotope mass-balance studies have had limited success balancing the global N2O budget thus far because the isotopic signatures of soil, freshwater, and marine sources are poorly constrained and a comprehensive analysis of global N2O stable isotope measurements has not been done. Here we used a robust analysis of all available in situ measurements to define key global N2O sources. We showed that the marine source is isotopically distinct from soil and freshwater N2O (the continental source). Further, the global average source (sum of all natural and anthropogenic sources) is largely controlled by soils and freshwaters. These findings substantiate past modelling studies that relied on several assumptions about the global N2O cycle. Finally, a two-box-model and a Bayesian isotope mixing model revealed marine and continental N2O sources have relative contributions of 24-26% and 74-76% to the total, respectively. Further, the Bayesian modeling exercise indicated the N2O flux from freshwaters may be much larger than currently thought.

Project description:The tooth enamel from the human remains of ten archaeological individuals belonging to a chalcolithic site at Inamgaon, District Pune, Maharashtra, were analysed for stable carbon and oxygen isotope compositions. The human remains of the involved individuals come from three consecutive periods: Period I (1600-1400 BC; n = 2), Period II (1400-1000 BC; n = 4), and Period III (1000-700 BC; n = 4). Enamel carbonate of twenty teeth (n = 20), two from each individual, were analysed to understand the inter- and intra-individual variations in isotope ratios across the three habitational periods. The acquired dataset will help in understanding isotope baseline values for the region in the prehistoric context. The subsequent research works in the region can reuse our data in collation with other datasets for comparative investigations.

Project description:Measures (e.g. ?15N, ?13C, %C, %N and C:N) derived from animal tissues are commonly used to estimate diets and trophic interactions. Since tissue samples are often exposed to air or kept chilled in ice over a short-term during sample preparation, they may degrade. Herein, we hypothesize that tissue decomposition will cause changes in these measures. In this study, we kept marine fish, crustacean and mollusc tissues in air or ice over 120 h (5 days). We found that tissue decomposition in air enriched ?15N (range 0.6‰ to 1.3‰) and ?13C (0.2‰ to 0.4‰), decreased %N (0.47 to 3.43 percentage points from staring values of ~13%) and %C (4.53 to 8.29 percentage points from starting values of ~43%), and subsequently increased C:N ratio (0.14 to 0.75). In air, while such changes to ?13C were relatively minor and therefore likely tolerable, changes in ?15N, %N, %C and C:N ratio should be interpreted with caution. Ice effectively reduced the extent to which decomposition enriched ?15N (? 0.4‰) and ?13C (? 0.2‰), and eliminated decomposition in C:N ratio, %N and %C. In our second experiment, for fish tissues in either air or ice over 120 h, we observed no effects of decomposition on relationships between lipid content, C:N ratio, and ??13C (change in ?13C after lipid removal), which are employed to correct ?13C for samples containing lipid. We also confirmed that lipid in tissues caused large errors when estimating ?13C (mean ± standard error = -1.8‰ ± 0.1‰, range -0.6‰ to -3.8‰), and showed both lipid extraction and mathematical correction performed equally well to correct for lipids when estimating ?13C. We, therefore, recommend that specimens of marine animals should be kept in ice during sample preparation for a short-term, as it is an effective means for minimizing changes of the stable isotope measures in their tissue.

Project description:The evaluation of extraction protocols for untargeted metabolomics approaches is still difficult. We have applied a novel stable isotope-assisted workflow for untargeted LC-HRMS-based plant metabolomics , which allows for the first time every detected feature to be considered for method evaluation. The efficiency and complementarity of commonly used extraction solvents, namely 1 + 3 (v/v) mixtures of water and selected organic solvents (methanol, acetonitrile or methanol/acetonitrile 1 + 1 (v/v)), with and without the addition of 0.1% (v/v) formic acid were compared. Four different wheat organs were sampled, extracted and analysed by LC-HRMS. Data evaluation was performed with the in-house-developed MetExtract II software and R. With all tested solvents a total of 871 metabolites were extracted in ear, 785 in stem, 733 in leaf and 517 in root samples, respectively. Between 48% (stem) and 57% (ear) of the metabolites detected in a particular organ were found with all extraction mixtures, and 127 of 996 metabolites were consistently shared between all extraction agent/organ combinations. In aqueous methanol, acidification with formic acid led to pronounced pH dependency regarding the precision of metabolite abundance and the number of detectable metabolites, whereas extracts of acetonitrile-containing mixtures were less affected. Moreover, methanol and acetonitrile have been found to be complementary with respect to extraction efficiency. Interestingly, the beneficial properties of both solvents can be combined by the use of a water-methanol-acetonitrile mixture for global metabolite extraction instead of aqueous methanol or aqueous acetonitrile alone.

Project description:Stable carbon isotopic data (δ13C) of 41 individual plant species was collected from long-term grazed and ungrazed pastures in Oklahoma, USA. These data can serve as a library of stable carbon isotope values for Southern mixed-grass prairie species. Seventeen warm-season (C4) and twenty-four cool-season (C3) plants were identified and collected from the United States Department of Agriculture (USDA) Southern Plains Experimental Range (SPER). Plant samples were dried at 55°C, and ground finely. The δ13C isotopic compositions were determined using a Europa Scientific automated nitrogen carbon analyzer (ANCA/NT) with a Solid/Liquid Preparation Module (Dumas combustion sample preparation system) coupled to a Europa 20-20 Stable isotope analyzer continuous flow isotope ratio mass spectrometer (Sercon Ltd, formerly Europa Scientific Ltd., Crewe, England). These data can be used as end members in isotopic mixing models or in paleoecology to correlate soil ages with plant species composition. Data from plant species provides information about soil organic carbon sequestration and possible long-term climate change.

Project description:BackgroundFish fin is a widely used, non-lethal sample material in studies using stable isotopes to assess the ecology of fishes. However, fish fin is composed of two distinct tissues (ray and membrane) which may have different stable isotope values and are not homogeneously distributed within a fin. As such, estimates of the stable isotope values of a fish may vary according to the section of fin sampled.MethodsTo assess the magnitude of this variation, we analysed carbon (δ13C), nitrogen (δ15N), hydrogen (δ2H) and oxygen (δ18O) stable isotopes of caudal fin from juvenile, riverine stages of Atlantic salmon (Salmo salar) and brown trout (Salmo trutta). Individual fins were sub-sectioned into tip, mid and base, of which a further subset were divided into ray and membrane.FindingsIsotope variation between fin sections, evident in all four elements, was primarily related to differences between ray and membrane. Base sections were13C depleted relative to tip (~1‰) with equivalent variation evident between ray and membrane. A similar trend was evident in δ2H, though the degree of variation was far greater (~10‰). Base and ray sections were 18O enriched (~2‰) relative to tip and membrane, respectively. Ray and membrane sections displayed longitudinal variation in 15N mirroring that of composite fin (~1‰), indicating that variation in15N values was likely related to ontogenetic variation.ConclusionsTo account for the effects of intra-fin variability in stable isotope analyses we suggest that researchers sampling fish fin, in increasing priority, 1) also analyse muscle (or liver) tissue from a subsample of fish to calibrate their data, or 2) standardize sampling by selecting tissue only from the extreme tip of a fin, or 3) homogenize fins prior to analysis.
