Project description:Antimicrobial resistances are emerging as one main threat to worldwide human health and are expected to kill 10 million people by 2050. Intensive livestock husbandry, along with biogas digestate, are considered as one of the biggest ARG reservoirs. Despite major concerns, little information is available on the diversity and abundance of various ARGs in small to large scale pig farms and biogas digestate slurry in Germany, followed by their consequent removal using microfiltration (MF)-nanofiltration (NF) process. Here, we report the identification and quantification of 189 ARGs in raw manure and digestate samples, out of which 66 ARGs were shared among manures and 53 ARGs were shared among both manure and digestate samples. The highest reported total ARG copy numbers in a single manure sampling site was 1.15 × 108 copies/100 µL. In addition, we found the absolute concentrations of 37 ARGs were above 105 copies/100 μL. Filtration results showed that the highly concentrated ARGs (except aminoglycoside resistance ARGs) in feed presented high log retention value (LRV) from 3 to as high as 5 after the MF-NF process. Additionally, LRV below 2 was noticed where the initial absolute ARG concentrations were ≤103 copies/100 μL. Therefore, ARG removal was found to be directly proportional to its initial concentration in the raw manure and in digestate samples. Consequently, some ARGs (tetH, strB) can still be found within the permeate of NF with up to 104 copies/100 μL.

Project description:Composting is recognized as an effective strategy for the sustainable use of organic wastes, but also as an important emission source of nitrous oxide (N2O) contributing to global warming. The effects of calcium superphosphate (CaSSP) on N2O production during composting are reported to be controversial, and the intrinsic microbial mechanism remains unclear. Here, a pig manure windrow composting experiment lasting for ~60 days was performed to evaluate the effects of CaSSP amendment (5%, w/w) on N2O fluxes in situ, and to determine the denitrifiers' response, and their driving factors. Results indicated that CaSSP amendment significantly reduced N2O emissions as compared to the control pile (maximum N2O emission rate reduced by 64.5% and total emission decreased by 49.8%). CaSSP amendment reduced the abundance of nirK gene encoding for nitrite reductase, while the abundance of nosZ gene (N2O reductase) was enriched. Finally, we built a schematic model and indicated that the abundance of nirK gene was likely to play a key role in mediating N2O production, which were correlated with NH4+-N and NO3--N changing responsive to CaSSP. Our finding implicates that CaSSP application could be a potential strategy for N2O mitigation in manure windrow composting, and the revealed microbial mechanism is helpful for deepening the understanding of the interaction among N-cycle functional genes, physicochemical factors, and greenhouse gases (GHG) emissions.

Project description:Global pork production has an annual growth of approximately 2.1%, and its economic and environmental impact are related with the treatment of waste in the production chain. There is little evidence of research advances to generate alternatives for using these wastes. The lack of research related to microalgae cultivation using digestate produced by porcine residues generates negative environmental impact, inadequate and inefficient technologies, low recovery and use of waste and loss of value and competitiveness in the market. The available literature focuses mainly on the treatment of anaerobic digestion liquid effluents for the removal of components, but not on the generation of value-added products. Therefore, there is a need to collect the available information, analyze it and propose other new methodologies. This article presents the information obtained from conducting a systematic review of the literature with a bibliometric and a comparative analysis; achieving an analysis of the temporal and geographical distribution, the main topics, the most influential players, the degree of maturity of the research and different strategies collected for microalgae-based swine manure digestate treatment. In this way, it was possible to capture an overview of the current state of the development of research focused on the use of digestate for the cultivation of microalgae, visualizing important aspects as the evolution of publications, identifying China and USA as the main players in research, biomass and wastewater as potential topics also Spirulina, Astaxanthin and beta-carotene as the main products based on microalgae. Thus, achieving an structure, organized and synthesized landscape of scientific and technological knowledge available for the proposal of investigations that allow the use of anaerobic digestion liquid effluents as cultivation medium for microalgae.

Project description:This study investigated the impact of composting substrate types on the bacterial community structure and dynamics during composting processes. To this end, pig manure (PM), chicken manure (CM), a mixture of PM and CM (PM + CM), and a mixture of PM, CM and anaerobic digestion residue (ADR) (PM + CM + ADR) were selected for thermophilic composting. The bacterial community structure and dynamics during the composting process were detected and analysed by polymerase chain reaction-denaturing gradient gel electrophoresis (DGGE) coupled with a statistic analysis. The physical-chemical analyses indicated that compared to single-material composting (PM, CM), co-composting (PM + CM, PM + CM + ADR) could promote the degradation of organic matter and strengthen the ability of conserving nitrogen. A DGGE profile and statistical analysis demonstrated that co-composting, especially PM + CM + ADR, could improve the bacterial community structure and functional diversity, even in the thermophilic stage. Therefore, co-composting could weaken the screening effect of high temperature on bacterial communities. Dominant sequencing analyses indicated a dramatic shift in the dominant bacterial communities from single-material composting to co-composting. Notably, compared with PM, PM + CM increased the quantity of xylan-degrading bacteria and reduced the quantity of human pathogens.

Project description:Manure and digestate liquid fractions are nutrient-rich effluents that can be fractionated and concentrated using membranes. However, these membranes tend to foul due to organic matter, solids, colloids, and inorganic compounds including calcium, ammonium, sodium, sulfur, potassium, phosphorus, and magnesium contained in the feed. This review paper is intended as a theoretical and practical tool for the decision-making process during design of membrane-based systems aiming at processing manure liquid fractions. Firstly, this review paper gives an overview of the main physico-chemical characteristics of manure and digestates. Furthermore, solid-liquid separation technologies are described and the complexity of the physico-chemical variables affecting the separation process is discussed. The main factors influencing membrane fouling mechanisms, morphology and characteristics are described, as well as techniques covering membrane inspection and foulant analysis. Secondly, the effects of the feed characteristics, membrane operating conditions (pressure, cross-flow velocity, temperature), pH, flocculation-coagulation and membrane cleaning on fouling and membrane performance are presented. Finally, a summary of techniques for specific recovery of ammonia-nitrogen, phosphorus and removal of heavy metals for farm effluents is also presented.
