Classification of Alzheimer's Disease Leveraging Multi-task Machine Learning Analysis of Speech and Eye-Movement Data.

ABSTRACT: Alzheimer's disease (AD) is a progressive neurodegenerative condition that results in impaired performance in multiple cognitive domains. Preclinical changes in eye movements and language can occur with the disease, and progress alongside worsening cognition. In this article, we present the results from a machine learning analysis of a novel multimodal dataset for AD classification. The cohort includes data from two novel tasks not previously assessed in classification models for AD (pupil fixation and description of a pleasant past experience), as well as two established tasks (picture description and paragraph reading). Our dataset includes language and eye movement data from 79 memory clinic patients with diagnoses of mild-moderate AD, mild cognitive impairment (MCI), or subjective memory complaints (SMC), and 83 older adult controls. The analysis of the individual novel tasks showed similar classification accuracy when compared to established tasks, demonstrating their discriminative ability for memory clinic patients. Fusing the multimodal data across tasks yielded the highest overall AUC of 0.83 ± 0.01, indicating that the data from novel tasks are complementary to established tasks.

Project description:Many machine learning and pattern classification methods have been applied to the diagnosis of Alzheimer's disease (AD) and its prodromal stage, i.e., mild cognitive impairment (MCI). Recently, rather than predicting categorical variables as in classification, several pattern regression methods have also been used to estimate continuous clinical variables from brain images. However, most existing regression methods focus on estimating multiple clinical variables separately and thus cannot utilize the intrinsic useful correlation information among different clinical variables. On the other hand, in those regression methods, only a single modality of data (usually only the structural MRI) is often used, without considering the complementary information that can be provided by different modalities. In this paper, we propose a general methodology, namely multi-modal multi-task (M3T) learning, to jointly predict multiple variables from multi-modal data. Here, the variables include not only the clinical variables used for regression but also the categorical variable used for classification, with different tasks corresponding to prediction of different variables. Specifically, our method contains two key components, i.e., (1) a multi-task feature selection which selects the common subset of relevant features for multiple variables from each modality, and (2) a multi-modal support vector machine which fuses the above-selected features from all modalities to predict multiple (regression and classification) variables. To validate our method, we perform two sets of experiments on ADNI baseline MRI, FDG-PET, and cerebrospinal fluid (CSF) data from 45 AD patients, 91 MCI patients, and 50 healthy controls (HC). In the first set of experiments, we estimate two clinical variables such as Mini Mental State Examination (MMSE) and Alzheimer's Disease Assessment Scale-Cognitive Subscale (ADAS-Cog), as well as one categorical variable (with value of 'AD', 'MCI' or 'HC'), from the baseline MRI, FDG-PET, and CSF data. In the second set of experiments, we predict the 2-year changes of MMSE and ADAS-Cog scores and also the conversion of MCI to AD from the baseline MRI, FDG-PET, and CSF data. The results on both sets of experiments demonstrate that our proposed M3T learning scheme can achieve better performance on both regression and classification tasks than the conventional learning methods.

Project description:Decoding movement related intentions is a key step to implement BMIs. Decoding EEG has been challenging due to its low spatial resolution and signal to noise ratio. Metric learning allows finding a representation of data in a way that captures a desired notion of similarity between data points. In this study, we investigate how metric learning can help finding a representation of the data to efficiently classify EEG movement and pre-movement intentions. We evaluate the effectiveness of the obtained representation by comparing classification the performance of a Support Vector Machine (SVM) as a classifier when trained on the original representation, called Euclidean, and representations obtained with three different metric learning algorithms, including Conditional Entropy Metric Learning (CEML), Neighborhood Component Analysis (NCA), and the Entropy Gap Metric Learning (EGML) algorithms. We examine different types of features, such as time and frequency components, which input to the metric learning algorithm, and both linear and non-linear SVM are applied to compare the classification accuracies on a publicly available EEG data set for two subjects (Subject B and C). Although metric learning algorithms do not increase the classification accuracies, their interpretability using an importance measure we define here, helps understanding data organization and how much each EEG channel contributes to the classification. In addition, among the metric learning algorithms we investigated, EGML shows the most robust performance due to its ability to compensate for differences in scale and correlations among variables. Furthermore, from the observed variations of the importance maps on the scalp and the classification accuracy, selecting an appropriate feature such as clipping the frequency range has a significant effect on the outcome of metric learning and subsequent classification. In our case, reducing the range of the frequency components to 0–5 Hz shows the best interpretability in both Subject B and C and classification accuracy for Subject C. Our experiments support potential benefits of using metric learning algorithms by providing visual explanation of the data projections that explain the inter class separations, using importance. This visualizes the contribution of features that can be related to brain function.

Project description:Alzheimer's disease (AD) is a progressive neurodegenerative disorder, accounting for nearly 60% of all dementia cases. The occurrence of the disease has been increasing rapidly in recent years. Presently about 46.8 million individuals suffer from AD worldwide. The current absence of effective treatment to reverse or stop AD progression highlights the importance of disease prevention and early diagnosis. Brain structural Magnetic Resonance Imaging (MRI) has been widely used for AD detection as it can display morphometric differences and cerebral structural changes. In this study, we built three machine learning-based MRI data classifiers to predict AD and infer the brain regions that contribute to disease development and progression. We then systematically compared the three distinct classifiers, which were constructed based on Support Vector Machine (SVM), 3D Very Deep Convolutional Network (VGGNet) and 3D Deep Residual Network (ResNet), respectively. To improve the performance of the deep learning classifiers, we applied a transfer learning strategy. The weights of a pre-trained model were transferred and adopted as the initial weights of our models. Transferring the learned features significantly reduced training time and increased network efficiency. The classification accuracy for AD subjects from elderly control subjects was 90%, 95%, and 95% for the SVM, VGGNet and ResNet classifiers, respectively. Gradient-weighted Class Activation Mapping (Grad-CAM) was employed to show discriminative regions that contributed most to the AD classification by utilizing the learned spatial information of the 3D-VGGNet and 3D-ResNet models. The resulted maps consistently highlighted several disease-associated brain regions, particularly the cerebellum which is a relatively neglected brain region in the present AD study. Overall, our comparisons suggested that the ResNet model provided the best classification performance as well as more accurate localization of disease-associated regions in the brain compared to the other two approaches.
