Synthesis of Macrocyclic Lactones and Dilactones Using Olive Oil.

ABSTRACT: Macrocyclic lactones have redolent characteristics of muscones that originate from the rectal musk organs of the musk deer. These lactones are the primary raw material in the flavor and fragrance industry and are also found within the cyclic frameworks of various bioactive molecules. Due to great demand, many efforts have been made for their synthesis; however, strategies generating a large number of macrocyclic analogues from renewable resources have not been fully realized and are urgently required. Here, we outline a sustainable, straightforward, and eco-friendly approach to synthesize high-valued macrocyclic lactones utilizing olive oil under greener reaction conditions. The outlined method allows us to turn biomass into valuable 12- to 29-membered lactones and dilactones.

Project description:Macrocyclic lactones can be prepared from lactams and hydroxyacid derivatives via an efficient 3- or 4-atom iterative ring expansion protocol. The products can also be expanded using amino acid-based linear fragments, meaning that macrocycles with precise sequences of hydroxy- and amino acids can be assembled in high yields by "growing" them from smaller rings, using a simple procedure in which high dilution is not required. The method should significantly expedite the practical synthesis of diverse nitrogen containing macrolide frameworks.

Formation of macrocyclic lactones in the Paterno-Buchi dimerization reaction.

Project description:Furan-2-ylmethyl 2-oxoacetates 1a,b, in which the furan ring and the carbonyl moiety were embedded intramolecularly, were synthesized from commercially available furan-2-ylmethanol and their photochemical reaction (h? > 290 nm) was investigated. Twelve-membered macrocyclic lactones 2a,b with C(i) symmetry including two oxetane-rings, which are the Paternò-Büchi dimerization products, were isolated in ca. 20% yield. The intramolecular cyclization products, such as 3-alkoxyoxetane and 2,7-dioxabicyclo[2.2.1]hept-5-ene derivatives, were not detected in the photolysate.

Treatment of MDR1 mutant dogs with macrocyclic lactones.

Project description:P-glycoprotein, encoded by the multidrug resistance gene MDR1, is an ATP-driven drug efflux pump which is highly expressed at the blood-brain barrier of vertebrates. Drug efflux of macrocyclic lactones by P-glycoprotein is highly relevant for the therapeutic safety of macrocyclic lactones, as thereby GABA-gated chloride channels, which are confined to the central nervous system in vertebrates, are protected from high drug concentrations that otherwise would induce neurological toxicity. A 4-bp deletion mutation exists in the MDR1 gene of many dog breeds such as the Collie and the Australian Shepherd, which results in the expression of a non-functional P-glycoprotein and is associated with multiple drug sensitivity. Accordingly, dogs with homozygous MDR1 mutation are in general prone to neurotoxicity by macrocyclic lactones due to their increased brain penetration. Nevertheless, treatment of these dogs with macrocyclic lactones does not inevitably result in neurological symptoms, since, the safety of treatment highly depends on the treatment indication, dosage, route of application, and the individual compound used as outlined in this review. Whereas all available macrocyclic lactones can safely be administered to MDR1 mutant dogs at doses usually used for heartworm prevention, these dogs will experience neurological toxicity following a high dose regimen which is common for mange treatment in dogs. Here, we review and discuss the neurotoxicological potential of different macrocyclic lactones as well as their treatment options in MDR1 mutant dogs.

Fraud detection and quality assessment of olive oil using ultrasound.

Project description:Today, food safety is recognized as one of the most important human priorities, so effective and new policies have been implemented to improve and develop the position of effective laws in the food industry. Extra virgin olive oil (EVOO) has many amazing benefits for human body's health. Due to the nutritional value and high price of EVOO, there is a lot of cheating in it. The ultrasound approach has many advantages in the food studies, and it is fast and nondestructive for quality evaluation. In this study, to fraud detection of EVOO four ultrasonic properties of oil in five levels of adulteration (5%, 10%, 20%, 35%, and 50%) were extracted. The 2 MHz ultrasonic probes were used in the DOI 1,000 STARMANS diagnostic ultrasonic device in a "probe holding mechanism." The four extracted ultrasonic features include the following: "percentage of amplitude reduction, time of flight (TOF), the difference between the first and second maximum amplitudes of the domain (in the time-amplitude diagram), and the ratio of the first and second maximum of amplitude." Seven classification algorithms including "Naïve Bayes, support vector machine, gradient boosting classifier, K-nearest neighbors, artificial neural network, logistic regression, and AdaBoost" were used to classify the preprocessed data. Results showed that the Naïve Bayes algorithm with 90.2% provided the highest accuracy among the others, and the support vector machine and gradient boosting classifier with 88.2% were in the next ranks.

Understanding Olive Oil Stability Using Filtration and High Hydrostatic Pressure.

Project description:Veiled extra virgin olive oil (VEVOO) is very attractive on the global market. A study was performed to highlight the role of different amounts of water and microorganisms on the evolution of VEVOO quality during storage, using the selective effects of the application of individual or combined filtration and high hydrostatic pressure (HHP) treatments. Four oil processing trials were carried out in four replicates, resulting in a full factorial design with two independent fixed factors: filtration and HPP treatments. The turbidity of all the olive oil samples was characterized. Furthermore, all the olive oil samples were analysed for legal parameters, volatile organic compounds and phenolic compounds during the storage tests. The microbial contamination in the presence of a high level of water activity (>0.6 Aw) was related to the formation of volatile aroma compounds, which were responsible for the "fusty" sensory defect. Furthermore, high water activity values were related to an increase in the hydrolytic degradation rate of the phenolic compounds. The oil turbidity has to be planned and controlled, starting from adjustment of the water content and application of good manufacturing practices.

Olive Oil Benefits from Sesame Oil Blending While Extra Virgin Olive Oil Resists Oxidation during Deep Frying.

Project description:Fresh potatoes were deep-fried in olive oil (OO), extra virgin olive oil (EVOO), and their blends with 5%, 10%, and 20% v/v sesame oil (SO). This is the first report on the use of sesame oil as a natural source of antioxidants during olive oil deep frying. The oil was evaluated for anisidine value (AV), free fatty acids (FFAs), extinction coefficient (K232 and K270), Trolox equivalent antioxidant capacity (TEAC), and total phenols (TPs) until the total polar compounds (TPCs) reached 25%. Sesame lignan transformations were monitored through reversed-phase HPLC. While the TPCs in olive oils increased at a steady rate, the addition of 5%, 10%, and 20% v/v SO delayed TPCs' formation for 1, 2, and 3 h, respectively. The addition of 5%, 10%, and 20% v/v SO increased the olive oil frying time by 1.5 h, 3.5 h, and 2.5 h, respectively. The addition of SO to OO reduced the secondary oxidation products' formation rate. The AV for EVOO was lower than OO and all tested blends, even those with EVOO. EVOO was more resistant to oxidation than OO, as measured by the TPCs and TEAC, while the frying time rose from 21.5 to 25.25 h when EVOO replaced OO. The increase in frying time for OO but not for EVOO, after SO addition, points to a niche market for EVOO in deep frying.

Proteomics of extra virgin olive oil (EVOO)

Project description:Proteins and peptides are minor components of vegetal oils. The presence of these compounds in virgin olive oil was first reported in 2001, but the nature of the olive oil proteome is still a puzzling question for food science researchers. In this project, we have compiled for a first time a comprehensive proteomic dataset of olive fruit and fungal proteins that are present at low but measurable concentrations in a vegetable oil from a crop of great agronomical relevance as olive (Olea europaea L.). Accurate mass nLC-MS data were collected in high definition direct data analysis (HD-DDA) mode using the ion mobility separation step. Protein identification was performed using the Mascot Server v2.2.07 software (Matrix Science) against an ad hoc database made of olive protein entries. Starting from this proteomic record, the impact of these proteins on olive oil stability and quality could be tested. Moreover, the effect of olive oil proteins on human health and their potential use as functional food components could be also evaluated. In addition, this dataset provides a resource for use in further functional comparisons across other vegetable oils, and also expands the proteomic resources to non-model species, thus also allowing further comparative inter-species studies.

Human in vivo nutrigenomic effect of olive oil

Project description:Olive oil is protective against risk factors for cardiovascular and cancer diseases. A nutrigenomic approach was performed to assess whether olive oil, the main fat of the Mediterranean diet modifies the gene expression in human peripheral blood mononuclear cells. Six healthy male volunteers ingested, at fasting state, 50 ml of olive oil, and continued with the same olive oil as a source of raw fat (25ml/day) during 3 weeks. Prior to intervention a 1-week washout period with sunflower oil as the only source of fat was followed. During the 3 days before, and on the intervention day, a very low phenolic compound diet was followed. At baseline (0h), at post ingestion (6h), and at fasting state after 3 weeks of sustained consumption of olive oil total RNA was isolated from PBMC.
