Project description:The coordination chemistry of bisphosphonates with Yb(3+) was investigated to evaluate the potential of the UV-vis based detection method using the Yb(3+)-pyrocatechol complexation reaction as a sensor for bisphosphonates. The complexation chemistry of Yb(3+) with phosphate and ATP analogs was previously described (E. Gaidamauskas, K. Saejueng, A.A. Holder, S. Bharuah, B.A. Kashemirov, D.C. Crans, C.E. McKenna, J. Biol. Inorg. Chem. 13 (2008) 1291-1299), and we here studied the complexation chemistry of bisphosphonates in this system. The spectrophotometric assay yields direct evidence for formation of a 4:3 metal to ligand complex at neutral pH. Direct evidence for Yb(3+):methylenebis(phosphonate) complexes with 1:1 and 1:2 stoichiometry was also obtained by potentiometry at acidic and basic pH. Direct evidence for complex formation was obtained using (1)H NMR spectroscopy although the stoichiometry was not accessed at neutral pH. Our results suggest that the spectroscopic observation of the YbPV complex can be used to conveniently measure concentrations of bisphosphonates down to 2-3 microM.

Project description:BackgroundFor clinicians, it is important to rely on accurate laboratory results for patient care and optimal use of health care resources. We sought to explore our observations that urine protein:creatinine ratios (PrCr) ≥30 mg/mmol are seen not infrequently associated with normal pregnancy outcome.MethodsUrine samples were collected prospectively from 160 pregnant women attending high-risk maternity clinics at a tertiary care facility. Urinary protein was measured using a pyrocatechol violet assay and urinary creatinine by an enzymatic method on Vitros analysers. Maternal/perinatal outcomes were abstracted from hospital records.Results91/233 (39.1%) samples had a PrCr ≥30 mg/mmol, especially when urinary creatinine concentration was <3 mM (94.1%) vs. ≥3 mM (16.4%) (p < 0.001). When using the last sample before delivery, 47/160 (29.4%) had a PrCr ≥30 mg/mmol in diluted urine vs. only 17/160 (15.4%) in more concentrated urine (p < 0.001); PrCr positive results were also more frequent among the 32 (20.0%) women with known normal pregnancy outcome (90.9% vs. 0) (p < 0.001). Using the same analyser, 0.12 g/L urinary protein was 'detected' in deionised water. Re-analysis of data from two cohorts revealed substantially less inflation of PrCr in dilute urine using a pyrogallol red assay.ConclusionsRandom urinary PrCr was overestimated in dilute urine when tested using a common pyrocatechol violet dye-based method. This effect was reduced in cohorts when pyrogallol red assays were used. False positive results can impact on diagnosis and patient care. This highlights the need for both clinical and laboratory quality improvement projects and standardization of laboratory protein measurement.

Project description:Metal complexation reactions can be used effectively as sensors to measure concentrations of phosphate and phosphate analogs. Recently, a method was described for the detection of phosphate or ATP in aqueous solution based on the displacement by these ligands of pyrocatechol violet (PV) from a putative 2:1 (Yb3+)2PV complex. We have not been able to reproduce this stoichiometry and report this work in order to correct the coordination chemistry upon which sensor applications are based. In our work, colorimetric and spectrophotometric detection of phosphate was confirmed qualitatively (blue PV+Yb3+; yellow+Pi); however, the sequence of visual changes on the titration of PV with 2 equiv. of Yb3+ and back titration with ATP as described previously could not be reproduced. In contrast to the linear response to Pi that was reported previously, the absorbance response at 443 or 623 nm was found to be sigmoidal using the recommended 2:1 Yb3+:PV solution (100 microM:50 microM, pH 7, HEPES). Furthermore, both continuous variation titration and molar ratio analysis (Job plot) experiments are consistent with 1:1, not 2:1, YbPV complex stoichiometry at pH 7 in HEPES buffer, indicating that the deviation from linearity is produced by excess Yb3+. Indeed, using a 1:1 Yb3+:PV ratio produces a linear response in DeltaAbs at 443 or 623 nm on back titration with analyte (phosphate or ATP). In addition, speciation analysis of the Yb-ATP system demonstrates that a 1:1 complex containing Yb3+ and ATP predominates in solution at microM metal ion and ATP concentrations. Paramagnetic 1H NMR spectroscopy directly establishes the formation of Yb3+-solute complexes in dilute aqueous solution. The 1:1 YbPV complex can be used for the colorimetric measurement of phosphate and ATP concentrations from approximately 2 microM.

Project description:Cysteine plays an essential role in maintaining cellular redox homeostasis and perturbations in cysteine concentration are associated with cardiovascular disease, liver disease, and cancer. 19F MRI is a promising modality for detecting cysteine in biology due to its high tissue penetration and negligible biological background signal. Herein we report fluorinated macrocyclic copper complexes that display a 19F NMR/MRI turn-on response following reduction of the Cu(ii) complexes by cysteine. The reactivity with cysteine was studied by monitoring the appearance of a robust diamagnetic 19F signal following addition of cysteine in conjunction with UV-vis and EPR spectroscopies. Importantly, complexes with -CH2CF3 tags display good water solubility. Studies with this complex in HeLa cells demonstrate the applicability of these probes to detect cysteine in complex biological environments.

Project description:The synthesis and characterization of a novel florescent chemosensor 1 with two different types of cationic binding sites have been reported in this work, which is a calix[4]crown derivative in 1,3-alternate conformation bearing two 2-phenyl-5-(4-dimethylaminopyenyl)-1,3,4-oxadiazole units. The recognition behaviors of 1 in dichloromethane/acetonitrile solution to alkali metal ions (Na+ and K+), alkaline earth metal ions (Mg2+ and Ca2+), and transition metal ions (Co2+, Ni2+, Zn2+, Cd2+, Cu2+, Mn2+, and Ag+) have been investigated by UV-Vis and fluorescence spectra. The fluorescence of 1 might be quenched selectively by Cu2+ due to the photo-induced electron transfer mechanism, and the quenched emission from 1 could be partly revived by the addition of Ca2+ or Mg2+; thus, the receptor 1 might be worked as an on-off switchable fluorescent chemosensor triggered by metal ion exchange.

Project description:An effective dual-DNAzyme-based unimolecular probe design employing intramolecular signal transduction is demonstrated. The probe is composed of three domains: a DNA-cleaving DNAzyme, a substrate, and an HRP-mimicking DNAzyme. When the probe meets its target, cleavage of the substrate by the DNA-cleaving DNAzyme activates the HRP-mimicking DNAzyme, producing a colorimetric signal. The Cu(2+)-dependent DNAzyme engineered to demonstrate this design revealed a sensitivity corresponding to 65 ppb, which is sufficient to detect Cu(2+) in drinking water. The new probe has excellent selectivity toward Cu(2+). This three-component design is simple and easy to engineer. It may provide the basis for future development of other nucleic acid-based probes for toxicological and environmental monitoring.

Project description:Quick and effective detection of biothiols in biological fluids has gained increasing attention due to its vital biological functions. In this paper, a novel reversible fluorescence chemosensor (L-Cu2+) based on a benzocoumarin-Cu2+ ensemble has been developed for the detection of biothiols (Cys, Hcy and GSH) in human urine. The chemosensing ensemble (L-Cu2+) contains a 2:1 stoichiometry structure between fluorescent ligand L and paramagnetic Cu2+. L was found to exclusively bond with Cu2+ ions accompanied with a dramatic fluorescence quenching maximum at 443 nm and an increase of an absorbance band centered at 378 nm. Then, the in situ generated fluorescence sluggish ensemble, L-Cu2+, was successfully used as a chemosensor for the detection of biothiols with a fluorescence "OFF-ON" response modality. Upon the addition of biothiols, the decomplexation of L-Cu2+ led to the liberation of the fluorescent ligand, L, resulting in the recovery of fluorescence and absorbance spectra. Studies revealed that L-Cu2+ possesses simple synthesis, excellent stability, high sensitivity, reliability at a broad pH range and desired renewability (at least 5 times). The practical application of L-Cu2+ was then demonstrated by the detection of biothiols in human urine sample.

Project description:The adjustment of the emitting wavelength of carbon dots (CDs) is usually realized by changing the raw materials, reaction temperature, or time. This paper reported the effective synthesis of multicolor photoluminescent CDs only by changing the solvent in a one-step solvothermal method, with 1,2,4,5-tetraaminobenzene as both the novel carbon source and nitrogen source. The emission wavelengths of the as-prepared CDs ranged from 527 to 605 nm, with quantum yields (QYs) reaching 10.0% to 47.6%, and it was successfully employed as fluorescence ink. The prepared red-emitting CDs (R-CDs, ?em = 605 nm) and yellow-emitting CDs (Y-CDs, ?em = 543 nm) were compared through multiple characterization methods, and their luminescence mechanism was studied. It was discovered that the large particle size, the existence of graphite Ns, and oxygen-containing functional groups are beneficial to the formation of long wavelength-emitting CDs. Y-CDs responded to crystal violet, and its fluorescence could be quenched. This phenomenon was thus employed to develop a detection method for crystal violet with a linear range from 0.1 to 11 µM and a detection limit of 20 nM.

Project description:Glyphosate, which has been widely reported to be a toxic pollutant, is often present at trace amounts in the environment. In this study, a novel copper-aluminum metal hydroxide doped graphene nanoprobe (labeled as CuAl-LDH/Gr NC) was first developed to construct a non-enzymatic electrochemical sensor for detection trace glyphosate. The characterization results showed that the synthesized CuAl-LDH had a high-crystallinity flowered structure, abundant metallic bands and an intercalated functional group. After mixed with Gr, the nanocomposites provided a larger surface area and better conductivity. The as-prepared CuAl-LDH/Gr NC dramatically improved the enrichment capability for glyphosate to realize the stripping voltammetry detection. The logarithmic linear detection range of the sensor was found to be 2.96 × 10-9-1.18 × 10-6 mol L-1 with the detection limit of 1 × 10-9 mol L-1 with excellent repeatability, good stability and anti-interference ability. Further, the sensor achieved satisfactory recovery rates in spiked surface water, ranging from 97.64% to 108.08%, demonstrating great accuracy and practicality.

Project description:The metal-binding capabilities of the spiropyran family of molecular switches have been explored for several purposes from sensing to optical circuits. Metal-selective sensing has been of great interest for applications ranging from environmental assays to industrial quality control, but sensitive metal detection for field-based assays has been elusive. In this work, we demonstrate colorimetric copper sensing at low micromolar levels. Dimethylamine-functionalized spiropyran (SP1) was synthesized and its metal-sensing properties were investigated using UV-vis spectrophotometry. The formation of a metal complex between SP1 and Cu2+ was associated with a color change that can be observed by the naked eye as low as ≈6 μM and the limit of detection was found to be 0.11 μM via UV-vis spectrometry. Colorimetric data showed linearity of response in a physiologically relevant range (0-20 μM Cu2+) with high selectivity for Cu2+ ions over biologically and environmentally relevant metals such as Na+, K+, Mn2+, Ca2+, Zn2+, Co2+, Mg2+, Ni2+, Fe3+, Cd2+, and Pb2+. Since the color change accompanying SP1-Cu2+ complex formation could be detected at low micromolar concentrations, SP1 could be viable for field testing of trace Cu2+ ions.
