Photographic High-Dynamic-Range Scalar Visualization.

ABSTRACT: We propose a photographic method to show scalar values of high dynamic range (HDR) by color mapping for 2D visualization. We combine (1) tone-mapping operators that transform the data to the display range of the monitor while preserving perceptually important features, based on a systematic evaluation, and (2) simulated glares that highlight high-value regions. Simulated glares are effective for highlighting small areas (of a few pixels) that may not be visible with conventional visualizations; through a controlled perception study, we confirm that glare is preattentive. The usefulness of our overall photographic HDR visualization is validated through the feedback of expert users.

DEVILS: a tool for the visualization of large datasets with a high dynamic range.

Project description:The number of grey values that can be displayed on monitors and be processed by the human eye is smaller than the dynamic range of image-based sensors. This makes the visualization of such data a challenge, especially with specimens where small dim structures are equally important as large bright ones, or whenever variations in intensity, such as non-homogeneous staining efficiencies or light depth penetration, becomes an issue. While simple intensity display mappings are easily possible, these fail to provide a one-shot observation that can display objects of varying intensities. In order to facilitate the visualization-based analysis of large volumetric datasets, we developed an easy-to-use ImageJ plugin enabling the compressed display of features within several magnitudes of intensities. The Display Enhancement for Visual Inspection of Large Stacks plugin (DEVILS) homogenizes the intensities by using a combination of local and global pixel operations to allow for high and low intensities to be visible simultaneously to the human eye. The plugin is based on a single, intuitively understandable parameter, features a preview mode, and uses parallelization to process multiple image planes. As output, the plugin is capable of producing a BigDataViewer-compatible dataset for fast visualization. We demonstrate the utility of the plugin for large volumetric image data.

High Dynamic Range Nanowire Resonators.

Project description:Dynamic range quantifies the linear operation regime available in nanomechanical resonators. Nonlinearities dominate the response of flexural beams in the limit of very high aspect ratio and very small diameter, which leads to expectation of low dynamic range for nanowire resonators in general. However, the highest achievable dynamic range for nanowire resonators with practical dimensions remains to be determined. We report dynamic range measurements on singly clamped silicon nanowire resonators reaching remarkably high values of up to 90 dB obtained with a simple harmonic actuation scheme. We explain these measurements by a comprehensive theoretical examination of dynamic range in singly clamped flexural beams including the effect of tapering, a usual feature of semiconductor nanowires. Our analysis reveals the nanowire characteristics required for broad linear operation, and given the relationship between dynamic range and mass sensing performance, it also enables analytical determination of mass detection limits, reaching atomic-scale resolution for feasible nanowires.

Precise Nanosizing with High Dynamic Range Holography.

Project description:Optical sensing is one of the key enablers of modern diagnostics. Especially label-free imaging modalities hold great promise as they eliminate labeling procedures prior to analysis. However, scattering signals of nanometric particles scale with their volume square. This unfavorable scaling makes it extremely difficult to quantitatively characterize intrinsically heterogeneous clinical samples, such as extracellular vesicles, as their signal variation easily exceeds the dynamic range of currently available cameras. Here, we introduce off-axis k-space holography that circumvents this limitation. By imaging the back-focal plane of our microscope, we project the scattering signal of all particles onto all camera pixels, thus dramatically boosting the achievable dynamic range to up to 110 dB. We validate our platform by detecting and quantitatively sizing metallic and dielectric particles over a 200 × 200 μm field of view and demonstrate that independently performed signal calibrations allow correctly sizing particles made from different materials. Finally, we present quantitative size distributions of extracellular vesicle samples.

Real-time high dynamic range laser scanning microscopy.

Project description:In conventional confocal/multiphoton fluorescence microscopy, images are typically acquired under ideal settings and after extensive optimization of parameters for a given structure or feature, often resulting in information loss from other image attributes. To overcome the problem of selective data display, we developed a new method that extends the imaging dynamic range in optical microscopy and improves the signal-to-noise ratio. Here we demonstrate how real-time and sequential high dynamic range microscopy facilitates automated three-dimensional neural segmentation. We address reconstruction and segmentation performance on samples with different size, anatomy and complexity. Finally, in vivo real-time high dynamic range imaging is also demonstrated, making the technique particularly relevant for longitudinal imaging in the presence of physiological motion and/or for quantification of in vivo fast tracer kinetics during functional imaging.

High dynamic range processing for magnetic resonance imaging.

Project description:PurposeTo minimize feature loss in T1- and T2-weighted MRI by merging multiple MR images acquired at different TR and TE to generate an image with increased dynamic range.Materials and methodsHigh Dynamic Range (HDR) processing techniques from the field of photography were applied to a series of acquired MR images. Specifically, a method to parameterize the algorithm for MRI data was developed and tested. T1- and T2-weighted images of a number of contrast agent phantoms and a live mouse were acquired with varying TR and TE parameters. The images were computationally merged to produce HDR-MR images. All acquisitions were performed on a 7.05 T Bruker PharmaScan with a multi-echo spin echo pulse sequence.ResultsHDR-MRI delineated bright and dark features that were either saturated or indistinguishable from background in standard T1- and T2-weighted MRI. The increased dynamic range preserved intensity gradation over a larger range of T1 and T2 in phantoms and revealed more anatomical features in vivo.ConclusionsWe have developed and tested a method to apply HDR processing to MR images. The increased dynamic range of HDR-MR images as compared to standard T1- and T2-weighted images minimizes feature loss caused by magnetization recovery or low SNR.

High fidelity sampling schedules for NMR spectra of high dynamic range.

Project description:The ability to reconstruct non-uniformly sampled (NUS) NMR spectra has mostly been accepted. Still a concern is lingering regarding artifacts from sampling non-uniformly. As experienced, some sampling schedules yield better results than others. Finding a useful schedule is relatively trivial for a low dynamic range spectrum and a conservative sparsity, but not so when the dynamic range is large and/or when extreme sparsity is used. High dynamic range is typically found in NOESY and spectra of metabolites, where quantification of peak heights is desired at high fidelity. Extreme sparsity is desired when high throughput is a goal. In all cases, selecting a poor sampling schedule can create unnecessary artifacts. Effectively, it is important to select a sampling schedule that provides a signal-to-artifact apex ratio (SAAR) value in par or better than the signal-to-noise ratio (SNR) value. Notably, by signal-to-artifact apex ratio we consider reconstruction fidelity as the apex intensity likeness, i.e., as the true signal to the tallest artifact. We show that the quality of reconstruction depends on the particular sampling schedule. We evaluate the reconstruction quality in the frequency domain following a matched Lorentz-to-Gauss transform plus common apodization and Fourier Transform. As the Lorentz-to-Gauss transform improves resolution and reduces ridges we include this when defining the Signal-to-Artifact Apex Ratio (SAAR) metric. This metric measures the ratio of simulated reconstructed peak height to the tallest artifact of reconstruction in a spectrum without noise. Once a NUS schedule is found with an optimal SAAR it will be satisfactory for all spectra recorded with the same parameter set. Tables with good seed values are provided in the supplement.

Ultra-high dynamic range quantum measurement retaining its sensitivity.

Project description:Quantum sensors are highly sensitive since they capitalise on fragile quantum properties such as coherence, while enabling ultra-high spatial resolution. For sensing, the crux is to minimise the measurement uncertainty in a chosen range within a given time. However, basic quantum sensing protocols cannot simultaneously achieve both a high sensitivity and a large range. Here, we demonstrate a non-adaptive algorithm for increasing this range, in principle without limit, for alternating-current field sensing, while being able to get arbitrarily close to the best possible sensitivity. Therefore, it outperforms the standard measurement concept in both sensitivity and range. Also, we explore this algorithm thoroughly by simulation, and discuss the T-2 scaling that this algorithm approaches in the coherent regime, as opposed to the T-1/2 of the standard measurement. The same algorithm can be applied to any modulo-limited sensor.

High dynamic range characterization of the trauma patient plasma proteome.

Project description:Although human plasma represents an attractive sample for disease biomarker discovery, the extreme complexity and large dynamic range in protein concentrations present significant challenges for characterization, candidate biomarker discovery, and validation. Herein we describe a strategy that combines immunoaffinity subtraction and subsequent chemical fractionation based on cysteinyl peptide and N-glycopeptide captures with two-dimensional LC-MS/MS to increase the dynamic range of analysis for plasma. Application of this "divide-and-conquer" strategy to trauma patient plasma significantly improved the overall dynamic range of detection and resulted in confident identification of 22,267 unique peptides from four different peptide populations (cysteinyl peptides, non-cysteinyl peptides, N-glycopeptides, and non-glycopeptides) that covered 3,654 different proteins with 1,494 proteins identified by multiple peptides. Numerous low abundance proteins were identified, exemplified by 78 "classic" cytokines and cytokine receptors and by 136 human cell differentiation molecules. Additionally a total of 2,910 different N-glycopeptides that correspond to 662 N-glycoproteins and 1,553 N-glycosylation sites were identified. A panel of the proteins identified in this study is known to be involved in inflammation and immune responses. This study established an extensive reference protein database for trauma patients that provides a foundation for future high throughput quantitative plasma proteomic studies designed to elucidate the mechanisms that underlie systemic inflammatory responses.

A high-sensitivity MEMS gravimeter with a large dynamic range.

Project description:Precise measurement of variations in the local gravitational acceleration is valuable for natural hazard forecasting, prospecting, and geophysical studies. Common issues of the present gravimetry technologies include their high cost, high mass, and large volume, which can potentially be solved by micro-electromechanical-system (MEMS) technology. However, the reported MEMS gravimeter does not have a high sensitivity and a large dynamic range comparable with those of the present commercial gravimeters, lowering its practicability and ruling out worldwide deployment. In this paper, we introduce a more practical MEMS gravimeter that has a higher sensitivity of 8 μGal/√Hz and a larger dynamic range of 8000 mGal by using an advanced suspension design and a customized optical displacement transducer. The proposed MEMS gravimeter has performed the co-site earth tides measurement with a commercial superconducting gravimeter GWR iGrav with the results showing a correlation coefficient of 0.91.

Event-Based Color Segmentation With a High Dynamic Range Sensor.

Project description:This paper introduces a color asynchronous neuromorphic event-based camera and a methodology to process color output from the device to perform color segmentation and tracking at the native temporal resolution of the sensor (down to one microsecond). Our color vision sensor prototype is a combination of three Asynchronous Time-based Image Sensors, sensitive to absolute color information. We devise a color processing algorithm leveraging this information. It is designed to be computationally cheap, thus showing how low level processing benefits from asynchronous acquisition and high temporal resolution data. The resulting color segmentation and tracking performance is assessed both with an indoor controlled scene and two outdoor uncontrolled scenes. The tracking's mean error to the ground truth for the objects of the outdoor scenes ranges from two to twenty pixels.
