Molecular Screening of Microorganisms Associated with Discolored Wood in Dead European Beech Trees Suffered from Extreme Drought Event Using Next Generation Sequencing.

ABSTRACT: Drought events weaken trees and make them vulnerable to attacks by diverse plant pathogens. Here, we propose a molecular method for fast screening of microorganisms associated with European beech decline after an extreme drought period (2018) in a forest of Thuringia, Germany. We used Illumina sequencing with a recent bioinformatics approach based on DADA2 to identify archaeal, bacterial, and fungal ASVs (amplicon sequence variants) based on bacterial and archaeal 16S and fungal ITS genes. We show that symptomatic beech trees are associated with both bacterial and fungal plant pathogens. Although the plant pathogen sequences were detected in both discolored and non-discolored wood areas, they were highly enriched in the discolored wood areas. We show that almost each individual tree was associated with a different combination of pathogens. Cytospora spp. and Neonectria coccinea were among the most frequently detected fungal pathogens, whereas Erwinia spp. and Pseudomonas spp. were the dominant bacterial plant pathogens. We demonstrate that bacterial plant pathogens may be of major importance in beech decline.

Project description:Trees adjust multiple structural and functional organ-specific characteristics, "traits", to cope with diverse soil conditions. Studies on traits are widely used to uncover ecological species adaptability to varying environments. However, fine-root traits are rarely studied for methodological reasons. We analyzed the adaptability of the fine-root systems of European beech and Norway spruce to extreme drought within species-specific tree groups at Kranzberger Forst (Germany), focusing on the seasonality of morphological, physiological, and biochemical key traits in view of carbon (C) and nitrogen dynamics. We hypothesized that fine roots of both species adjust to seasonal drought: with beech representing a "fast" (i.e. with fast C turnover), and spruce a "slow" (i.e. with long-term C retention) ecological strategy. We identified three functional fine-root categories, based on root function (absorptive or transport fine roots), and mycorrhizal status of the absorptive fine-roots (mycorrhizal or non-mycorrhizal). Solely the non-mycorrhizal absorptive roots adjusted in a species-specific manner supporting fine-root ecological strategy hypothesis. During drought, beech produced thin ephemeral (absorptive non-mycorrhizal) fine roots with high specific fine-root area and high respiratory activity, representing fast C turnover and enabling effective resource exploitation. These adjustments reflect a "fast" ecological strategy. Conversely, spruce absorptive fine roots did not respond to the soil moisture deficit by growth but instead increased root suberization. Drastically lowered respiratory activity of this functional category facilitated C retention and structural persistence during drought, indicating a "slow" ecological strategy in spruce. Absorptive mycorrhizal fine roots maintained respiration throughout the drought event in both tree species, but in spruce this was the only fine-root category with high respiration. This suggests, that spruce relies heavily on mycorrhizal associations as a method of drought resistance. Accumulation of non-structural carbohydrates and high C concentrations were observed in the transport fine roots of both species, indicating drought-induced osmotic protection of these roots. Thus, functional classification enabled us to determine that fine-root branches of each species are not tied to one sole ecological strategy. The suggested approach helps to better understand the complex interplay between structure and function belowground.

Project description:Climate projections propose that drought stress will become challenging for establishing trees. The magnitude of stress is dependent on tree species, provenance, and most likely also highly influenced by soil quality. European Beech (Fagus sylvatica) is of major ecological and economical importance in Central European forests. The species has an especially wide physiological and ecological amplitude enabling growth under various soil conditions within its distribution area in Central Europe. We studied the effects of extreme drought on beech saplings (second year) of four climatically distinct provenances growing on different soils (sandy loam and loamy sand) in a full factorial pot experiment. Foliar δ13C, δ15N, C, and N as well as above- and belowground growth parameters served as measures for stress level and plant growth. Low-quality soil enhanced the effect of drought compared with qualitatively better soil for the above- and belowground growth parameters, but foliar δ13C values revealed that plant stress was still remarkable in loamy soil. For beeches of one provenance, negative sandy soil effects were clearly smaller than for the others, whereas for another provenance drought effects in sandy soil were sometimes fatal. Foliar δ15N was correlated with plant size during the experiment. Plasticity of beech provenances in their reaction to drought versus control conditions varied clearly. Although a general trend of declining growth under control or drought conditions in sandy soil was found compared to loamy soil, the magnitude of the effect of soil quality was highly provenance specific. Provenances seemed to show adaptations not only to drought but also to soil quality. Accordingly, scientists should integrate information about climatic pre-adaptation and soil quality within the home range of populations for species distribution modeling and foresters should evaluate soil quality and climatic parameters when choosing donor populations for reforestation projects.

Project description:BackgroundThe incidence of Lyme borreliosis and other tick-borne diseases is increasing in Europe and North America. There is currently much interest in identifying the ecological factors that determine the density of infected ticks as this variable determines the risk of Lyme borreliosis to vertebrate hosts, including humans. Lyme borreliosis is caused by the bacterium Borrelia burgdorferi sensu lato (s.l.) and in western Europe, the hard tick Ixodes ricinus is the most important vector.MethodsOver a 15-year period (2004-2018), we monitored the monthly abundance of I. ricinus ticks (nymphs and adults) and their B. burgdorferi s.l. infection status at four different elevations on a mountain in western Switzerland. We collected climate variables in the field and from nearby weather stations. We obtained data on beech tree seed production (masting) from the literature, as the abundance of Ixodes nymphs can increase dramatically 2 years after a masting event. We used generalized linear mixed effects models and AIC-based model selection to identify the ecological factors that influence inter-annual variation in the nymphal infection prevalence (NIP) and the density of infected nymphs (DIN).ResultsWe found that the NIP decreased by 78% over the study period. Inter-annual variation in the NIP was explained by the mean precipitation in the present year, and the duration that the DNA extraction was stored in the freezer prior to pathogen detection. The DIN decreased over the study period at all four elevation sites, and the decrease was significant at the top elevation. Inter-annual variation in the DIN was best explained by elevation site, year, beech tree masting index 2 years prior and the mean relative humidity in the present year. This is the first study in Europe to demonstrate that seed production by deciduous trees influences the density of nymphs infected with B. burgdorferi s.l. and hence the risk of Lyme borreliosis.ConclusionsPublic health officials in Europe should be aware that masting by deciduous trees is an important predictor of the risk of Lyme borreliosis.
