Long-Term Nitrogen Deposition Alters Ectomycorrhizal Community Composition and Function in a Poplar Plantation.

ABSTRACT: The continuous upsurge in soil nitrogen (N) enrichment has had strong impacts on the structure and function of ecosystems. Elucidating how plant ectomycorrhizal fungi (EMF) mutualists respond to this additional N will facilitate the rapid development and implementation of more broadly applicable management and remediation strategies. For this study, we investigated the responses of EMF communities to increased N, and how other abiotic environmental factors impacted them. Consequently, we conducted an eight-year N addition experiment in a poplar plantation in coastal eastern China that included five N addition levels: 0 (N0), 50 (N1), 100 (N2), 150 (N3), and 300 (N4) kg N ha-1 yr-1. We observed that excessive N inputs reduced the colonization rate and species richness of EMF, and altered its community structure and functional traits. The total carbon content of the humus layer and available phosphorus in the mineral soil were important drivers of EMF abundance, while the content of ammonium in the humus layer and mineral soil determined the variations in the EMF community structure and mycelium foraging type. Our findings indicated that long-term N addition induced soil nutrient imbalances that resulted in a severe decline in EMF abundance and loss of functional diversity in poplar plantations.

Project description:Cunninghamia lanceolata (Lamb.) Hook. has been widely planted in subtropical China to meet increasing timber demands, leading to short-rotation practices that deplete soil nutrients. However, increased nitrogen (N) deposition offsets soil N depletion. While long-term experimental data investigating the coupled effects related to short rotation practices and increasing N deposition are scarce, applying model simulations may yield insights. In this study, the CenW3.1 model was validated and parameterized using data from pure C. lanceolata plantations. The model was then used to simulate various changes in long-term productivity. Results indicated that responses of productivity of C. lanceolata plantation to increased N deposition were more related to stand age than N addition, depending on the proportion and age of growing forests. Our results have also shown a rapid peak in growth and N dynamics. The peak is reached sooner and is higher under higher level of N deposition. Short rotation lengths had a greater effect on productivity and N dynamics than high N deposition levels. Productivity and N dynamics decreased as the rotation length decreased. Total productivity levels suggest that a 30-year rotation length maximizes productivity at the 4.9 kg N ha(-1) year(-1) deposition level. For a specific rotation length, higher N deposition levels resulted in greater overall ecosystem C and N storage, but this positive correlation tendency gradually slowed down with increasing N deposition levels. More pronounced differences in N deposition levels occurred as rotation length decreased. To sustain C. lanceolata plantation productivity without offsite detrimental N effects, the appropriate rotation length is about 20-30 years for N deposition levels below 50 kg N ha(-1) year(-1) and about 15-20 years for N deposition levels above 50 kg N ha(-1) year(-1). These results highlight the importance of assessing N effects on carbon management and the long-term productivity of forest ecosystems.

Project description:Soil microorganisms are key regulators for plant growth and ecosystem health of forest ecosystem. Although previous research has demonstrated that soil microorganisms are greatly affected by understory nitrogen (N) addition, little is known about the effects of canopy N addition (CNA) and understory management on soil microorganisms in forests. In this study, we conducted a full designed field experiment with four treatments: CNA (25 kg N ha-1 year-1), understory removal (UR), canopy N addition, and understory removal (CNAUR) (25 kg N ha-1 year-1), and control in a Chinese fir plantation. High-throughput sequencing and qPCR techniques were used to determine the abundance, diversity, and composition of bacterial and fungal communities in three soil layers. Our results showed that CNA increased bacterial diversity in the 10-20 cm soil layer but decreased bacterial abundance in the 20-40 cm soil layer and fungal diversity in the 0-10 cm soil layer. UR increased bacterial abundance only in the 20-40 cm soil layer. CNA, not UR significantly altered the compositions of soil bacterial and fungal community compositions, especially in the 0-20 cm soil layer. CNA sharply reduced the relative abundance of copiotrophic taxa (i.e., taxa in the bacterial phylum Proteobacteria and the orders Eurotiales and Helotiales in the fungal phylum Ascomycota) but increased the relative abundance of oligotrophic taxa (i.e., in the bacterial phylum Verrucomicrobia). RDA analysis revealed that soil pH, DON, and DOC were the main factors associated with the variation in bacterial and fungal communities. Our findings suggest that short-term CNA changes both soil bacterial and fungal communities, with stronger responses in the surface and middle soil than in the deep soil layer, and that UR may enhance this effect on the soil bacterial abundance. This study improves our understanding of soil microorganisms in plantations managed with understory removal and that experience increases in N deposition.

Project description:Microbial diversity is an important indicator of soil fertility and plays an indispensable role in farmland ecosystem sustainability. The short-term effects of fertilization and rhizobium inoculation on soil microbial diversity and community structure have been explored extensively; however, few studies have evaluated their long-term effects. Here, we applied quantitative polymerase chain reaction (qPCR) and amplicon sequencing to characterize the effect of 10-year fertilizer and rhizobium inoculation on bacterial communities in soybean bulk and rhizosphere soils at the flowering-podding and maturity stages. Four treatments were examined: non-fertilization control (CK), phosphorus and potassium fertilization (PK), nitrogen and PK fertilization (PK + N), and PK fertilization and Bradyrhizobium japonicum 5821 (PK + R). Long-term co-application of rhizobium and PK promoted soybean nodule dry weight by 33.94% compared with PK + N, and increased soybean yield by average of 32.25%, 5.90%, and 5.00% compared with CK, PK, and PK + N, respectively. The pH of PK + R was significantly higher than that of PK and PK + N at the flowering-podding stage. The bacterial abundance at the flowering-podding stage was positively correlated with soybean yield, but not at the maturity stage. The significant different class Gemmatimonadetes, and the genera Gemmatimonas, and Ellin6067 in soil at the flowering-podding stage were negatively correlated with soybean yield. However, the bacterial community at class and genus levels at maturity had no significant effect on soybean yield. The key bacterial communities that determine soybean yield were concentrated in the flowering-podding stage, not at maturity stage. Rhizosphere effect, growth period, and treatment synergies resulted in significant differences in soil bacterial community composition. Soil organic matter (OM), total nitrogen (TN), pH, and available phosphorus (AP) were the main variables affecting bacterial community structure. Overall, long-term co-application of rhizobium and fertilizer not only increased soybean yield, but also altered soil bacterial community structure through niche reconstruction and microbial interaction. Rhizobium inoculation plays key role in reducing nitrogen fertilizer application and promoting sustainable agriculture practices.

Project description:Soil extracellular enzymes play a significant role in the N mineralization process. However, few studies have documented the linkage between enzyme activity and the microbial community that performs the function. This study examined the effects of inorganic and organic N fertilization on soil microbial communities and their N mineralization functions over 4 years. Soils were collected from silage corn field plots with four contrasting N treatments: control (no additional N), ammonium sulfate (AS; 100 and 200 kg of N ha-1), and compost (200 kg of N ha-1). Illumina amplicon sequencing was used to comprehensively assess the overall bacterial community (16S rRNA genes), bacterial ureolytic community (ureC), and bacterial chitinolytic community (chiA). Selected genes involved in N mineralization were also examined using quantitative real-time PCR and metagenomics. Enzymes (and marker genes) included protease (npr and sub), chitinase (chiA), urease (ureC), and arginase (rocF). Compost significantly increased diversity of overall bacterial communities even after one application, while ammonium fertilizers had no influence on the overall bacterial communities over four seasons. Bacterial ureolytic and chitinolytic communities were significantly changed by N fertilization. Compost treatment strongly elevated soil enzyme activities after 4 years of repeated application. Functional gene abundances were not significantly affected by N treatments, and they were not correlated with corresponding enzyme activities. N mineralization genes were recovered from soil metagenomes based on a gene-targeted assembly. Understanding how the structure and function of soil microbial communities involved with N mineralization change in response to fertilization practices may indicate suitable agricultural management practices that improve ecosystem services while reducing negative environmental consequences.IMPORTANCE Agricultural N management practices influence the enzymatic activities involved in N mineralization. However, specific enzyme activities do not identify the microbial species directly involved in the measured process, leaving the link between the composition of the microbial community and the production of key enzymes poorly understood. In this study, the application of high-throughput sequencing, real-time PCR, and metagenomics shed light on how the abundance and diversity of microorganisms involved in N mineralization respond to N management. We suggest that N fertilization has significantly changed bacterial ureolytic and chitinolytic communities.
