Project description:Here, we report data of the principal component analysis (PCA) assessment and clustering analysis related to low-temperature thermal hydrolysis process (THP) for enhancing the anaerobic digestion (AD) of sludge in wastewater treatment plants (WWTPs) with primary sludge fermentation (Azizi et al., 2021). The PCA was examined to pinpoint the influence of different THP schemes on the variations of macromolecular compounds solubilization after low-temperature THP and the relative performances in enhancing methane potential in AD. We established 2 experimental setups with a total of 18 treatment conditions (3 exposure times, 30, 60, and 90 min at three temperature levels 50, 70 and 90 °C) in comparison to the untreated control samples. Scheme-1 comprises the THP of a mixture of (1:1 vol ratio) fermented primary sludge (FPS) and thickened waste activated sludge (TWAS); while scheme-2 comprised the THP of TWAS only. The factors employed in the assessment of the PCA encompassed the variations in the macromolecular compounds and other solubilization metrics. This included the variations in the levels of carbohydrates, lipids, proteins, and solubilization of chemical oxygen demand (COD) and volatile suspended solids (VSS). Furthermore, the evaluation considered the changes of volatile fatty acids (VFAs) and total ammonia nitrogen (TAN) with respect to time and temperature. The assessment of PCA classified the THP based on their differences and alterations that occurred after the treatment. The indices of the PCA assessments differed based on the factors of concern and the focus of each individual PCA assessment. In every individual PCA assessment, the respective contribution to the total variance in PCA analysis was calculated and manifested by the highest distribution of the principal components (PCs) axis PC1 and PC2. The differences in distributions of PCs after various PCA examinations can describe the relative influence of THP schemes and the most significant variables that can trigger major differences among THP conditions. The comparative differences demonstrated by PCA support the potential investigations of the efficiency of THPs conditions and their performance categories.

Project description:Pretreatment of waste-activated sludge (WAS) is an effective way to destabilize sludge floc structure and release organic matter for improving sludge digestion efficiency. Nonetheless, information on the impact of WAS pretreatment on digestion sludge microbiomes, as well as mechanistic insights into how sludge pretreatment improves digestion performance, remains elusive. In this study, a genome-centric metagenomic approach was employed to investigate the digestion sludge microbiome in four sludge digesters with different types of feeding sludge: WAS pretreated with 0.25 mol/liter alkaline/acid (APAD), WAS pretreated with 0.8 mol/liter alkaline/acid (HS-APAD), thermally pretreated WAS (thermal-AD), and fresh WAS (control-AD). We retrieved 254 metagenome-assembled genomes (MAGs) to identify the key functional populations involved in the methanogenic digestion process. These MAGs span 28 phyla, including 69 yet-to-be-cultivated lineages, and 30 novel lineages were characterized with metabolic potential associated with hydrolysis and fermentation. Interestingly, functional populations involving carbohydrate digestion were enriched in APAD and HS-APAD, while lineages related to protein and lipid fermentation were enriched in thermal-AD, corroborating the idea that different substrates are released from alkaline/acid and thermal pretreatments. Among the major functional populations (i.e., fermenters, syntrophic acetogens, and methanogens), significant correlations between genome sizes and abundance of the fermenters were observed, particularly in APAD and HS-APAD, which had improved digestion performance.IMPORTANCE Wastewater treatment generates large amounts of waste-activated sludge (WAS), which consists mainly of recalcitrant microbial cells and particulate organic matter. Though WAS pretreatment is an effective way to release sludge organic matter for subsequent digestion, detailed information on the impact of the sludge pretreatment on the digestion sludge microbiome remains scarce. Our study provides unprecedented genome-centric metagenomic insights into how WAS pretreatments change the digestion sludge microbiomes, as well as their metabolic networks. Moreover, digestion sludge microbiomes could be a unique source for exploring microbial dark matter. These results may inform future optimization of methanogenic sludge digestion and resource recovery.

Project description:Pretreatments have been successfully used to shorten the HRT of anaerobic digestion (AD) of sewage sludge, but they become ineffective for low-organic sewage sludge, with HRT > 10 days. Herein, a new process using alkaline hydrolysis-anaerobic digestion has been developed to solve this problem. Firstly, maximum organic matter in the sludge was dissolved by strong alkaline hydrolysis (pH > 11) in a two-stage alkaline hydrolysis system (TSAHS). Secondly, only the supernatant of the sludge that contained most of the methane potential was applied for AD. The operational conditions were optimized and the process mechanism was also analyzed. The results showed that under optimum operational conditions, above 19% of the organic matter in the sludge was released into the supernatant after alkaline hydrolysis in TSAHS, and the supernatant for AD achieved a methane production of 392 mL CH4 per g COD. The process attained a methane production of 0.26 m3 CH4 per kg VS and a VS reduction of 43.5%, while the HRT was only 12 h. The advantage of the mechanism was that the alkaline neutralization capacity of the sludge maintained a proper pH value for the supernatant from TSAHS, which benefited subsequent AD. It is concluded that the new process based on the dissolution of organic matter can attain a short digestion time for low-organic sludge.

Project description:Three-dimensional printed drug development is nowadays an active area in the pharmaceutical industry, where the search for an appropriate edible carrier that permits the thermal processing of the mixture at temperature levels that are safe for the drug is an important field of study. Here, potato starch and hydroxypropyl cellulose based mixtures loaded with paracetamol up to 50% in weight were processed by hot melt extrusion at 85 °C to test their suitability to be thermally processed. The extruded mixtures were tested by liquid chromatography to analyze their release curves and were thermally characterized. The drug recovery was observed to be highly dependent on the initial moisture level of the mixture, the samples being prepared with an addition of water at a ratio of 3% in weight proportional to the starch amount, highly soluble and easy to extrude. The release curves showed a slow and steady drug liberation compared to a commercially available paracetamol tablet, reaching the 100% of recovery at 60 min. The samples aged for 6 weeks showed slower drug release curves compared to fresh samples, this effect being attributable to the loss of moisture. The paracetamol loaded mixture in powder form was used to print pills with different sizes and geometries in a fused deposition modelling three-dimensional printer modified with a commercially available powder extrusion head, showing the potential of this formulation for use in personalized medicine.

Project description:Non-hydrolyzed polyacrylamide (PAM) and partially hydrolyzed polyacrylamide (HPAM) are commonly used polymers in various industrial applications, including in oil and gas production operations. Understanding the microbial utilization of such polymers can contribute to improved recovery processes and help to develop technologies for polymer remediation. Microbial communities enriched from oilfield produced water (PW) and activated sludge from Alberta, Canada were assessed for their ability to utilize PAM and HPAM as nitrogen and carbon sources at 50 °C. Microbial growth was determined by measuring CO2 production, and viscosity changes and amide concentrations were used to determine microbial utilization of the polymers. The highest CO2 production was observed in incubations wherein HPAM was added as a nitrogen source for sludge-derived enrichments. Our results showed that partial deamination of PAM and HPAM occurred in both PW and sludge microbial cultures after 34 days of incubation. Whereas viscosity changes were not observed in cultures when HPAM or PAM was provided as the only carbon source, sludge enrichment cultures amended with HPAM and glucose showed significant decreases in viscosity. 16S rRNA gene sequencing analysis indicated that microbial members from the family Xanthomonadaceae were enriched in both PW and sludge cultures amended with HPAM or PAM as a nitrogen source, suggesting the importance of this microbial taxon in the bio-utilization of these polymers. Overall, our results demonstrate that PAM and HPAM can serve as nitrogen sources for microbial communities under the thermophilic conditions commonly found in environments such as oil and gas reservoirs.

Project description:We describe here for the first time the low temperature superplasticity of nanostructured low carbon steel (microalloyed with V, N, Mn, Al, Si, and Ni). Low carbon nanograined/ultrafine-grained (NG/UFG) bulk steel was processed using a combination of cold-rolling and annealing of martensite. The complex microstructure of NG/UFG ferrite and 50-80 nm cementite exhibited high thermal stability at 500 °C with low temperature elongation exceeding 100% (at less than 0.5 of the absolute melting point) as compared to the conventional fine-grained (FG) counterpart. The low temperature superplasticity is adequate to form complex components. Moreover, the low strength during hot processing is favorable for decreasing the spring back and minimize die loss.

Project description:The aim of this study was to estimate bentazone's potential to leach to groundwater in the Arenosols developed from sand, Luvisols developed from loamy sand or sandy loam, and Luvisols or Cambisols developed from loess, and to identify the major factors influencing bentazone's fate in the soils. Potato and maize cultivations were simulated using the FOCUS PELMO 5.5.3 pesticide leaching model. The amount of bentazone reaching groundwater was highly sensitive to degradation parameters, water-holding capacity, evapotranspiration, organic carbon content, and pH. The highest bentazone concentrations in percolate were noted in Arenosols. The risk of bentazone concentration exceeding 0.1 μg/L was low only in Arenosols with high organic carbon content (3.0% for topsoil or higher). In Luvisols developed from loamy sand or sandy loam, the estimated bentazone concentrations in percolate were highly dependent on the climate. In Luvisols or Cambisols developed from loess, concentrations of >0.1 μg/L were the least likely due to the high water-holding capacity and high organic carbon content of these soils. The study also revealed that the FOCUS Hamburg scenario, representing the coarsest soils in the European Union with relatively low organic carbon content, does not reflect the leaching potential of Arenosols and Luvisols.

Project description:The potential of periodate (PI) in sludge anaerobic digestion is not tapped, although it has recently attracted great research interest in organic contaminants removal and pathogens inactivation in wastewater treatment. This is the first work to demonstrate significant improvement in methane generation from waste activated sludge (WAS) with PI pretreatment and to provide underlying mechanisms. Biochemical methane potential tests indicated that methane yield enhanced from 100.2 to 146.3 L per kg VS (VS, volatile solids) with PI dosages from 0 to 100 mg per g TS (TS, total solids). Electron spin resonance showed PI could be activated without extra activator addition, which might be attributed to the native transition metals (e.g., Fe2+) in WAS, thereby generating hydroxyl radical (•OH), superoxide radicals (•O2 -), and singlet oxygen (1O2). Further scavenging tests demonstrated all of them synergistically promoted WAS disintegration, and their contributions were in the order of •O2 - > •OH > 1O2, leading to the release of substantial biodegradable substances (i.e., proteins and polysaccharides) into the liquid phase for subsequent biotransformation. Moreover, fluorescence and ultraviolet spectroscopy analyses indicated the recalcitrant organics (especially lignocellulose and humus) could be degraded by reducing their aromaticity under oxidative stress of PI, thus readily for methanogenesis. Microbial community analysis revealed some microorganisms participating in hydrolysis, acidogenesis, and acetoclastic methanogenesis were enriched after PI pretreatment. The improved key enzyme activities and up-regulated metabolic pathways further provided direct evidence for enhanced methane production. This research was expected to broaden the application scope of PI and provide more diverse pretreatment choices for energy recovery through anaerobic digestion.

Project description:In this work, we describe the role of the different layers in perovskite solar cells to achieve reproducible, ~16% efficient perovskite solar cells. We used a planar device architecture withPedotPSS on the bottom, followed by the perovskite layer and an evaporated C60 layer before deposition of the top electrode. No high temperature annealing step is needed, which also allows processing on flexible plastic substrates. Only the optimization of all of these layers leads to highly efficient and reproducible results. In this work, we describe the effects of different processing conditions, especially the influence of the C60 top layer on the device performance.
