S100B dysregulation during brain development affects synaptic SHANK protein networks via alteration of zinc homeostasis.

ABSTRACT: Autism Spectrum Disorders (ASD) are caused by a combination of genetic predisposition and nongenetic factors. Among the nongenetic factors, maternal immune system activation and zinc deficiency have been proposed. Intriguingly, as a genetic factor, copy-number variations in S100B, a pro-inflammatory damage-associated molecular pattern (DAMP), have been associated with ASD, and increased serum S100B has been found in ASD. Interestingly, it has been shown that increased S100B levels affect zinc homeostasis in vitro. Thus, here, we investigated the influence of increased S100B levels in vitro and in vivo during pregnancy in mice regarding zinc availability, the zinc-sensitive SHANK protein networks associated with ASD, and behavioral outcomes. We observed that S100B affects the synaptic SHANK2 and SHANK3 levels in a zinc-dependent manner, especially early in neuronal development. Animals exposed to high S100B levels in utero similarly show reduced levels of free zinc and SHANK2 in the brain. On the behavioral level, these mice display hyperactivity, increased stereotypic and abnormal social behaviors, and cognitive impairment. Pro-inflammatory factors and zinc-signaling alterations converge on the synaptic level revealing a common pathomechanism that may mechanistically explain a large share of ASD cases.

Project description:The communication of neurons is primarily maintained by synapses, which play a crucial role in the functioning of the nervous system. Therefore, synaptic failure may critically impair information processing in the brain and may underlie many neurodegenerative diseases. A number of studies have suggested that synaptic failure may preferentially target neurons with high connectivity (i.e., network hubs). As a result, the activity of these highly connected neurons can be significantly affected. It has been speculated that anesthetics regulate conscious state by affecting synaptic transmission at these network hubs and subsequently reducing overall coherence in the network activity. In addition, hubs in cortical networks are shown to be more vulnerable to amyloid deposition because of their higher activity within the network, causing decrease in coherence patterns and eventually Alzheimer's disease (AD). Here, we investigate how synaptic failure can affect spatio-temporal dynamics of scale free networks, having a power law scaling of number of connections per neuron - a relatively few neurons (hubs) with a lot of emanating or incoming connections and many cells with low connectivity. We studied two types of synaptic failure: activity-independent and targeted, activity-dependent synaptic failure. We defined scale-free network structures based on the dominating direction of the connections at the hub neurons: incoming and outgoing. We found that the two structures have significantly different dynamical properties. We show that synaptic failure may not only lead to the loss of coherence but unintuitively also can facilitate its emergence. We show that this is because activity-dependent synaptic failure homogenizes the activity levels in the network creating a dynamical substrate for the observed coherence increase. Obtained results may lead to better understanding of changes in large-scale pattern formation during progression of neuro-degenerative diseases targeting synaptic transmission.

Project description:IntroductionThere is a close and complex interaction between the elements in the aboveground-underground ecosystem during the growth and development of plants. Specifically, when the aboveground part of plants is infected by pathogens, it induces the plant rhizosphere to synthesize specific root exudates. Consequently, a group of beneficial rhizosphere soil bacteria is recruited to help plants resist diseases. However, the changes in the rhizosphere soil bacterial community of plants under infection by oomycete pathogens remain unknown.MethodsThree experimental treatments were set up in this experiment: soils inoculated with P. nicotianae, no-inoculation with P. nicotianae, and a control. The control treatment was composed of soils without transplanted tobacco plants, with the pathogen inoculated twice at an interval of eight days to ensure a successful P. nicotianae infection. P. nicotianae inoculation treatments were designed using the hyphal block inoculation method. In the non-inoculation treatment, tobacco plants were grown normally without pathogen inoculation. The tobacco plants were grown in a greenhouse.ResultsThis study demonstrates that tobacco plants recruit microorganisms at the rhizosphere level as a defense mechanism against disease after infection by the oomycete pathogen Phytophthora nicotianae. Specific rhizosphere soil bacteria were screened in vitro to promote tobacco growth in a biofilm-forming manner, which induced the systemic resistance of the plants to P. nicotianae. The recruitment of rhizosphere soil bacteria to the inter-root zone of tobacco plants after infection by P. nicotianae can help subsequently cultivated tobacco plants in the same soil resist pathogen infestation.DiscussionIn conclusion, the present study confirms that infestation caused by oomycete pathogens alters the composition of the plant rhizosphere soil bacterial community and recruits a specific group of beneficial microorganisms that induce disease resistance and promote plant growth, thereby maximizing the protection of progeny grown in the same soil against the disease.
