Stain-free identification of tissue pathology using a generative adversarial network to infer nanomechanical signatures.

ABSTRACT: Intraoperative frozen section analysis can be used to improve the accuracy of tumour margin estimation during cancer resection surgery through rapid processing and pathological assessment of excised tissue. Its applicability is limited in some cases due to the additional risks associated with prolonged surgery, largely from the time-consuming staining procedure. Our work uses a measurable property of bulk tissue to bypass the staining process: as tumour cells proliferate, they influence the surrounding extra-cellular matrix, and the resulting change in elastic modulus provides a signature of the underlying pathology. In this work we accurately localise atomic force microscopy measurements of human liver tissue samples and train a generative adversarial network to infer elastic modulus from low-resolution images of unstained tissue sections. Pathology is predicted through unsupervised clustering of parameters characterizing the distributions of inferred values, achieving 89% accuracy for all samples based on the nominal assessment (n = 28), and 95% for samples that have been validated by two independent pathologists through post hoc staining (n = 20). Our results demonstrate that this technique could increase the feasibility of intraoperative frozen section analysis for use during resection surgery and improve patient outcomes.

Project description:Drug design is an important area of study for pharmaceutical businesses. However, low efficacy, off-target delivery, time consumption, and high cost are challenges and can create barriers that impact this process. Deep Learning models are emerging as a promising solution to perform de novo drug design, i.e., to generate drug-like molecules tailored to specific needs. However, stereochemistry was not explicitly considered in the generated molecules, which is inevitable in targeted-oriented molecules. This paper proposes a framework based on Feedback Generative Adversarial Network (GAN) that includes optimization strategy by incorporating Encoder-Decoder, GAN, and Predictor deep models interconnected with a feedback loop. The Encoder-Decoder converts the string notations of molecules into latent space vectors, effectively creating a new type of molecular representation. At the same time, the GAN can learn and replicate the training data distribution and, therefore, generate new compounds. The feedback loop is designed to incorporate and evaluate the generated molecules according to the multiobjective desired property at every epoch of training to ensure a steady shift of the generated distribution towards the space of the targeted properties. Moreover, to develop a more precise set of molecules, we also incorporate a multiobjective optimization selection technique based on a non-dominated sorting genetic algorithm. The results demonstrate that the proposed framework can generate realistic, novel molecules that span the chemical space. The proposed Encoder-Decoder model correctly reconstructs 99% of the datasets, including stereochemical information. The model's ability to find uncharted regions of the chemical space was successfully shown by optimizing the unbiased GAN to generate molecules with a high binding affinity to the Kappa Opioid and Adenosine [Formula: see text] receptor. Furthermore, the generated compounds exhibit high internal and external diversity levels 0.88 and 0.94, respectively, and uniqueness.

Project description:Background Animal illness is a disturbance in an animal’s natural condition that disrupts or changes critical functions. Concern over animal illnesses stretches back to the earliest human interactions with animals and is mirrored in early religious and magical beliefs. Animals have long been recognized as disease carriers. Man has most likely been bitten, stung, kicked, and gored by animals for as long as he has been alive; also, early man fell ill or died after consuming the flesh of deceased animals. Man has recently learned that numerous invertebrates are capable of transferring disease-causing pathogens from man to man or from other vertebrates to man. These animals, which function as hosts, agents, and carriers of disease, play a significant role in the transmission and perpetuation of human sickness. Thus, there is a need to detect unhealthy animals from a whole group of animals. Methods In this study, a deep learning-based method is used to detect or separate out healthy-unhealthy animals. As the dataset contains a smaller number of images, an image augmentation-based method is used prior to feed the data in the deep learning network. Flipping, scale-up, sale-down and orientation is applied in the combination of one to four to increase the number of images as well as to make the system robust from these variations. One fuzzy-based brightness correction method is proposed to correct the brightness of the image. Lastly, semi-supervised generative adversarial network (SGAN) is used to detect the healthy-unhealthy animal images. As per our knowledge, this is the first article which is prepared to detect healthy-unhealthy animal images. Results The outcome of the method is tested on augmented COCO dataset and achieved 91% accuracy which is showing the efficacy of the method. Conclusions A novel two-fold animal healthy-unhealthy detection system is proposed in this study. The result gives 91.4% accuracy of the model and detects the health of the animals in the pictures accurately. Thus, the system improved the literature on healthy-unhealthy animal detection techniques. The proposed approach may effortlessly be utilized in many computer vision systems that could be confused by the existence of a healthy-unhealthy animal.
