Draft Genome Sequence of a Poly-γ-Glutamic Acid-Producing Isolate, Bacillus paralicheniformis Strain bcasdu2018/01.

ABSTRACT: Bacillus paralicheniformis bcasdu2018/01 was isolated from the indoor environment of a chemistry laboratory. As part of the extracellular matrix, this isolate produces copious amounts of poly-γ-glutamic acid (γ-PGA). Here, we report the 4.25-Mbp draft genome assembly of the organism with an average G+C content of 45.92%.

This sequence will provide further help for the biosynthesis of γ-PGA and will greatly facilitate research efforts in metabolic engineering of B. subtilis subsp. natto strain CGMCC 2108.

Improvement of Bacillus subtilis for poly-γ-glutamic acid production by genome shuffling.

Project description:Poly-γ-glutamic acid (γ-PGA) is a promising microbial polymer with potential applications in industry, agriculture and medicine. The use of high γ-PGA-producing strains is an effective approach to improve productivity of γ-PGA. In this study, we developed a mutant, F3-178, from Bacillus subtilis GXA-28 using genome shuffling. The morphological characteristics of F3-178 and GXA-28 were not identical. Compared with GXA-28 (18.4 ± 0.8 g l-1 ), the yield of γ-PGA was 1.9-fold higher in F3-178 (34.3 ± 1.2 g l-1 ). Results from batch fermentation in 3.7 l fermenter showed that F3-178 was satisfactory for industrial production of γ-PGA. Metabolic studies suggested that the higher γ-PGA yield in F3-178 could be attributed to increased intracellular flux and uptake of extracellular glutamate. Real-time PCR indicated that mRNA level of pgsB in F3-178 was 18.8-fold higher than in GXA-28, suggesting the higher yield might be related to the overexpression of genes involved in γ-PGA production. This study demonstrated that genome shuffling can be used for rapid improvement of γ-PGA strains, and the possible mechanism for the improved phenotype was also explored at the metabolic and transcriptional levels.

Poly-L-gamma-glutamic acid production by recombinant Bacillus subtilis without pgsA gene.

Project description:Poly-gamma-glutamic acid (PGA) is a promising bio-based polymer that shares many functions with poly (acrylic acid) and its derivatives. Thus, technologies for efficient production and molecular size control of PGA are required to expand the application of this useful biopolymer. In Bacillus strains, PGA is synthesized by the PgsBCA protein complex, which is encoded by the pgsBCA gene operon, otherwise is known as ywsC and ywtAB operons and/or capBCA operon. Hence, we investigated responsible components of the PgsBCA complex in B. subtilis for over-production of PGA. In particular, we constructed genomic pgsBCA gene-deletion mutants of B. subtilis. And also, we assembled high copy-number plasmids harboring σA-dependent promoter, leading to high-level expression of all combinations of pgsBCA, pgsBC, pgsBA, pgsCA, pgsB, pgsC, and/or pgsA genes. Subsequently, PGA production of the transformed B. subtilis mutant was determined in batch fermentation using medium supplemented with L-glutamate. PGA production by the transformants introduced with pgsBC genes (lacking the genomic pgsBCA genes) was 26.0 ± 3.0 g L-1, and the enantiomeric ratio of D- and L-glutamic acid (D/L-ratio) in the produced PGA was 5/95. In contrast, D/L-ratio of produced PGA by the transformants introduced with pgsBCA genes (control strains) was 75/25. In conclusion, B. subtilis without pgsA gene could over-produce PGA with an L-rich enantiomeric ratio.

Metabolic engineering of Bacillus amyloliquefaciens LL3 for enhanced poly-γ-glutamic acid synthesis.

Project description:Poly-γ-glutamic acid (γ-PGA) is a biocompatible and biodegradable polypeptide with wide-ranging applications in foods, cosmetics, medicine, agriculture and wastewater treatment. Bacillus amyloliquefaciens LL3 can produce γ-PGA from sucrose that can be obtained easily from sugarcane and sugar beet. In our previous work, it was found that low intracellular glutamate concentration was the limiting factor for γ-PGA production by LL3. In this study, the γ-PGA synthesis by strain LL3 was enhanced by chromosomally engineering its glutamate metabolism-relevant networks. First, the downstream metabolic pathways were partly blocked by deleting fadR, lysC, aspB, pckA, proAB, rocG and gudB. The resulting strain NK-A6 synthesized 4.84 g l-1 γ-PGA, with a 31.5% increase compared with strain LL3. Second, a strong promoter PC 2up was inserted into the upstream of icd gene, to generate strain NK-A7, which further led to a 33.5% improvement in the γ-PGA titre, achieving 6.46 g l-1 . The NADPH level was improved by regulating the expression of pgi and gndA. Third, metabolic evolution was carried out to generate strain NK-A9E, which showed a comparable γ-PGA titre with strain NK-A7. Finally, the srf and itu operons were deleted respectively, from the original strains NK-A7 and NK-A9E. The resulting strain NK-A11 exhibited the highest γ-PGA titre (7.53 g l-1 ), with a 2.05-fold improvement compared with LL3. The results demonstrated that the approaches described here efficiently enhanced γ-PGA production in B. amyloliquefaciens fermentation.

Poly-Gamma-Glutamic Acid Secretion Protects Bacillus subtilis from Zinc and Copper Intoxication.

Project description:Zinc and copper are essential micronutrients that serve as a cofactors for numerous enzymes. However, when present at elevated concentrations, zinc and copper are highly toxic to bacteria. To combat the effects of zinc and copper excess, bacteria have evolved a wide array of defense mechanisms. Here, we show that the Gram-positive soil bacterium, Bacillus subtilis, produces the extracellular polymeric substance, poly-gamma-glutamate (γ-PGA) as a protective mechanism in response to zinc and copper excess. Furthermore, we provide evidence that zinc and copper dependent γ-PGA production is independent of the DegS-DegQ two-component regulatory system and likely occurs at a posttranscriptional level through the small protein, PgsE. These data provide new insight into bacterial metal resistance mechanisms and contribute to our understanding of the regulation of bacterial γ-PGA biosynthesis. IMPORTANCE Zinc and copper are potent antimicrobial compounds. As such, bacteria have evolved a diverse range of tools to prevent metal intoxication. Here, we show that the Gram-positive model organism, Bacillus subtilis, produces poly-gamma-glutamic acid (γ-PGA) as a protective mechanism against zinc and copper intoxication and that zinc and copper dependent γ-PGA production occurs by a yet undefined mechanism independent of known γ-PGA regulation pathways.

Poly-?-Glutamic Acid (PGA)-Producing Bacillus Species Isolated from Kinema, Indian Fermented Soybean Food.

Project description:Kinema, an ethnic fermented, non-salted and sticky soybean food is consumed in the eastern part of India. The stickiness is one of the best qualities of good kinema preferred by consumers, which is due to the production of poly-?-glutamic acid (PGA). Average load of Bacillus in kinema was 10(7) cfu/g and of lactic acid bacteria was 10(3) cfu/g. Bacillus spp. were screened for PGA-production and isolates of lactic acid bacteria were also tested for degradation of PGA. Only Bacillus produced PGA, none of lactic acid bacteria produced PGA. PGA-producing Bacillus spp. were identified by phenotypic characterization and also by 16S rRNA gene sequencing as Bacillus subtilis, B. licheniformis and B. sonorensis.

Properties of Poly-γ-Glutamic Acid Producing-Bacillus Species Isolated From Ogi Liquor and Lemon-Ogi Liquor.

Project description:Poly-γ-glutamic acid (γPGA) is a natural and promising biopolymer synthesized by Bacillus spp. during fermentation. This study isolated Bacillus spp. from ogi steep liquor (OSL) and lemon-ogi steep liquor (LOSL) using standard methods and determined the γPGA-producing ability. The antimicrobial and angiotensin-converting enzyme (ACE) inhibitory activities of γPGA polymer were evaluated and isolates were sequenced. Four isolates (TA004, TA006, TA011, TA012) selected based on phenotypic characterization and stickiness (<15 cm) showed antibacterial activity against different pathogens with the highest activity found in TA004 (22.5 mm) and least in TA011 (16.6 mm). Furthermore, time-kill assay showed that the combined γPGA polymer was more effective and demonstrated bactericidal activity over individual γPGA which are bacteriostatic in nature. All γPGA polymer exhibited ACE properties except TA011. The highest IC50 was observed in TA006 (0.11 mg/ml) and least in TA004 (0.35 mg/ml). TA004 had the highest molecular weight (261 kDa) while TA011 had the least (194.97 kDa). In addition, all γPGA exhibited characteristic peaks at 3413-3268 cm-1 and 1722-1664 cm-1 that corresponded to amine N-H stretching intensities and C = O stretching in COOH. Bacillus isolates were identified as TA004 (B. subtilis-GenBank: MH782061), TA006 (B. amyloliquefaciens- GenBank: MH782075), TA011 (B. subtilis- GenBank: MH782088), TA012 (B. subtilis- GenBank: MH782083). OSL and LOSL have the potential for developing functional foods with a valuable effect on health.

Solution scattering study of the Bacillus subtilis PgdS enzyme involved in poly-γ-glutamic acids degradation.

Project description:The PgdS enzyme is a poly-γ-glutamic (γ-PGA) hydrolase, which has potential application for a controllable degradation of γ-PGA by enzymatic depolymerization; however, the structure of PgdS is still unknown. Here, to study in detail the full-length PgdS structure, we analyze the low-resolution architecture of PgdS hydrolase from Bacillus subtilis in solution using small angle X-ray scattering (SAXS) method. Combining with other methods, like dynamic light scattering and mutagenesis analyses, a model for the full length structure and the possible substrate delivery route of PgdS are proposed. The results will provide useful hints for future investigations into the mechanisms of γ-PGA degradation by the PgdS hydrolase and may provide valuable practical information.
