Bioremediation of malachite green dye by two bacterial strains isolated from textile effluents.

ABSTRACT: Globally, water pollution from the textile industries is an alarming issue. Malachite Green dye of the triphenylmethane group is an extensively used dye in the fabric industries that is emitted through textile wastewater. This study aimed to isolate and characterize potential Malachite Green (MG) dye degrading bacteria from textile effluents. Different growth and culture parameters such as temperature, pH and dye concentration were optimized to perform the dye-degradation assay using different concentrations of MG dye in the mineral salt medium. A photo-electric-colorimeter was used to measure the decolorizing activity of bacteria at different time intervals after aerobic incubation. Two potential bacterial strains of Enterobacter spp. CV-S1 (accession no: MH450229) and Enterobacter spp. CM-S1 (accession no: MH447289) were isolated from textile effluents exhibiting potential MG dye decoloring efficiency. Further, the RAPD analysis and 16S rRNA sequencing confirmed the genetic differences of the isolated strains. Enterobacter sp CV-S1 and Enterobacter sp CM-S1 can completely decolor MG dye up to 15 mg/L under shaking condition without any requirement of sole carbon source. Thus, these two bacteria have the potency to be utilized in the textile wastewater treatment plant.

Project description:Aurivillius oxides ferroelectric layered materials are formed by bismuth oxide and pseu-do-perovskite layers. They have a good ionic conductivity, which is beneficial for various photo-catalyzed reactions. Here, we synthesized ultra-thin nanosheets of two different Aurivillius oxides, Bi2WO6 (BWO) and Bi2MoO6 (BMO), by using a hard-template process. All materials were characterized through XRD, TEM, FTIR, TGA/DSC, DLS/ELS, DRS, UV-Vis. Band gap material (Eg) and potential of the valence band (EVB) were calculated for BWO and BMO. In contrast to previous reports on the use of multi composite materials, a new procedure for photocatalytic efficient BMO nanosheets was developed. The procedure, with an additional step only, avoids the use of composite materials, improves crystal structure, and strongly reduces impurities. BWO and BMO were used as photocatalysts for the degradation of the water pollutant dye malachite green (MG). MG removal kinetics was fitted with Langmuir-Hinshelwood model obtaining a kinetic constant k = 7.81 × 10-2 min-1 for BWO and k = 9.27 × 10-2 min-1 for BMO. Photocatalytic dye degradation was highly effective, reaching 89% and 91% MG removal for BWO and BMO, respectively. A control experiment, carried out in the absence of light, allowed to quantify the contribution of adsorption to MG removal process. Adsorption contributed to MG removal by a 51% for BWO and only by a 19% for BMO, suggesting a different degradation mechanism for the two photocatalysts. The advanced MG degradation process due to BMO is likely caused by the high crystallinity of the material synthetized with the new procedure. Reuse tests demonstrated that both photocatalysts are highly active and stable reaching a MG removal up to 95% at the 10th reaction cycle. These results demonstrate that BMO nanosheets, synthesized with an easy additional step, achieved the best degradation performance, and can be successfully used for environmental remediation applications.

Project description:There is high demand for herbicides based on the necessity to increase crop production to satisfy world-wide demands. Nevertheless, there are negative impacts of herbicide use, manifesting as selection for resistant weeds, production of toxic metabolites from partial degradation of herbicides, changes in soil microbial communities and biogeochemical cycles, alterations in plant nutrition and soil fertility, and persistent environmental contamination. Some herbicides damage non-target microorganisms via directed interference with host metabolism and via oxidative stress mechanisms. For these reasons, it is necessary to identify sustainable, efficient methods to mitigate these environmental liabilities. Before the degradation process can be initiated by microbial enzymes and metabolic pathways, microorganisms need to tolerate the oxidative stresses caused by the herbicides themselves. This can be achieved via a complex system of enzymatic and non-enzymatic antioxidative stress systems. Many of these response systems are not herbicide specific, but rather triggered by a variety of substances. Collectively, these nonspecific response systems enhance the survival and fitness potential of microorganisms. Biodegradation studies and remediation approaches have relied on individually selected strains to effectively remediate herbicides in the environment. Nevertheless, it has been shown that microbial communication systems that modulate social relationships and metabolic pathways inside biofilm structures among microorganisms are complex; therefore, use of isolated strains for xenobiotic degradation needs to be enhanced using a community-based approach with biodegradation pathway integration. Bioremediation efforts can use omics-based technologies to gain a deeper understanding of the molecular complexes of bacterial communities to achieve to more efficient elimination of xenobiotics. With this knowledge, the possibility of altering microbial communities is increased to improve the potential for bioremediation without causing other environmental impacts not anticipated by simpler approaches. The understanding of microbial community dynamics in free-living microbiota and those present in complex communities and in biofilms is paramount to achieving these objectives. It is also essential that non-developed countries, which are major food producers and consumers of pesticides, have access to these techniques to achieve sustainable production, without causing impacts through unknown side effects.

Project description:Phytogenic magnetic nanoparticles (PMNPs) were fabricated using plant leaves' extract of Fraxinus chinensis Roxb. and then, the surfaces of the PMNPs were functionalized by 3-mercaptopropionic acid (3-MPA) to investigate the adsorptive removal of the toxic dye malachite green (MG) from aqueous solutions. The preparation and coating of 3-MPA on the surface of the PMNPs was confirmed and characterized using different techniques, which are UV-visible spectroscopy, Fourier transform infrared spectroscopy (FTIR), X-ray diffractometry (XRD), scanning electron microscopy with integrated energy dispersive X-ray analysis (SEM-EDX), transmission electron microscopy (TEM), X-ray photoelectron spectroscopy (XPS), vibrating sample magnetometry (VSM), Brunauer-Emmett-Teller (BET) analysis and thermogravimetric analysis (TGA). The hysteresis loops of 3-MPA@PMNPs depicted an excellent superparamagnetic nature with saturation magnetization values of 50.95 emu g-1. The prepared material showed the highest adsorptive rate (98.57% MG removal within 120 min) and an estimated comparable adsorptive capacity of 81.2 mg g-1 at 25 °C. The experimental data were well fitted to the Langmuir isotherm, indicating the monolayer adsorption of MG onto 3-MPA@PMNPs. Furthermore, the kinetic data agreed well with the pseudo-second-order model, indicating the removal of MG by chemisorption and/or ion-exchange mechanism. Thermodynamic study confirmed that the adsorption of MG was exothermic and spontaneous. The high adsorptive removal of the dye not only persisted over a wide pH range (6-12), but the material also demonstrated high selectivity in the presence of co-existing ions (i.e. Pd2+ and Cd2+) along with the fastest separation times (35 s) from aqueous solutions. The recovered adsorbent (3-MPA@PMNPs) was reused five times and maintained a removal efficiency of more than 85%. Therefore, the prepared novel 3-MPA@PMNPs can be employed as an alternative low-cost sorbent material for the removal cationic dyes from textile wastewater. In addition, this green nanotechnology/strategy can easily be implemented in low-economy countries for wastewater treatment.
