ZnO Tips Dotted with Au Nanoparticles-Advanced SERS Determination of Trace Nicotine.

ABSTRACT: Long-term exposure to nicotine causes a variety of human diseases, such as lung damage/adenocarcinoma, nausea and vomiting, headache, incontinence and heart failure. In this work, as a surface-enhanced Raman scattering (SERS) substrate, zinc oxide (ZnO) tips decorated with gold nanoparticles (AuNPs) are fabricated and designated as ZnO/Au. Taking advantage of the synergistic effect of a ZnO semiconductor with morphology of tips and AuNPs, the ZnO/Au-based SERS assay for nicotine demonstrates high sensitivity and the limit of detection 8.9 × 10-12 mol/L is reached, as well as the corresponding linear dynamic detection range of 10-10-10-6 mol/L. Additionally, the signal reproducibility offered by the SERS substrate could realize the reliable determination of trace nicotine in saliva.

Au Nanoparticles (NPs) Decorated Co Doped ZnO Semiconductor (Co400-ZnO/Au) Nanocomposites for Novel SERS Substrates.

Project description:Au nanoparticles were decorated on the surface of Co-doped ZnO with a certain ratio of Co2+/Co3+ to obtain a novel semiconductor-metal composite. The optimal substrate, designated as Co400-ZnO/Au, is beneficial to the promotion of separation efficiency of electron and hole in a semiconductor excited under visible laser exposure, which the enhances localized surface plasmon resonance (LSPR) of the Au nanoparticles. As an interesting finding, during Co doping, quantum dots of ZnO are generated, which strengthen the strong semiconductor metal interaction (SSSMI) effect. Eventually, the synergistic effect effectively advances the surface enhancement Raman scattering (SERS) performance of Co400-ZnO/Au composite. The enhancement mechanism is addressed in-depth by morphologic characterization, UV-visible, X-ray diffraction, photoluminescence, X-ray photoelectron spectroscopy, density functional theory, and finite difference time domain (FDTD) simulations. By using Co400-ZnO/Au, SERS detection of Rhodamine 6G presents a limit of detection (LOD) of 1 × 10-9 M. As a real application, the Co400-ZnO/Au-based SERS method is utilized to inspect tyramine in beer and the detectable concentration of 1 × 10-8 M is achieved. In this work, the doping strategy is expected to realize a quantum effect, triggering a SSSMI effect for developing promising SERS substrates in future.

Assembled Au/ZnO Nano-Urchins for SERS Sensing of the Pesticide Thiram.

Project description:In this paper, we are relating a significant improvement of the SERS effect achieved with assembled Au/ZnO nano-urchins. This improvement is realized thanks to an excellent capacity of adsorption (denoted K) for thiram molecules on these plasmonic nano-urchins, which is a key point to be taken into account for obtaining a SERS spectrum. Moreover, this outlook may be employed for different types of plasmonic substrates and for a wide number of molecules. We studied the capacity of the assembled Au/ZnO nano-urchins to be sensitive to the pesticide thiram, which adsorbs well on metals via the metal-sulfur bond. For the thiram detection, we found a limit concentration of 10 pM, a value of this capacity of adsorption (K) of 9.5 × 106 M-1 and a factor of analytical enhancement equal to 1.9 × 108.

Au Nanoparticles@Si Nanowire Oligomer Arrays for SERS: Dimers Are Best.

Project description:We report the synthesis of vertically aligned silicon nanowire (VA-SiNW) oligomer arrays coated with Au nanoparticle (NP) monolayers via a combination of colloidal lithography, metal-assisted chemical etching, and directed NP assembly. Arrays of SiNW monomers (i.e., isolated NWs), dimers, and tetramers are synthesized, decorated with AuNPs, and tested for their performance in surface-enhanced Raman spectroscopy. The ∼20 nm AuNPs easily enter within the ca. 40 nm gaps of the SiNW oligomers, thus reaching the hot spot region. At 785 nm excitation, the AuNPs@SiNW dimer arrays provide the highest Raman signal, in agreement with electromagnetic simulations showing a high electric field enhancement at the Au/Si interface within the dimer gap region.

ZnO Nanocages Decorated with Au@AgAu Yolk-Shell Nanomaterials for SERS-Based Detection of Hyperuricemia.

Project description:Surface-enhanced Raman scattering (SERS) is widely recognized as a highly sensitive technology for chemical detection and biological sensing. In SERS-based biomedical applications, developing highly efficient sensing platforms based on SERS plays a pivotal role in monitoring disease biomarker levels and facilitating the early detection of cancer biomarkers. Hyperuricemia, characterized by abnormally high concentrations of uric acid (UA) in the blood, was associated with a range of diseases, such as gouty arthritis, heart disease, and acute kidney injury. Recent reports have demonstrated the correlation between UA concentrations in blood and tears. In this work, we report the fabrication of SERS substrates utilizing ZnO nanocages and yolk-shell-structured plasmonic nanomaterials for the noninvasive detection of UA in tears. This innovative SERS substrate enables noninvasive and sensitive detection of UA to prevent hyperuricemia-related diseases.

SERS-PCR assays of SARS-CoV-2 target genes using Au nanoparticles-internalized Au nanodimple substrates.

Project description:The reverse transcription-polymerase chain reaction (RT-PCR) method has been adopted worldwide to diagnose severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Although this method has good sensitivity and specificity, there is a need to develop a more rapid diagnostic technology, given the virus's rapid spread. However, the RT-PCR method takes a long time to diagnose SARS-CoV-2 because of the required thermocycling steps. Therefore, we developed a surface-enhanced Raman scattering (SERS)-PCR detection method using an AuNP-internalized Au nanodimple substrate (AuNDS) to shorten the diagnosis time by reducing the number of thermocycling steps needed to amplify the DNA. For the representative target markers, namely, the envelope protein (E) and RNA-dependent RNA polymerase (RdRp) genes of SARS-CoV-2, 25 RT-PCR thermocycles are required to reach a detectable threshold value, while 15 cycles are needed for magnetic bead-based SERS-PCR when the initial DNA concentration was 1.00× 105 copies/μL. However, only 8 cycles are needed for the AuNDS-based SERS-PCR. The corresponding detectable target DNA concentrations were 3.36 × 1012, 3.28 × 109, and 2.56 × 107 copies/μL, respectively. Therefore, AuNDS-based SERS-PCR is seen as being a new molecular diagnostic platform that can shorten the time required for the thermocycling steps relative to the conventional RT-PCR.

Au-Coated ZnO Surface-Enhanced Raman Scattering (SERS) Substrates: Synthesis, Characterization, and Applications in Exosome Detection.

Project description:Developing a biomolecular detection method that minimizes photodamage while preserving an environment suitable for biological constituents to maintain their physiological state is expected to drive new diagnostic and mechanistic breakthroughs. In addition, ultra-sensitive diagnostic platforms are needed for rapid and point-of-care technologies for various diseases. Considering this, surface-enhanced Raman scattering (SERS) is proposed as a non-destructive and sensitive approach to address the limitations of fluorescence, electrochemical, and other optical detection techniques. However, to advance the applications of SERS, novel approaches that can enhance the signal of substrate materials are needed to improve reproducibility and costs associated with manufacture and scale-up. Due to their physical properties and synthesis, semiconductor-based nanostructures have gained increasing recognition as SERS substrates; however, low signal enhancements have offset their widespread adoption. To address this limitation and assess the potential for use in biological applications, zinc oxide (ZnO) was coated with different concentrations (0.01-0.1 M) of gold (Au) precursor. When crystal violet (CV) was used as a model target with the synthesized substrates, the highest enhancement was obtained with ZnO coated with 0.05 M Au precursor. This substrate was subsequently applied to differentiate exosomes derived from three cell types to provide insight into their molecular diversity. We anticipate this work will serve as a platform for colloidal hybrid SERS substrates in future bio-sensing applications.

Localized Energy Band Bending in ZnO Nanorods Decorated with Au Nanoparticles.

Project description:Surface decoration by means of metal nanostructures is an effective way to locally modify the electronic properties of materials. The decoration of ZnO nanorods by means of Au nanoparticles was experimentally investigated and modelled in terms of energy band bending. ZnO nanorods were synthesized by chemical bath deposition. Decoration with Au nanoparticles was achieved by immersion in a colloidal solution obtained through the modified Turkevich method. The surface of ZnO nanorods was quantitatively investigated by Scanning Electron Microscopy, Transmission Electron Microscopy and Rutherford Backscattering Spectrometry. The Photoluminescence and Cathodoluminescence of bare and decorated ZnO nanorods were investigated, as well as the band bending through Mott-Schottky electrochemical analyses. Decoration with Au nanoparticles induced a 10 times reduction in free electrons below the surface of ZnO, together with a decrease in UV luminescence and an increase in visible-UV intensity ratio. The effect of decoration was modelled with a nano-Schottky junction at ZnO surface below the Au nanoparticle with a Multiphysics approach. An extensive electric field with a specific halo effect formed beneath the metal-semiconductor interface. ZnO nanorod decoration with Au nanoparticles was shown to be a versatile method to tailor the electronic properties at the semiconductor surface.

Three-Dimensional Dendritic Au-Ag Substrate for On-Site SERS Detection of Trace Molecules in Liquid Phase.

Project description:The development of a facile surface-enhanced Raman scattering (SERS) sensor for the on-site detection of trace molecules in liquid phase is a compelling need. In this paper, a three-dimensional (3D) dendritic Au-Ag nanostructure was constructed by a two-step electro displacement reaction in a capillary tube for the on-site liquid phase detection of trace molecules. The multiplasmon resonance mechanism of the dendritic Au-Ag structure was simulated using the finite-difference time domain (FDTD) method. It was confirmed that the highly branched 3D structure promoted the formation of high-density "hot spots" and interacted with the gold nanoparticles at the dendrite tip, gap, and surface to maximize the spatial electric field, which allowed for high signal intensification to be observed. More importantly, the unique structure of the capillary made it possible to achieve the on-site detection of trace molecules in liquids. Using Rhodamine 6G (R6G) solution as a model molecule, the 3D dendritic Au-Ag substrate exhibited a high detection sensitivity (10-13 mol/L). Furthermore, the developed sensor was applied to the detection of antibacterial agents, ciprofloxacin (CIP), with clear Raman characteristic peaks observed even at concentrations as low as 10-9 mol/L. The results demonstrated that the 3D dendritic Au-Ag sensor could successfully realize the rapid on-site SERS detection of trace molecules in liquids, providing a promising platform for ultrasensitive and on-site liquid sample analysis.

Au nanoparticles for SERS: Temperature-controlled nanoparticle morphologies and their Raman enhancing properties.

Project description:Quasi-fractal gold nanoparticles can be synthesized via a modified and temperature controlled procedure initially used for the synthesis of star-like gold nanoparticles. The surface features of nanoparticles lead to improved enhancement of Raman scattering intensity of analyte molecules due to the increased number of sharp surface features possessing numerous localized surface plasmon resonances (LSPR). The LSPR is affected by the size and shape of surface features as well as inter-nanoparticle interactions, as these affect the oscillation modes of electrons on the nanoparticle surfaces. The effect of the particle morphologies on the localized surface plasmon resonance (LSPR) and on the surface-enhancing capabilities of these nanoparticles is explored by comparing different nanoparticle morphologies and concentrations. We show that in a fixed nanoparticle concentration regime, quasi-fractal gold nanoparticles (gold nanocaltrop) provide the highest level of surface enhancement, whereas spherical nanoparticles provide the largest enhancement in a fixed gold concentration regime. The presence of highly branched features enables these nanoparticles to couple with a laser wavelength, despite having no strong absorption band and hence no single surface plasmon resonance. This cumulative LSPR may allow these nanoparticles to be used in a variety of applications in which laser wavelength flexibility is beneficial, such as in medical imaging applications where fluorescence at short laser wavelengths may be coupled with non-fluorescing long laser wavelengths for molecular sensing.
