Lectotypification of Chamaecyparishodginsii of the Cupressaceae.

ABSTRACT: Recent phylogenetic studies have suggested that the monotypic Fokienia A.Henry & H.H.Thomas is nested within Chamaecyparis Spach, which is in agreement with separate morphological studies. Here the authors confirm a previous taxonomic treatment that incorporated Fokieniahodginsii (Dunn) A.Henry & H.H.Thomas into Chamaecyparis based on the monophyly requirement of taxonomy, i.e. Chamaecyparishodginsii (Dunn) Rushforth. In addition, the type collection of the basionym Cupressushodginsii Dunn was found to contain three sheets of specimens, one in K including a vegetative branch (K000088294) and a separate ovulate cone (K001090486), a second one in A (A00022477), and a third one in IBSC (IBSC0016081). All three specimens are marked with Hongkong Herbarium No. 3505, but only the two specimens in K and IBSC possess similar handwriting of "Cupressushodginsii Dunn". The two specimens should be considered as syntypes according to the Shenzhen Code. The specimen in K is better preserved but it is a mixture according to the collection label: cones from Foochow (Fuzhou) and foliage from Yenping (Nanping). We lectotypified the name Cupressushodginsii with K000088294 because the specimen is well preserved and has enough characters for identification. Moreover, an ovulate cone (K001090486) is on the same sheet.

Lectotypification of Tsuga longibracteata W.C.Cheng (Pinaceae).

Project description:W.C.Cheng did not clearly indicate the herbarium repository of the type specimen (Y.Tsiang 7712) when he described Tsuga longibracteata W.C.Cheng. Later, researchers suggested that the type is either in NAS or in PE. However, we found more than one duplicate of the type collection in both NAS and PE. Following the Shenzhen Code, we lectotypify the name T. longibracteata with Y.Tsiang 7712 (PE00003223) that bears a handwritten identification of W.C.Cheng.

Phylogenetic placement and lectotypification of Pseudotryblidium neesii (Helotiales, Leotiomycetes).

Project description:A phylogenetic analysis of combined rDNA LSU and ITS sequence data was carried out to determine the phylogenetic placement of specimens identified as Pseudotryblidium neesii. The species forms a distinct clade within Dermateaceae (Helotiales, Leotiomycetes) with Rhizodermea veluwiensis and two Dermea species. The geographical distribution of this species, previously known only from Europe on Abies alba, is extended to north-western North America where it grows exclusively on A. grandis. The name P. neesii is lectotypified in order to disentangle the complicated nomenclature of the species. A new, detailed description of P. neesii with illustrations is provided after comparison of sequenced specimens with the type material. Furthermore, the new combination Pseudographis rufonigra (basionym Peziza rufonigra) is made for a fungus previously known as Pseudographis pinicola.

Plastome Phylogenomic and Biogeographical Study on Thuja (Cupressaceae).

Project description:Investigating the biogeographical disjunction of East Asian and North American flora is key to understanding the formation and dynamics of biodiversity in the Northern Hemisphere. The small Cupressaceae genus Thuja, comprising five species, exhibits a typical disjunct distribution in East Asia and North America. Owing to obscure relationships, the biogeographical history of the genus remains controversial. Here, complete plastomes were employed to investigate the plastome evolution, phylogenetic relationships, and biogeographic history of Thuja. All plastomes of Thuja share the same gene content arranged in the same order. The loss of an IR was evident in all Thuja plastomes, and the B-arrangement as previously recognized was detected. Phylogenomic analyses resolved two sister pairs, T. standishii-T. koraiensis and T. occidentalis-T. sutchuenensis, with T. plicata sister to T. occidentalis-T. sutchuenensis. Molecular dating and biogeographic results suggest the diversification of Thuja occurred in the Middle Miocene, and the ancestral area of extant species was located in northern East Asia. Incorporating the fossil record, we inferred that Thuja likely originated from the high-latitude areas of North America in the Paleocene with a second diversification center in northern East Asia. The current geographical distribution of Thuja was likely shaped by dispersal events attributed to the Bering Land Bridge in the Miocene and subsequent vicariance events accompanying climate cooling. The potential effect of extinction may have profound influence on the biogeographical history of Thuja.

Terpenoid Compositions of Resins from Callitris Species (Cupressaceae).

Project description:The environmental fate of conifer resins and their natural product compounds as mixtures is of importance for source, alteration, and transport studies. The compound compositions of resins of the common Callitris species (Cupressaceae) based on gas chromatography-mass spectrometry have not been reported. Results show that diterpenoids were the most abundant components and callitrisic acid was present in the resin extracts of all Callitris species analyzed. Significant amounts of 4-epi-pimaric and sandaracopimaric acids, with lesser communic, ozic, and lambertianic acids, were also in the mixtures. Phenolic diterpenoids, for example, ferruginol, hinokiol, were found in trace quantities in some samples. Thus, callitrisic acid and 4-epi-pimaric acid are the characteristic diterpenoids of Callitris species that are amenable to molecular biomarker analyses in geological or environmental applications.

The complete chloroplast genome of Juniperus rigida (Cupressaceae)

Project description:Abstract The complete chloroplast (cp) genome of Juniperus rigida was assembled using Illumina short reads. The assembled plastome showed a circular structure with an overall size of 127,720 bp. Inverted repeat (IRs) regions were absent from the J. rigida cp genome. A total of 119 genes were predicted, including 82 protein-coding genes, 33 tRNA genes, and 4 rRNA genes. The overall GC content of J. rigida cp genome was 34.92%. Phylogenetic analysis among J. rigida and 19 other Cupressaceae species showed that J. rigida clustered together with J. formosana. The J. rigida cp genome presented in this study will provide useful genetic resource for further evolutionary studies of the genus Juniperus as well as Cupressaceae.

Typification of Juniperus pingii W.C.Cheng (Cupressaceae).

Project description:W.C.Cheng validly published the name Juniperus pingii W.C.Cheng in 1944 by providing a validating Latin diagnosis in de Ferré (1944), but he failed to cite any specimen. He repeated the publication of the name in 1947 with the same Latin diagnosis; he thus published an isonym "J. pingii" under Art. 6 Note 2. Cheng (1947) lectotypified the name J. pingii when he designated W.C.Cheng 1015 as the "type" of the isonym. Farjon (2005) overlooked this early designation and his lectotypification of the name with the illustration from the 1944 protologue is not effective as the W.C.Cheng 1015 specimen is extant.

A lectotype is designated for this name, with an updated morphological description based on fresh material. The name Melastoma candidum f. albiflorum is proposed as a heterotypic synonym of Melastoma candidum.

DNA variation in a conifer, Cryptomeria japonica (Cupressaceae sensu lato).

Project description:We investigated the nucleotide variation of a conifer, Cryptomeria japonica, and the divergence between this species and its closest relative, Taxodium distichum, at seven nuclear loci (Acl5, Chi1, Ferr, GapC, HemA, Lcyb, and Pat). Samples of C. japonica were collected from three areas, Kantou-Toukai, Hokuriku, and Iwate. No apparent geographic differentiation was found among these samples. However, the frequency spectrum of the nucleotide polymorphism revealed excesses of intermediate-frequency variants, which suggests that the population was not panmictic and a constant size in the past. The average nucleotide diversity, pi, for silent sites was 0.00383. However, values of pi for silent sites vary among loci. Comparisons of polymorphism to divergence among loci (the HKA test) showed that the polymorphism at the Acl5 locus was significantly lower. We also observed a nearly significant excess of replacement polymorphisms at the Lcyb locus. These results suggested possibilities of natural selection acting at some of the loci. Intragenic recombination was detected only once at the Chi1 locus and was not detected at the other loci. The low level of population recombination rate, 4Nr, seemed to be due to both low level of recombination, r, and small population size, N.

Distribution of living Cupressaceae reflects the breakup of Pangea.

Project description:Most extant genus-level radiations in gymnosperms are of Oligocene age or younger, reflecting widespread extinction during climate cooling at the Oligocene/Miocene boundary [?23 million years ago (Ma)]. Recent biogeographic studies have revealed many instances of long-distance dispersal in gymnosperms as well as in angiosperms. Acting together, extinction and long-distance dispersal are likely to erase historical biogeographic signals. Notwithstanding this problem, we show that phylogenetic relationships in the gymnosperm family Cupressaceae (162 species, 32 genera) exhibit patterns expected from the Jurassic/Cretaceous breakup of Pangea. A phylogeny was generated for 122 representatives covering all genera, using up to 10,000 nucleotides of plastid, mitochondrial, and nuclear sequence per species. Relying on 16 fossil calibration points and three molecular dating methods, we show that Cupressaceae originated during the Triassic, when Pangea was intact. Vicariance between the two subfamilies, the Laurasian Cupressoideae and the Gondwanan Callitroideae, occurred around 153 Ma (124-183 Ma), when Gondwana and Laurasia were separating. Three further intercontinental disjunctions involving the Northern and Southern Hemisphere are coincidental with or immediately followed the breakup of Pangea.
