Complete Genome Sequence of a Thin-Sheath Mutant of the Phototropic Cyanobacterium Calothrix sp. Strain PCC 7716.

ABSTRACT: Calothrix sp. strain PCC 7716 is a filamentous cyanobacterium whose morphology is tapered, with basal-apical polarity. The apical filament shows positive phototropism toward white light or blue light. To elucidate the molecular basis of the phototropism, we determined the complete genome sequence of a spontaneous mutant of this strain that has a thin sheath and is suitable for genomic DNA extraction.

Sheathless mutant of Cyanobacterium Gloeothece sp. strain PCC 6909 with increased capacity to remove copper ions from aqueous solutions.

Project description:The cyanobacterium Gloeothece sp. strain PCC 6909 and its sheathless mutant were tested for their abilities to remove copper ions from aqueous solutions, with the aim of defining the role of the various outermost polysaccharidic investments in the removal of the metal ions. Microscopy studies and chemical analyses revealed that, although the mutant does not possess a sheath, it releases large amounts of polysaccharidic material (released exocellular polysaccharides [RPS]) into the culture medium. The RPS of the wild type and the mutant are composed of the same 11 sugars, although they are present in different amounts, and the RPS of the mutant possesses a larger amount of acidic sugars and a smaller amount of deoxysugars than the wild type. Unexpectedly, whole cultures of the mutant were more effective in the removal of the heavy metal than the wild type (46.3 +/- 3.1 and 26.7 +/- 1.5 mg of Cu(2+) removed per g of dry weight, respectively). Moreover, we demonstrated that the contribution of the sheath to the metal-removal capacity of the wild type is scarce and that the RPS of the mutant is more efficient in removing copper. This suggests that the metal ions are preferably bound to the cell wall and to RPS functional groups rather than to the sheath. Therefore, the increased copper binding efficiency observed with the sheathless mutant can be attributed to the release of a polysaccharide containing larger amounts and/or more accessible functional groups (e.g., carboxyl and amide groups).

Genomic structure of the cyanobacterium Synechocystis sp. PCC 6803 strain GT-S.

Project description:Synechocystis sp. PCC 6803 is the most popular cyanobacterial strain, serving as a standard in the research fields of photosynthesis, stress response, metabolism and so on. A glucose-tolerant (GT) derivative of this strain was used for genome sequencing at Kazusa DNA Research Institute in 1996, which established a hallmark in the study of cyanobacteria. However, apparent differences in sequences deviating from the database have been noticed among different strain stocks. For this reason, we analysed the genomic sequence of another GT strain (GT-S) by 454 and partial Sanger sequencing. We found 22 putative single nucleotide polymorphisms (SNPs) in comparison to the published sequence of the Kazusa strain. However, Sanger sequencing of 36 direct PCR products of the Kazusa strains stored in small aliquots resulted in their identity with the GT-S sequence at 21 of the 22 sites, excluding the possibility of their being SNPs. In addition, we were able to combine five split open reading frames present in the database sequence, and to remove the C-terminus of an ORF. Aside from these, two of the Insertion Sequence elements were not present in the GT-S strain. We have thus become able to provide an accurate genomic sequence of Synechocystis sp. PCC 6803 for future studies on this important cyanobacterial strain.

Novel DNA-binding proteins in the cyanobacterium Anabaena sp. strain PCC 7120.

Project description:As an approach towards elucidation of the biochemical regulation of the progression of heterocyst differentiation in Anabaena sp. strain PCC 7120, we have identified proteins that bind to a 150-bp sequence upstream from hepC, a gene that plays a role in the synthesis of heterocyst envelope polysaccharide. Such proteins were purified in four steps from extracts of vegetative cells of Anabaena sp. Two of these proteins (Abp1 and Abp2) are encoded by neighboring genes in the Anabaena sp. chromosome. The genes that encode the third (Abp3) and fourth (Abp4) proteins are situated at two other loci in that chromosome. Insertional mutagenesis of abp2 and abp3 blocked expression of hepC and hepA and prevented heterocyst maturation and aerobic fixation of N(2).

Global analysis of circadian expression in the cyanobacterium Synechocystis sp. strain PCC 6803.

Project description:Cyanobacteria are the only bacterial species found to have a circadian clock. We used DNA microarrays to examine circadian expression patterns in the cyanobacterium Synechocystis sp. strain PCC 6803. Our analysis identified 54 (2%) and 237 (9%) genes that exhibited circadian rhythms under stringent and relaxed filtering conditions, respectively. The expression of most cycling genes peaked around the time of transition from subjective day to night, suggesting that the main role of the circadian clock in Synechocystis is to adjust the physiological state of the cell to the upcoming night environment. There were several chromosomal regions where neighboring genes were expressed with similar circadian patterns. The physiological functions of the cycling genes were diverse and included a wide variety of metabolic pathways, membrane transport, and signal transduction. Genes involved in respiration and poly(3-hydroxyalkanoate) synthesis showed coordinated circadian expression, suggesting that the regulation is important for the supply of energy and carbon source in the night. Genes involved in transcription and translation also followed circadian cycling patterns. These genes may be important for output of the rhythmic information generated by the circadian clock. Our findings provided critical insights into the importance of the circadian clock on cellular physiology and the mechanism of clock-controlled gene regulation.

Cell surface-associated proteins in the filamentous cyanobacterium Anabaena sp. strain PCC 7120.

Project description:The cell surface senses environmental changes first and transfers signals into the cell. To understand the response to environmental changes, it is necessary to analyze cell surface components, particularly cell surface-associated proteins. We therefore investigated cell surface-associated proteins from the filamentous cyanobacterium Anabaena sp. strain PCC 7120. The cell surface-associated proteins extracted by an acidic buffer were resolved by SDS-PAGE. Eighteen proteins were identified from resolved bands by amino-terminal sequencing. Analysis of cell surface-associated proteins indicated that several proteins among them were involved in nucleic acid binding, protein synthesis, proteolytic activity and electron transfer, and other proteins were involved in the stress response.
