Project description:Carotenoids are isoprenoid pigments used by pharmaceutical, cosmetic, food and feed industry as antioxidants and colorants. Although traditional sources of carotenoids are fruits, vegetables and chemical synthesis, prospecting for alternative sinks of common and/or unusual carotenoids is important for the development of natural carotenoid industry. In this work, 30 pigmented bacterial strains from Fildes Peninsula in King George Island, Antarctica, were isolated and identified by 16S rRNA gene sequencing and classified in three phyla, Bacteroidetes, Firmicutes and Actinobacteria. After cells extraction, ten different carotenoids were identified based on the chromatographic and spectroscopic characteristic obtained by HPLC-PDA and HPLC-PDA-APCI-MS analyses. Strains assigned to Bacteroidetes affiliated to Flavobacterium, Chryseobacterium and Zobellia genera, presented a pigment profile composed of zeaxanthin, β-cryptoxanthin and β-carotene. Firmicutes strains of Planococcus genus produced a C50 carotenoid, identified as C.p. 450 glucoside. Actinobacteria isolates were mainly assigned to Arthrobacter genus, and few to Salinibacterium and Cryobacterium genera. Arthrobacter strains produced C50 carotenoids such as decaprenoxanthin and its glucosylated derivatives, as well as some C40 carotenoids such as lycopene which is used as synthesis precursors of the C50 carotenoids. Salinibacterium and Cryobacterium genera produced C.p. 450 free form and its glucosylated derivatives. Although most isolates produce carotenoids similar in diversity and quantity than those already reported in the literature, novel sources for C50 carotenoids results from this work. According to their carotenoid content, all isolates could be promising candidates for carotenoids production.

Project description:Antarctica harbors a great diversity of microorganisms, including bacteria, archaea, microalgae and yeasts. The Pseudomonas genus is one of the most diverse and successful bacterial groups described to date, but only eight species isolated from Antarctica have been characterized. Here, we present three potentially novel species isolated on King George Island. The most abundant isolates from four different environments, were genotypically and phenotypically characterized. Multilocus sequence analysis and 16S rRNA gene analysis of a sequence concatenate for six genes (16S, aroE, glnS, gyrB, ileS and rpoD), determined one of the isolates to be a new Pseudomonas mandelii strain, while the other three are good candidates for new Pseudomonas species. Additionally, genotype analyses showed the three candidates to be part of a new subgroup within the Pseudomonas fluorescens complex, together with the Antarctic species Pseudomonas antarctica and Pseudomonas extremaustralis. We propose terming this new subgroup P. antarctica. Likewise, phenotypic analyses using API 20 NE and BIOLOG® corroborated the genotyping results, confirming that all presented isolates form part of the P. fluorescens complex. Pseudomonas genus research on the Antarctic continent is in its infancy. To understand these microorganisms' role in this extreme environment, the characterization and description of new species is vital.

Project description:Exiguobacterium antarcticum is a psychotropic bacterium isolated for the first time from microbial mats of Lake Fryxell in Antarctica. Many organisms of the genus Exiguobacterium are extremophiles and have properties of biotechnological interest, e.g., the capacity to adapt to cold, which make this genus a target for discovering new enzymes, such as lipases and proteases, in addition to improving our understanding of the mechanisms of adaptation and survival at low temperatures. This study presents the genome of E. antarcticum B7, isolated from a biofilm sample of Ginger Lake on King George Island, Antarctic peninsula.

Project description:It has been proposed that Antarctic environments select microorganisms with unique biochemical adaptations, based on the tenet 'Everything is everywhere, but, the environment selects' by Baas-Becking. However, this is a hypothesis that has not been extensively evaluated. This study evaluated the fundamental prediction contained in this hypothesis-in the sense that species are structured in the landscape according to their local habitats-, using as study model the phylogenetic diversity of the culturable bacteria of Fildes Peninsula (King George Island, Antarctica). Eighty bacterial strains isolated from 10 different locations in the area, were recovered. Based on phylogenetic analysis of 16S rRNA gene sequences, the isolates were grouped into twenty-six phylotypes distributed in three main clades, of which only six are exclusive to Antarctica. Results showed that phylotypes do not group significantly by habitat type; however, local habitat types had phylogenetic signal, which support the phylogenetic niche conservatism hypothesis and not a selective role of the environment like the Baas-Becking hypothesis suggests. We propose that, more than habitat selection resulting in new local adaptations and diversity, local historical colonization and species sorting (i.e. differences in speciation and extinction rates that arise by interaction of species level traits with the environment) play a fundamental role on the culturable bacterial diversity in Antarctica.

Project description:The coasts of the West Antarctic Peninsula are strongly influenced by glacier meltwater discharge. The spatial structure and biogeochemical composition of inshore habitats are shaped by large quantities of terrigenous particulate material deposited in the vicinity of the coast, which impacts the pelagic and benthic ecosystems. We used a multitude of geochemical and environmental variables to identify the radius extension of the meltwater impact from the Fourcade Glacier into the fjord system of Potter Cove, King George Island. The k-means cluster algorithm, canonical correspondence analysis, variance analysis and Tukey's post hoc multiple comparison tests were applied to define and cluster coastal meltwater habitats. A minimum of 10 clusters were needed to classify the 8?km2 study area into meltwater fjord habitats (MFHs), fjord habitats and marine habitats. Strontium content in surface sediments is the main geochemical indicator for lithogenic creek discharge in Potter Cove. Furthermore, bathymetry, glacier distance and geomorphic positioning are the essential habitats explaining variables. The mean and maximum MFH extent amounted to 1?km and 2?km, respectively. Extrapolation of the identified meltwater impact ranges to King George Island coastlines, which are presently ice-covered bays and fjord areas, indicated an overall coverage of 200-400?km2 MFH, underpinning the importance of better understanding the biology and biogeochemistry in terrestrial marine transition zones.This article is part of the theme issue 'The marine system of the West Antarctic Peninsula: status and strategy for progress in a region of rapid change'.

Project description:Abstract Deschampsia antarctica Desv. can be found in diverse Antarctic habitats which may vary considerably in terms of environmental conditions and soil properties. As a result, the species is characterized by wide ecotypic variation in terms of both morphological and anatomical traits. The species is a unique example of an organism that can successfully colonize inhospitable regions due to its phenomenal ability to adapt to both the local mosaic of microhabitats and to general climatic fluctuations. For this reason, D. antarctica has been widely investigated in studies analyzing morphophysiological and biochemical responses to various abiotic stresses (frost, drought, salinity, increased UV radiation). However, there is little evidence to indicate whether the observed polymorphism is accompanied by the corresponding genetic variation. In the present study, retrotransposon?based iPBS markers were used to trace the genetic variation of D. antarctica collected in nine sites of the Arctowski oasis on King George Island (Western Antarctic). The genotyping of 165 individuals from nine populations with seven iPBS primers revealed 125 amplification products, 15 of which (12%) were polymorphic, with an average of 5.6% polymorphic fragments per population. Only one of the polymorphic fragments, observed in population 6, was represented as a private band. The analyzed specimens were characterized by low genetic diversity (uHe = 0.021, I = 0.030) and high population differentiation (FST = 0.4874). An analysis of Fu's FS statistics and mismatch distribution in most populations (excluding population 2, 6 and 9) revealed demographic/spatial expansion, whereas significant traces of reduction in effective population size were found in three populations (1, 3 and 5). The iPBS markers revealed genetic polymorphism of D. antarctica, which could be attributed to the mobilization of random transposable elements, unique features of reproductive biology, and/or geographic location of the examined populations. Deschampsia antarctica is among the two angiosperms considered as a native for the Antarctic. The retrotransposon?based iPBS markers applied in the study revealed low genetic variation of the species accompanied by surprisingly high population differentiation.

Project description:The paleoclimate during the Early Eocene in Maritime Antarctica is characterized by cool conditions without a pronounced dry season. Soils formed on volcanic material under such climate conditions in modern analogue environments are usually Andosols rich in nanocrystalline minerals without pedogenic smectite. The paleosols formed on volcanic material on King Georges Island are covered by basalts, dated by 6 new 40Ar/39Ar datings to 51-48 Ma, and are rich in smectite. A pedogenic origin of the smectites would suggest a semi-arid rather than a wet non-seasonal humid paleoclimate. To investigate the origin of the smectites in these paleosols we used X-ray diffraction and microscopic techniques. Minor mineralogical changes between the volcanic parent material and the paleosols and a homogenous distribution of smectites throughout the paleosol horizons indicate that these smectites were mainly inherited from the pyroclastic parent material, which was altered prior to surficial weathering. Nevertheless, the mineralogical properties, such as degree of crystallinity and octahedral site occupancy, of these smectites were modified during the ancient soil formation. Our findings highlight that trioctahedral smectites were a product of deuteric alteration of pyroclastic rocks and were progressively transformed to dioctahedral smectites during weathering in a soil environment on King George Island.

Project description:Chiral organochlorine compounds (OCs) were measured in various environmental matrices (air, soil and vegetation) from west Antarctica using high resolution gas chromatography coupled with high resolution mass spectrometry (HRGC/HRMS). They were generally detected at a global background level compared with the previous studies. α-HCH and PCB-183 was observed in all the matrices except PCB-183 in two soil samples, while PCB-95, -136, -149, -174, -176 and o,p'-DDT were detected in most air but only a few solid matrices. Enantiomeric fractions (EFs) indicated that nonracemic residues of chiral OCs occurred in all the matrices and a wide variation of the EF values was observed in the vegetation. There was significant discrepancy between the EF values of PCB-183 and the racemic values, indicating that stereoselective depletion of PCB-183 was probably associated with the water-air exchange. The EFs values of α-HCH were generally lower than the racemic values but no statistical difference was obtained in all the matrices except lichen, supporting the assumption that water-air exchange may make influence on long-range transport of α-HCH.

Project description:Here, we announce the draft genome sequence of Pseudomonas sp. strain AN3A02, isolated from the rhizosphere of one of the only two species of vascular plants existing in the Antarctic continent, Deschampsia antarctica Desv. This isolate, which inhibited the mycelial growth of Botrytis cinerea in dual culture, has a genome sequence of 6,778,644 bp, with a G+C content of 60.4%. These draft genome sequence data provide insight into the genetics underpinning the antifungal activity of this strain.
