FASN, SCD1 and ANXA9 gene polymorphism as genetic predictors of the fatty acid profile of sheep milk.

ABSTRACT: In this study, single nucleotide polymorphisms (SNPs) in the ANXA9 (annexin 9), FASN (fatty acid synthase) and SCD1 (stearoyl-CoA desaturase 1) genes were analyzed as factors influencing fatty acid profiles in milk from Zošľachtená valaška sheep. SNP in selected genes was identified using polymerase chain reaction (PCR) and restriction fragment length polymorphism (PCR-RFLP). The long-chain fatty acids profile in sheep milk was identified by gas chromatography. Statistical analysis of the SCD1/Cfr13I polymorphism showed that the milk of the homozygous AA animals was characterized by a lower (P < 0.05) share of C4:0, C6:0, C8:0, C10:0, C12:0, C14:0 in comparison to the homozygous CC sheep. The milk of heterozygous sheep was characterized by a higher (P < 0.05) proportion of C13:0 acid compared to the milk of sheep with the homozygous AA type. A higher (P < 0.05) level of saturated fatty acids (SFA) was found in the milk of CC genotype sheep compared to the AA genotype. Our results lead to the conclusion that the greatest changes were observed for the SCD1/Cfr13I polymorphism and the least significant ones for FASN/AciI. Moreover, it is the first evidence that milk from sheep with SCD1/Cfr13I polymorphism and the homozygous AA genotype showed the most desirable fatty acids profile.

Project description:The objectives of this study were as follows: 1) to establish whether feeding a source of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) to ewes during late gestation changes the fatty acid profile of colostrum, milk, ewe adipose tissue, and plasma and subsequently lamb plasma and red blood cells (RBC), and 2) to investigate the effects of EPA and DHA on mRNA expression in ewe adipose tissue. Eighty-four gestating ewes (28 pens, three per pen) were blocked by lambing day and assigned to a diet with an addition of fat at 0.39% of the DM during the last 50 d of gestation using Ca salts of a palm fatty acid distillate (PFAD) high in palmitic and oleic acids or EPA + DHA. Blood samples were taken from ewes on days 20, 1 (parturition), and 30 and from lambs on days 1 and 30 for plasma fatty acid analysis. Fatty analysis of lamb RBC was performed on day 1. Colostrum samples were taken at lambing and milk samples on day 30 for fatty acid analysis. Subcutaneous adipose tissue biopsies were taken from one ewe per pen on day 20 for fatty acid analysis and gene expression analysis of 27 genes. Treatment × day interactions (P < 0.10) were observed for several isomers of C18:1, with concentrations that were greater in plasma of EPA + DHA ewes on day 20, but were not different on day 1 or 30. Plasma concentrations of EPA tended to be greater (P = 0.07), whereas DHA was greater (P < 0.001) in EPA + DHA ewes compared with PFAD ewes. There was no difference in EPA or DHA in adipose tissue with EPA + DHA vs. PFAD supplementation (P > 0.10). Concentrations of fatty acids with 6 to 10 carbons were significantly increased (P < 0.05) in colostrum and milk of EPA + DHA ewes. There was a treatment × day interaction with EPA + DHA ewes yielding greater EPA (P = 0.03) and DHA (P = 0.04) concentrations than PFAD in colostrum, but not in milk. Treatment × day interactions (P < 0.05) were observed for several C18:1 isomers with concentrations that were greater in EPA + DHA ewe colostrum, but were not different between treatments in milk. In lamb plasma and RBC, EPA and DHA were not different between treatments (P > 0.10). The expression of fatty acid synthase and leptin was significantly increased (P < 0.05), whereas the expression of diacylglycerol acyltransferase 2 tended to be increased (P = 0.08) by supplementation of EPA + DHA vs. PFAD. These results suggest that supplementation with EPA and DHA to ewes during late gestation alters the fatty acid profile of plasma, colostrum, and milk and may increase lipogenesis.

Project description:Simple Summary Colostrum is of interest to the scientific community because of its nutritional and therapeutic capabilities. The aims of this study were to characterize the macro and micro composition of colostrum from Sarda dairy sheep and to compare it with the composition of the mature milk of the same breed. The results of this survey showed a large variation in the immunoglobulin concentration in colostrum, which could affect the acquisition of passive immunity by lambs. The strong correlation between immunoglobulin G and the total protein content suggests that this can be used to estimate the immunoglobulin content in sheep colostrum. The concentration of essential minerals is higher in colostrum than in milk as a result of mineral salt block supplementation at the end of gestation. Colostrum has a significantly different fatty acid profile than milk, and this is due to the specific needs of newborn lambs. Abstract Colostrum is a major source of immunity in ruminants. It allows the transfer of antibodies from the mother to the fetus, and it is the exclusive source of nutrients for the newborn. The objectives of this study were (i) to characterize the macro and the micro composition of colostrum; (ii) to analyze the antioxidant capacity, fatty acid profile, and essential and toxic mineral content of colostrum; and (iii) to compare FA profiles and the amount of trace elements between colostrum and mature milk. For these purposes, samples of colostrum and milk were collected from a representative sample of animals from eight sheep dairy farms in the north of Sardinia (Italy). Fat, proteins, and seven essential and toxic minerals were measured in all samples of colostrum and milk. Furthermore, the FA profile was also measured in both matrices, while total antioxidant capacity was measured only in colostrum samples. The average amounts of fat and protein (TP) concentration in colostrum were 7.8% and 16%, respectively. Additionally, an average amount of 40 ± 20 g dm−3 was found for immunoglobulin G (IgG). As regards the antioxidant capacity of colostrum, a large variation was observed between samples from different farms for test 2, 2′-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS), which was 30 ± 10% (mean ± standard deviation). High levels of selenium (Se), zinc (Zn), and copper (Cu) were found in colostrum (200 µg kg−1, 25,000 µg kg−1, and 1200 µg kg−1, respectively). A strong positive correlation between TP and IgG was observed (r = 91%). In colostrum, the amount of IgG is positively correlated with Se and Zn, as they are essential minerals to the immune system. The FA profile demonstrated higher levels of medium and long chain fatty acids in colostrum than in mature milk, and this is mainly true for arachidonic acid (ARA), ecosapentaenoic acid (EPA), docosapentaenoic acid (DPA), and docosahexaenoic acid (DHA). This study provided new information on the quality of colostrum in Sarda dairy sheep and showed the different composition of fatty acids between colostrum and mature milk.
