Stimulus-specific plasticity of macaque V1 spike rates and gamma.

ABSTRACT: When a visual stimulus is repeated, average neuronal responses typically decrease, yet they might maintain or even increase their impact through increased synchronization. Previous work has found that many repetitions of a grating lead to increasing gamma-band synchronization. Here, we show in awake macaque area V1 that both repetition-related reductions in firing rate and increases in gamma are specific to the repeated stimulus. These effects show some persistence on the timescale of minutes. Gamma increases are specific to the presented stimulus location. Further, repetition effects on gamma and on firing rates generalize to images of natural objects. These findings support the notion that gamma-band synchronization subserves the adaptive processing of repeated stimulus encounters.

Project description:Under natural conditions, animals repeatedly encounter the same visual scenes, objects or patterns repeatedly. These repetitions constitute statistical regularities, which the brain captures in an internal model through learning. A signature of such learning in primate visual areas V1 and V4 is the gradual strengthening of gamma synchronization. We used a V1-V4 Dynamic Causal Model (DCM) to explain visually induced responses in early and late epochs from a sequence of several hundred grating presentations. The DCM reproduced the empirical increase in local and inter-areal gamma synchronization, revealing specific intrinsic connectivity effects that could explain the phenomenon. In a sensitivity analysis, the isolated modulation of several connection strengths induced increased gamma. Comparison of alternative models showed that empirical gamma increases are better explained by (1) repetition effects in both V1 and V4 intrinsic connectivity (alone or together with extrinsic) than in extrinsic connectivity alone, and (2) repetition effects on V1 and V4 population input rather than output gain. The best input gain model included effects in V1 granular and superficial excitatory populations and in V4 granular and deep excitatory populations. Our findings are consistent with gamma reflecting bottom-up signal precision, which increases with repetition and, therefore, with predictability and learning.

Attention reduces stimulus-driven gamma frequency oscillations and spike field coherence in V1.

Project description:Rhythmic activity of neuronal ensembles has been proposed to play an important role in cognitive functions such as attention, perception, and memory. Here we investigate whether rhythmic activity in V1 of the macaque monkey (macaca mulatta) is affected by top-down visual attention. We measured the local field potential (LFP) and V1 spiking activity while monkeys performed an attention-demanding detection task. We show that gamma oscillations were strongly modulated by the stimulus and by attention. Stimuli that engaged inhibitory mechanisms induced the largest gamma LFP oscillations and the largest spike field coherence. Directing attention toward a visual stimulus at the receptive field of the recorded neurons decreased LFP gamma power and gamma spike field coherence. This decrease could reflect an attention-mediated reduction of surround inhibition. Changes in synchrony in V1 would thus be a byproduct of reduced inhibitory drive, rather than a mechanism that directly aids perceptual processing.

A quantitative theory of gamma synchronization in macaque V1.

Project description:Gamma-band synchronization coordinates brief periods of excitability in oscillating neuronal populations to optimize information transmission during sensation and cognition. Commonly, a stable, shared frequency over time is considered a condition for functional neural synchronization. Here, we demonstrate the opposite: instantaneous frequency modulations are critical to regulate phase relations and synchronization. In monkey visual area V1, nearby local populations driven by different visual stimulation showed different gamma frequencies. When similar enough, these frequencies continually attracted and repulsed each other, which enabled preferred phase relations to be maintained in periods of minimized frequency difference. Crucially, the precise dynamics of frequencies and phases across a wide range of stimulus conditions was predicted from a physics theory that describes how weakly coupled oscillators influence each other's phase relations. Hence, the fundamental mathematical principle of synchronization through instantaneous frequency modulations applies to gamma in V1 and is likely generalizable to other brain regions and rhythms.

Stimulus dependence of directed information exchange between cortical layers in macaque V1.

Project description:Perception and cognition require the integration of feedforward sensory information with feedback signals. Using different sized stimuli, we isolate spectral signatures of feedforward and feedback signals, and their effect on communication between layers in primary visual cortex of male macaque monkeys. Small stimuli elicited gamma frequency oscillations predominantly in the superficial layers. These Granger-causally originated in upper layer 4 and lower supragranular layers. Unexpectedly, large stimuli generated strong narrow band gamma oscillatory activity across cortical layers. They Granger-causally arose in layer 5, were conveyed through layer six to superficial layers, and violated existing models of feedback spectral signatures. Equally surprising, with large stimuli, alpha band oscillatory activity arose predominantly in granular and supragranular layers and communicated in a feedforward direction. Thus, oscillations in specific frequency bands are dynamically modulated to serve feedback and feedforward communication and are not restricted to specific cortical layers in V1.

Stimulus-specific plasticity in human visual gamma-band activity and functional connectivity.

Project description:Under natural conditions, the visual system often sees a given input repeatedly. This provides an opportunity to optimize processing of the repeated stimuli. Stimulus repetition has been shown to strongly modulate neuronal-gamma band synchronization, yet crucial questions remained open. Here we used magnetoencephalography in 30 human subjects and find that gamma decreases across ≈10 repetitions and then increases across further repetitions, revealing plastic changes of the activated neuronal circuits. Crucially, increases induced by one stimulus did not affect responses to other stimuli, demonstrating stimulus specificity. Changes partially persisted when the inducing stimulus was repeated after 25 minutes of intervening stimuli. They were strongest in early visual cortex and increased interareal feedforward influences. Our results suggest that early visual cortex gamma synchronization enables adaptive neuronal processing of recurring stimuli. These and previously reported changes might be due to an interaction of oscillatory dynamics with established synaptic plasticity mechanisms.

Stimulus-specific competitive selection in macaque extrastriate visual area V4.

Project description:Macaque visual area V4 has been implicated in the selective processing of stimuli. Prior studies of selection in area V4 have used spatially separate stimuli, thus confounding selection of retinotopic location with selection of the stimulus at that location. We asked whether V4 neurons can selectively respond to one of two differently colored stimuli even when they are spatially superimposed. We find that delaying one of the two stimuli leads to selective processing of the delayed stimulus by area V4 neurons. This selective processing persists when the stimuli move together across the visual field, thereby successively activating different populations of neurons. We also find that this effect is not a spatially global form of feature-based selection. We conclude that selective processing in area V4 is neither exclusively spatial nor feature-based and may thus be surface- or object-based.

Gain modulation by nicotine in macaque v1.

Project description:Acetylcholine is a ubiquitous cortical neuromodulator implicated in cognition. In order to understand the potential for acetylcholine to play a role in visual attention, we studied nicotinic acetylcholine receptor (nAChR) localization and function in area V1 of the macaque. We found nAChRs presynaptically at thalamic synapses onto excitatory, but not inhibitory, neurons in the primary thalamorecipient layer 4c. Furthermore, consistent with the release enhancement suggested by this localization, we discovered that nicotine increases responsiveness and lowers contrast threshold in layer 4c neurons. We also found that nAChRs are expressed by GABAergic interneurons in V1 but rarely by pyramidal neurons, and that nicotine suppresses visual responses outside layer 4c. All sensory systems incorporate gain control mechanisms, or processes which dynamically alter input/output relationships. We demonstrate that at the site of thalamic input to visual cortex, the effect of this nAChR-mediated gain is an enhancement of the detection of visual stimuli.

Electrophysiological assessment of auditory stimulus-specific plasticity in schizophrenia.

Project description:BackgroundDisrupted neuroplasticity may be an important aspect of the neural basis of schizophrenia. We used event-related brain potentials (ERPs) to assay neuroplasticity after auditory conditioning in chronic schizophrenia patients (SZ) and matched healthy control subjects (HC).MethodsSubjects (15 HC, 14 SZ) performed an auditory oddball task during electroencephalogram recording before and after auditory tetanic stimulation (Pre/Post Blocks). Each oddball block consisted of 1000-Hz and 1500-Hz standards and 400-Hz targets. During tetanic conditioning, 1000-Hz tones were presented at 11 Hz for 2.4 min. We analyzed the standard trials, comparing the ERPs evoked by the tetanized stimuli (1000 Hz tones: TS+) and untetanized stimuli (1500 Hz tones: TS-) in the Post Blocks with ERPs from the Pre Blocks (averaged into Baseline ERPs).ResultsIn Post Block 1 in HC, TS+ tones evoked a negative shift (60-350 msec) at right temporal electrodes relative to Baseline. No pre-/post-tetanus effects were found in SZ. In Post Block 2 in HC, TS+ tones evoked a positive shift (200-300 msec) at bilateral frontal electrodes. In SZ, TS+ tones evoked a positive shift (100-400 msec) at right frontotemporal electrodes. No pre-/post-tetanus effects were found in either subject group for the TS- tones. The right temporal Post Block 1 and 2 effects were correlated in SZ, suggesting a trade-off in the expression of these effects.ConclusionsThese results suggest that stimulus-specific auditory neuroplasticity is abnormal in schizophrenia. The electrophysiologic assessment of stimulus-specific plasticity may yield novel targets for drug treatment in schizophrenia.

Coding of chromatic spatial contrast by macaque V1 neurons.

Project description:Color perception relies on comparisons between adjacent lights, but how the brain performs these comparisons is poorly understood. To elucidate the underlying mechanisms, we recorded spiking responses of individual V1 neurons in macaque monkeys to pairs of stimuli within the classical receptive field (RF). We estimated the spatial-chromatic RF of each neuron and then presented customized colored edges using a closed-loop technique. We found that many double-opponent (DO) cells, which have spatially and chromatically opponent RFs, responded to chromatic contrast as a weighted sum, akin to how other V1 neurons responded to luminance contrast. Yet other neurons integrated chromatic signals nonlinearly, confirming that linear signal integration is not an obligate property of V1 neurons. The functional similarity of cone-opponent DO cells and cone non-opponent simple cells suggests that these two groups may share a common underlying circuitry, promotes the construction of image-computable models for full-color image representation, and sheds new light on V1 complex cells.

Synapse- and stimulus-specific local translation during long-term neuronal plasticity.

Project description:Long-term memory and synaptic plasticity require changes in gene expression and yet can occur in a synapse-specific manner. Messenger RNA localization and regulated translation at synapses are thus critical for establishing synapse specificity. Using live-cell microscopy of photoconvertible fluorescent protein translational reporters, we directly visualized local translation at synapses during long-term facilitation of Aplysia sensory-motor synapses. Translation of the reporter required multiple applications of serotonin, was spatially restricted to stimulated synapses, was transcript- and stimulus-specific, and occurred during long-term facilitation but not during long-term depression of sensory-motor synapses. Translational regulation only occurred in the presence of a chemical synapse and required calcium signaling in the postsynaptic motor neuron. Thus, highly regulated local translation occurs at synapses during long-term plasticity and requires trans-synaptic signals.
