Indigenous microbial communities in heavy oil show a threshold response to salinity.

ABSTRACT: Microbial degradation influences the quality of oil resources. The environmental factors that shape the composition of oil microbial communities are largely unknown because most samples from oil fields are impacted by anthropogenic oil production, perturbing the native ecosystem with exogenous fluids and microorganisms. We investigated the relationship between formation water geochemistry and microbial community composition in undisturbed oil samples. We isolated 43 microliter-sized water droplets naturally enclosed in the heavy oil of the Pitch Lake, Trinidad and Tobago. The water chemistry and microbial community composition within the same water droplet were determined by ion chromatography and 16S rRNA gene amplicon sequencing, respectively. The results revealed a high variability in ion concentrations and community composition between water droplets. Microbial community composition was mostly affected by the chloride concentration, which ranged from freshwater to brackish-sea water. Remarkably, microbial communities did not respond gradually to increasing chloride concentration but showed a sudden change to less diverse and uneven communities when exceeding a chloride concentration of 57.3 mM. The results reveal a threshold-regulated response of microbial communities to salinity, offering new insights into the microbial ecology of oil reservoirs.

Project description:Salinity stress is one of the critical environmental drivers of soil organic matter (SOM) decomposition in coastal ecosystems. Although the temperature sensitivity (Q10) of SOM decomposition has been widely applied in Earth system models to forecast carbon processes, the impact of salinity on SOM decomposition by restructuring microbial communities remains uncovered. Here, we conducted a microcosm experiment with soils collected from the coastal salt marsh in the Yellow River Estuary, which is subjected to strong dynamics of salinity due to both tidal flooding and drainage. By setting a gradient of salt solutions, soil salinity was adjusted to simulate salinity stress and soil carbon emission (CO2) rate was measured over the period. Results showed that as salinity increased, the estimated decomposition constants based on first-order kinetics gradually decreased at different temperatures. Below the 20‰ salinity treatments, which doubled the soil salinity, Q10 increased with increasing salinity; but higher salinity constrained the temperature-related response of SOM decomposition by inhibiting microbial growth and carbon metabolisms. Soil bacteria were more sensitive to salinity stress than fungi, which can be inferred from the response of microbial beta-diversity to changing salinity. Among them, the phylotypes assigned to Gammaproteobacteria and Bacilli showed higher salt tolerance, whereas taxa affiliated with Alphaproteobacteria and Bacteroidota were more easily inhibited by the salinity stress. Several fungal taxa belonging to Ascomycota had higher adaptability to the stress. As the substrate was consumed with the incubation, bacterial competition intensified, but the fungal co-occurrence pattern changed weakly during decomposition. Collectively, these findings revealed the threshold effect of salinity on SOM decomposition in coastal salt marshes and emphasized that salt stress plays a key role in carbon sequestration by regulating microbial keystone taxa, metabolisms, and interactions.

Project description:Soil salinization and heavy metal (HM) contamination are major challenges facing agricultural systems worldwide. Determining how soil microbial communities respond to these stress factors and identifying individual phylotypes with potential to tolerate these conditions while promoting plant growth could help prevent negative impacts on crop productivity. This study used amplicon sequencing and several bioinformatic programs to characterize differences in the composition and potential functional capabilities of soil bacterial, fungal, and archaeal communities in five agricultural fields that varied in salinity and HM concentrations within the Indus basin region of Pakistan. The composition of bacteria with the potential to fix atmospheric nitrogen (N) and produce the enzyme 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase were also determined. Microbial communities were dominated by: Euryarchaeota (archaea), Actinobacteria, Proteobacteria, Planctomycetota, Firimicutes, Patescibacteria and Acidobacteria (bacteria), and Ascomycota (fungi), and all soils contained phylotypes capable of N-fixation and ACC-deaminase production. Salinity influenced bacterial, but not archaeal or fungal communities. Both salinity and HM altered the relative abundance of many phylotypes that could potentially promote or harm plant growth. These stress factors also appeared to influence the potential functional capabilities of the microbial communities, especially in their capacity to cycle phosphorous, produce siderophores, and act as symbiotrophs or pathotrophs. Results of this study confirm that farms in this region are at risk due to salinization and excessive levels of some toxic heavy metals, which could negatively impact crop and human health. Changes in soil microbial communities and their potential functional capabilities are also likely to affect several critical agroecosystem services related to nutrient cycling, pathogen suppression, and plant stress tolerance. Many potentially beneficial phylotypes were identified that appear to be salt and HM tolerant and could possibly be exploited to promote these services within this agroecosystem. Future efforts to isolate these phylotypes and determine whether they can indeed promote plant growth and/or carry out other important soil processes are recommended. At the same time, identifying ways to promote the abundance of these unique phylotypes either through modifying soil and crop management practices, or developing and applying them as inoculants, would be helpful for improving crop productivity in this region.
