Measurement of progesterone in sheep using a commercial ELISA kit for human plasma.

ABSTRACT: Determination of serum or plasma progesterone (P4) concentrations is important to recognize pregnant and non-pregnant ewes, and also to predict the number of carried lambs. The 2 most common methodologies for the detection of plasma P4 are radioimmunoassay (RIA) and enzyme immunoassay (EIA). RIA is very expensive, and not all laboratories are equipped to perform this test; EIA is commercially available for human use, but only a few companies produce species-specific kits, which are expensive. We verified for ovine plasma a less expensive and easily available ELISA kit (DiaMetra) designed to quantify P4 in humans. Pools of ovine and human plasma were used to compare repeatability, accuracy, sensitivity, and stability of P4 measured by the DiaMetra kit. Repeatability data were within 15%, and accuracy values were ~90% for both plasma matrices. Stability data showed a loss of <20% for freeze-thaw and <30% for 30-d storage. All parameters were acceptable under international guidelines for method validation. The human ELISA kit was used successfully to quantify plasma P4 in 26 ewes during pregnancy until delivery. P4 concentrations were also correlated with the number of carried lambs.

Project description:BackgroundToxoplasmosis is one of the most common food borne zoonoses worldwide, and can be a serious life-threatening disease in the congenitally infected fetus and in immunosuppressed patients. Among food animals, sheep along with goats and pigs possess the highest incidence of T. gondii cysts in meat, and play a major role as a source of human infection.MethodsIn this study, a new commercial ELISA kit (PrioCHECK Toxoplasma Ab SR, Prionics Schlieren-Zurich, Switzerland) for the detection of anti-T. gondii antibodies in serum, plasma and meat juice of sheep, was evaluated by comparing it with the indirect fluorescent antibody test (IFAT), indirect haemagglutination test (IHA) and real-time PCR, on samples from experimentally inoculated and naturally exposed sheep.ResultsThe commercial ELISA detected the infection status in 50% and 100% of sheep orally inoculated with 10,000 T. gondii oocysts (n = 6), from two or three weeks post infection (wpi), respectively, both on serum and plasma samples. Meat juice from all experimentally inoculated sheep collected at slaughter (12 wpi) showed positive ELISA values. In naturally exposed sheep (n = 396), the ELISA showed a very good agreement with IFAT (kappa = 0.91-1.0) and IHA (kappa = 0.96-1.0) performed on serum; and a positive correlation was observed between ELISA values and IFAT titers. By a Receiver Operating Characteristics (ROC) curve analysis, the commercial ELISA had relative sensitivities between 93.33% and 100%, and relative specificities between 96.87% and 100% respect to IFAT and IHA, depending on the considered cut-off value and animal groups tested. Furthermore, the ELISA correctly recognized all animals reacting positive in real-time PCR. The ELISA results on meat juice agreed with those on serum samples in all experimentally inoculated animals, and in 94 out of 96 (97.9%) naturally exposed sheep, when meat juice was tested at a 1:10 dilution.ConclusionThe commercial ELISA kit evaluated in this study could represent a valuable tool to improve the surveillance and reporting system for T. gondii in sheep populations at the farm level or for diagnosis at the slaughterhouse, contributing to the control of this widespread zoonosis.

Project description:The financial impact of early embryonic loss in Australia may be as high as $137 million AUD/year. Embryos may be lost due to environmental conditions, or maternal factors such as nutrition or progesterone (P4) profiles. However, studies on the supplementation of P4 during early pregnancy have returned contradictory results, partly as a reliable method of detecting embryos in the early stages of gestation (<day 20) has yet be established. As such, Merino ewes (n = 62) were either not supplemented (control) or were given exogenous P4 at the time of insemination (day 0) or 3 days later (day 3). Transrectal ultrasound (TRUS) was performed on day 10, 12, 14, 17, 19 and 29 following laparoscopic artificial insemination. Transcutaneous ultrasound (TCUS) was performed on day 54 to confirm pregnancy and peripheral blood was collected for hormone analysis on day 19 to compare the accuracy of all three pregnancy diagnosis methods. Data were then analysed in developmental periods. The percentage of ewes detected as pregnant by TRUS during pre-, peri- and post implantation was 66% (41/62; day 12 and 14), 61% (38/62; day 17 and 19) and 58% (36/62; day 29), respectively. TCUS during established gestation recorded a pregnancy rate of 60% (37/62). The sensitivity of TRUS to correctly diagnose ewes as pregnant during pre-, peri- and post implantation was 68% (25/37), 89% (33/37) and 100% (36/36), respectively, while the sensitivity to correctly identify multiples was 49% (16/33), 60% (21/35) and 97% (34/35), respectively (P<0.05). The majority of embryonic loss occurred between pre- and peri- implantation (0.9±0.15 per ewe; P<0.001). No further loss was recorded after this point. Ewes that were given P4 at day 0 had significantly higher embryonic loss (77%) compared to the control (52%) and day 3-ewes (56%; P<0.05). These results show TRUS is a viable tool for investigating early embryonic loss and that the variability noted in previous P4 supplementation studies may be due to variation in time and length of treatment.
