Development of an Autochthonous Microbial Consortium for Enhanced Bioremediation of PAH-Contaminated Soil.

ABSTRACT: The main objectives of this study were to isolate bacteria from soil chronically contaminated with polycyclic aromatic hydrocarbons (PAHs), develop an autochthonous microbial consortium, and evaluate its ability to degrade PAHs in their native contaminated soil. Strains with the best bioremediation potential were selected during the multi-stage isolation process. Moreover, to choose bacteria with the highest bioremediation potential, the presence of PAH-degrading genes (pahE) was confirmed and the following tests were performed: tolerance to heavy metals, antagonistic behavior, phytotoxicity, and antimicrobial susceptibility. In vitro degradation of hydrocarbons led to the reduction of the total PAH content by 93.5% after the first day of incubation and by 99.22% after the eighth day. Bioremediation experiment conducted in situ in the contaminated area resulted in the average reduction of the total PAH concentration by 33.3% after 5 months and by over 72% after 13 months, compared to the concentration recorded before the intervention. Therefore, this study implicates that the development of an autochthonous microbial consortium isolated from long-term PAH-contaminated soil has the potential to enhance the bioremediation process.

Project description:The formation of more polar and toxic polycyclic aromatic hydrocarbon (PAH) transformation products is one of the concerns associated with the bioremediation of PAH-contaminated soils. Soil contaminated with coal tar (prebioremediation) from a former manufactured gas plant (MGP) site was treated in a laboratory scale bioreactor (postbioremediation) and extracted using pressurized liquid extraction. The soil extracts were fractionated, based on polarity, and analyzed for 88 PAHs (unsubstituted, oxygenated, nitrated, and heterocyclic PAHs). The PAH concentrations in the soil tested, postbioremediation, were lower than their regulatory maximum allowable concentrations (MACs), with the exception of the higher molecular weight PAHs (BaA, BkF, BbF, BaP, and IcdP), most of which did not undergo significant biodegradation. The soil extract fractions were tested for genotoxicity using the DT40 chicken lymphocyte bioassay and developmental toxicity using the embryonic zebrafish (Danio rerio) bioassay. A statistically significant increase in genotoxicity was measured in the unfractionated soil extract, as well as in four polar soil extract fractions, postbioremediation (p < 0.05). In addition, a statistically significant increase in developmental toxicity was measured in one polar soil extract fraction, postbioremediation (p < 0.05). A series of morphological abnormalities, including peculiar caudal fin malformations and hyperpigmentation in the tail, were measured in several soil extract fractions in embryonic zebrafish, both pre- and postbioremediation. The increased toxicity measured postbioremediation is not likely due to the 88 PAHs measured in this study (including quinones), because most were not present in the toxic polar fractions and/or because their concentrations did not increase postbioremediation. However, the increased toxicity measured postbioremediation is likely due to hydroxylated and carboxylated transformation products of the 3- and 4-ring PAHs (PHE, 1MPHE, 2MPHE, PRY, BaA, and FLA) that were most degraded.

Project description:This research examined the bioremediation of pesticides (Carbofuran and Paraquat) contaminated farmyard soil using compost and Nitrogen, Phosphorus, and Potassium (NPK) fertilizer. Microcosms representing each treatment were set-up in triplicates. Biostimulation was done using two concentrations (0.5 % and 1.0 % w/w) of NPK fertilizer and compost, following pesticides application at recommended rates [Carbofuran (1 g/kg) and Paraquat (5 ml/kg)] and four times the recommended rates. Two control soils were set-up; Abiotic control (sterile farmyard soil + pesticide) and Control (farmyard soil without treatment). Monitoring of the dynamics in microbial community abundance, and pesticide residues during the biostimulation period was done weekly for 28 days, using standard enumeration method, and High Performance Liquid Chromatography (HPLC), respectively. At the end of the monitoring period, considerable reduction in pesticide residues across the treatment set-ups was recorded. In Carbofuran-treated soils, there were no complete, but considerable losses in residual pesticide, however, in most of the Paraquat-treated soils, there were complete losses within 21 days. Lower pesticide residues were recorded in set-ups amended with compost than NPK, across both Carbofuran and Paraquat-treated soils. After pesticides application, decreases in microbial counts were recorded at Day 7 across all the treatments, followed by increases from Day 14-21, then decreases at Day 28. Microbial counts were lower in Carbofuran than in Paraquat-treated soils irrespective of nutrient (compost and NPK) amendments. Bacterial and fungal counts were in the magnitude of 106 and 105 CFU/g soil, respectively. Also, increased counts were recorded for Actinomycetes, Nitrifiers, Phosphate solubilizers across all treatments, and were in magnitude of 103-104 CFU/g soil. Soil microorganisms could breakdown and eliminate large concentrations of Carbofuran and Paraquat in compost-amended soils than in NPK-amended soils. This study suggests that bioremediation of pesticides contaminated soils can be achieved and enhanced by stimulating the indigenous microbial community with requisite nutrients (compost).
