Achnanthidiumgladius sp. nov. (Bacillariophyceae) - a new monoraphid diatom species from Indonesia.

ABSTRACT: A new monoraphid diatom species Achnanthidiumgladius sp. nov. is described from Indonesia. The description is based on molecular data (18SV4), morphological analysis and comparison with similar species. According to molecular data, Achnanthidiumgladius sp. nov. is closely related to Achnanthidiumminutissimum. Morphologically, the new species differs from similar species by the absence of a fascia on raphe valve, cell size, and striae density and pattern. The new species is only known from the type locality in Indonesia. Comparison with close related species is given.

Achnanthidiumbratanense sp. nov. (Bacillariophyceae, Achnanthidiaceae), a new diatom from the Lake Bratan (Bali, Indonesia).

Project description:A new species, Achnanthidiumbratanense, is described from Lake Bratan, located on the island of Bali (Indonesia). The morphology of this species was analyzed with light (LM) and scanning electron microscopy (SEM). A.bratanense is characterized by linear-elliptic to nearly elliptic valves with convex margins and rounded, broadly subcapitate apices. The striae of this species are hardly discernable under LM; they are weakly radiate throughout the valve and composed of one to four large transapically elongated areolae of different length and shape. The most similar taxon to A.bratanense is A.macrocephalum, a species described from Sumatra, another Indonesian island. The differences of A.bratanense from similar taxa are discussed.

Encyonopsis indonesica sp. nov. (Bacillariophyceae, Cymbellales), a new diatom from the ancient lake Matano (Sulawesi, Indonesia).

Project description:A new species, Encyonopsis indonesica, is described from the ancient lake Matano, Sulawesi island, Indonesia. The morphology of this species was studied by means of light and scanning electron microscopy. E. indonesica has a remarkable valve ultrastructure. The valve surface is ornamented with numerous longitudinal siliceous ribs and siliceous verrucae. Valve face delineated from the mantle by a thickened marginal ridge. Raised sterna border the raphe branches. Raphe is distinctly undulate with distal ends hooked strongly to the ventral side. The only similar species to E. indonesica is Amphora dissimilis described from New Caledonia. Comparison of both taxa is given and A. dissimilis is transferred to Encyonopsis. The taxonomic placement of both taxa is evaluated, and the phenomenon of external siliceous ornamentation is discussed.

A new diatom species P. hallegraeffii sp. nov. belonging to the toxic genus Pseudo-nitzschia (Bacillariophyceae) from the East Australian Current.

Project description:A new species belonging to the toxin producing diatom genus Pseudo-nitzschia, P. hallegraeffii sp. nov., is delineated and described from the East Australian Current (EAC). Clonal cultures were established by single cell isolation from phytoplankton net hauls collected as part of a research expedition in the EAC region in 2016 on the RV Investigator. Cultures were assessed for their morphological and genetic characteristics, their sexual compatibility with other Pseudo-nitzschia species, and their ability to produce domoic acid. Light and transmission electron microscopy revealed cells which differed from their closest relatives by their cell width, rows of poroids, girdle band structure and density of band straie. Phylogenetic analyses based on sequencing of nuclear-encoded ribosomal deoxyribonucleic acid (rDNA) regions showed this novel genotype clustered within the P. delicatissima complex, but formed a discrete clade from its closest relatives P. dolorosa, P. simulans, P. micropora and P. delicatissima. Complementary base changes (CBCs) were observed in the secondary structure of the 3' nuclear ribosomal transcribed spacer sequence region (ITS2) between P. hallegraeffii sp. nov. and its closest related taxa, P. simulans and P. dolorosa. Under laboratory conditions, and in the absence of any zooplankton cues, strains of P. hallegraeffii sp. nov. did not produce domoic acid (DA) and were not sexually compatible with any other Pseudo-nitzschia clones tested. A total of 18 Pseudo-nitzschia species, including three confirmed toxigenic species (P. cuspidata, P. multistriata and P. australis) have now been unequivocally confirmed from eastern Australia.

Achnanthidium tinea sp. nov. - a new monoraphid diatom (Bacillariophyceae) species, described on the basis of molecular and morphological approaches.

Project description:A new monoraphid diatom species Achnanthidium tinea Tseplik, Kulikovskiy, Kociolek & Maltsev, sp. nov. is described from Indonesia. The species is described on the basis of molecular and morphological analyses. According to molecular data the new species belongs to the clade that includes strains of Achnanthidium minutissimum, Achnanthidium saprophilum and Achnanthidium digitatum. Morphologically, the new species differs quite significantly from other species of the same genus because of linear-elliptic valves with almost parallel sides and strongly radiate striae and a butterfly-shaped fascia on the raphe valve. The morphology and phylogeny of the new species are discussed, and thoughts on the current state of the taxonomy of the genus Achnanthidium are expressed. Our work shows the importance of using molecular data in diatom systematics and also demonstrates the need to investigate rarely studied regions of our planet.

A Description of Biremis panamae sp. nov., a New Diatom Species from the Marine Littoral, with an Account of the Phylogenetic Position of Biremis D.G. Mann et E.J. Cox (Bacillariophyceae).

Project description:Here we present a formal description of Biremis panamae Barka, Witkowski et Weisenborn sp. nov., which was isolated from the marine littoral environment of the Pacific Ocean coast of Panama. The description is based on morphology (light and electron microscopy) and the rbcL, psbC and SSU sequences of one clone of this species. The new species is included in Biremis due to its morphological features; i.e. two marginal rows of foramina, chambered striae, and girdle composed of numerous punctate copulae. The new species also possesses a striated valve face which is not seen in most known representatives of marine littoral Biremis species. In this study we also present the relationship of Biremis to other taxa using morphology, DNA sequence data and observations of auxosporulation. Our results based on these three sources point to an evolutionary relationship between Biremis, Neidium and Scoliopleura. The unusual silicified incunabular caps present in them are known otherwise only in Muelleria, which is probably related to the Neidiaceae and Scoliotropidaceae. We also discuss the relationship between Biremis and the recently described Labellicula and Olifantiella.

Morphology and phylogeny of Nitzschianandorii sp. nov. (Bacillariophyceae), a new small-celled lanceolate species from a post-mining reservoir.

Project description:Post-mining reservoirs are distinguished by characteristic environmental conditions where specific diatom communities can be observed. Reservoirs created as a part of the reclamation plan after human mining activities are marked by unique chemical and physical water parameters. In the course of research on the diatoms from Bogdałów reservoir, we examined the taxonomic and morphological diversity of Nitzschia taxa from the section Lanceolatae occurring in a post-mining lignite reservoir. Our study describes a new species of Nitzschia from a post-mining reservoir, Nitzschianandorii Olszyński, Zakrzewski & Żelazna-Wieczorek, sp. nov. Morphometry and morphology analyses of new species were performed with light and scanning electron microscopy. Chloroplast morphology analysis was conducted with differential interference contrast microscopy and confocal laser scanning microscopy. Molecular data from SSU 18S, rbcL and psbC sequences were obtained from cultures of this taxon. Differential diagnosis of Nitzschianandorii Olszyński, Zakrzewski & Żelazna-Wieczorek, sp. nov. with co-occurring taxa: N.lacuum and N.alpinobacillum was performed using morphological traits and nMDS analysis of the valves' morphometry.

Three new Scandinavian species of Culicoides (Culicoides): Culicoides boyi sp. nov., Culicoides selandicus sp. nov. and Culicoides kalix sp. nov. (Diptera: Ceratopogonidae).

Project description:In the context of a major monitoring program of Culicoides in Denmark and Sweden due to the appearance of bluetongue disease in 2007-2008, a large number of specimens were collected by light traps and sorted morphologically, with COI barcodes generated for selected specimens.Three species are described as new to science based on both morphological and molecular data: Culicoides (Culicoides) boyi sp. nov. (Denmark: Jutland), C. (C.) selandicus sp. nov. (Denmark: Zealand) and C. (C.) kalix sp. nov. (Sweden: Norrbotten). All are diagnosed morphologically as well as by molecular barcoding. A key to slide-mounted females of all Scandinavian species of Culicoides (Culicoides) is presented.

Diversity in the Globally Distributed Diatom Genus Chaetoceros (Bacillariophyceae): Three New Species from Warm-Temperate Waters.

Project description:Chaetoceros is one of the most species rich, widespread and abundant diatom genera in marine and brackish habitats worldwide. It therefore forms an excellent model for in-depth biodiversity studies, assessing morphological and genetic differentiation among groups of strains. The global Chaetoceros lorenzianus complex presently comprises three species known to science. However, our recent studies have shown that the group includes several previously unknown species. In this article, 50 strains, mainly from high latitudes and from warm-temperate waters, were examined morphologically and genetically and the results compared with those of field studies from elsewhere. The strains clustered into five groups, two of which are formed by C. decipiens Cleve and C. mitra (Bailey) Cleve, respectively. Their species descriptions are emended based on samples collected close to the type localities. The three other groups are formed by new species, C. elegans sp. nov., C. laevisporus sp. nov. and C. mannaii sp. nov. Characters used to distinguish each species are: orientation of setae, shape and size of the apertures, shape, size and density of the poroids on the setae and, at least in some species, characters of the resting spores. Our aim is to cover the global species diversity in this complex, as correct species delineation is the basis for exploring biodiversity, distribution of organisms, interactions in the food web and effects of environmental changes.

Ceratocystis larium sp. nov., a new species from Styrax benzoin wounds associated with incense harvesting in Indonesia.

Project description:Styrax benzoin trees, native to the island Sumatra, Indonesia are wounded to produce resin that is collected and burned as incense. These wounds on trees commonly develop into expanding cankers that lead to tree death. The aim of this study was to consider whether Ophiostomatoid fungi, typically associated with wounds on trees might be associated with resin harvesting on S. benzoin. Samples were collected from the edges of artificially induced wounds, and particularly where cankers and staining of the vascular tissue was evident. Tissue samples were incubated in moist chambers and carrot baiting was also used to detect the presence of Ceratocystis spp. Fruiting structures with morphology typical of species in the C. fimbriata s.l. species complex and species in the anamorph genus Thielaviopsis were found, on both the incubated wood and the carrot baits. DNA sequences were generated for the Internal Transcribed Spacer regions 1 and 2 including the 5.8S rRNA gene, part of the beta-tubulin and the Transcription Elongation Factor 1-alpha gene regions. These data were compared with those of other species in the C. fimbriata s.l. species complex and Thielaviopsis using phylogenetic analysis. Morphology of the isolates in culture as well as phylogenetic inference showed that the Thielaviopsis sp. present on the wounds was T. basicola. The Ceratocystis sp. from S. benzoin represents a new taxon in the C. fimbriata s.l. complex described here as C. larium sp. nov.
