Complete mitochondrial genome sequence of Abraximorpha davidii (Lepidoptera: Hesperiidae: Tagiadinae).

ABSTRACT: Taxonomic status and phylogenetic position of some skippers within Hesperiidae remains a controversial issue, here, we sequenced and analyzed the complete mitochondrial genome (mitogenome) of Abraximorpha davidii, one of species in Hesperiidae. This mitogenome is 15,469 bp long and encodes 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), and two ribosomal RNA unit genes (rRNAs). The overall base composition of the mitogenome is A 40.2%, T 41.4%, C 11.2%, and G 7.2%, with a high A + T content of 81.6%. Except for cox1 starting with CGA, all other PCGs start with the standard ATN codons (seven ATG and five ATT). Most of the PCGs terminate with the stop codon TAA, whereas cox1, cox2, nad5, and nad4 end with the incomplete codon T-. Phylogenetic analysis showed that A. davidii is closely related to Daimio tethys and Tagiades vajuna, then this clade clusters Ctenoptilum vasava and Celaenorrhinus maculosa.

The 15327 bp mitogenome covers 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), 2 ribosomal RNA genes (rRNAs), and an A+T rich region. Its gene order is typical for mitogenomes of Lepidoptera. Phylogenetic analysis places Burara striata as a sister to Hasora, and Choaspes as a sister to both of these genera.

Complete mitochondrial genome of the Small-Branded Swift: Pelopidas mathias (Lepidoptera, Hesperiidae).

Project description:We report the complete mitochondrial genome of the Small-Branded Swift: Pelopidas mathias, which is an important pest of rice. The total length of the circular double-stranded mitogenome is 15,524 bp, containing 13 protein-coding genes (PCGs), 22 transfer RNAs (tRNAs), 2 ribosomal RNAs (rRNAs) and a non-coding AT-rich region with the nucleotide base composition of 40.07% A, 40.83% T, 11.59% C, and 7.51% G, showing a relatively strong AT bias. The gene order and organization are consistent with typical Lepidoptera species. This work will provide molecular data support for the study of the phylogeny and evolution in the family Hesperiidae.

Complete mitochondrial genome of the silver stripped skipper, Leptalina unicolor (Lepidoptera: Hesperiidae).

Project description:The silver stripped skipper, Leptalina unicolor Bremer and Grey, 1853 (Lepidoptera: Hesperiidae), is listed as endangered insect in South Korea. We sequenced the whole genome (15,854 bp) of L. unicolor species using Next-Generation Sequencing method and the subsequent gap-filling method. This genome included a set of typical genes and one major non-coding A + T-rich region, with an arrangement identical to that observed in most lepidopteran genomes. Twelve protein-coding genes (PCGs) had the typical ATN start codon, whereas COI had the atypical CGA codon that is frequently found in the start region of the lepidopteran COI. The 757-bp long A + T-rich region was the second largest among completely sequenced Hesperiidae, which ranged from 234 to 793 bp. Phylogenetic reconstruction was performed by maximum-likelihood method using the concatenated sequences of 13 PCGs and two rRNAs of available species of Hesperiidae, including that of L. unicolor (a total of 28 species). The resulting phylogeny provided strong support for monophyletic Heteropterinae in which L. unicolor belongs, with the highest nodal support and a sister relationship between current L. unicolor and co-subfamilial species Carterocephalus silvicola with a bootstrap value of 91%.

The Complete Mitochondrial Genome of Ctenoptilum vasava (Lepidoptera: Hesperiidae: Pyrginae) and Its Phylogenetic Implication.

Project description:We here report the first complete mitochondrial (mt) genome of a skipper, Ctenoptilum vasava Moore, 1865 (Lepidoptera: Hesperiidae: Pyrginae). The mt genome of the skipper is a circular molecule of 15,468 bp, containing 2 ribosomal RNA genes, 24 putative transfer RNA (tRNA), genes including an extra copy of trnS (AGN) and a tRNA-like insertion trnL (UUR), 13 protein-coding genes and an AT-rich region. All protein-coding genes (PCGs) are initiated by ATN codons and terminated by the typical stop codon TAA or TAG, except for COII which ends with a single T. The intergenic spacer sequence between trnS (AGN) and ND1 genes also contains the ATACTAA motif. The AT-rich region of 429 bp is comprised of nonrepetitive sequences, including the motif ATAGA followed by an 19 bp poly-T stretch, a microsatellite-like (AT)(3) (TA)(9) element next to the ATTTA motif, an 11 bp poly-A adjacent to tRNAs. Phylogenetic analyses (ML and BI methods) showed that Papilionoidea is not a natural group, and Hesperioidea is placed within the Papilionoidea as a sister to ((Pieridae + Lycaenidae) + Nymphalidae) while Papilionoidae is paraphyletic to Hesperioidea. This result is remarkably different from the traditional view where Papilionoidea and Hesperioidea are considered as two distinct superfamilies.

Complete mitochondrial genome of banana skipper Erionota torus Evans (Lepidoptera: Hesperiidae) and phylogenetic analysis.

Project description:Erionota torus (Evans, 1941) is a banana pest and is mainly distributed in Southeast Asia and the Pacific regions. The complete mitogenome of E. torus (GenBank accession number MW586888) is 15,987 bp in size, including 13 protein-coding genes, 22 transfer RNAs, 2 ribosomal RNAs genes, and a noncoding A + T-rich region. The A + T-rich region is located between 12S rRNA and tRNAMet . The base composition of the whole E. torus mitogenome is 39.68% for A, 7.30% for G, 41.55% for T, and 11.47% for C, with a high AT content of 81.23%. The phylogeny analysis indicated that E. torus had a close relationship with Notocrypta curvifascia. The present data could contribute to the further detailed phylogeographic analysis and provide a comprehensive control strategy for this banana pest.

The complete mitochondrial genomes of two skipper genera (Lepidoptera: Hesperiidae) and their associated phylogenetic analysis.

Project description:The systematic positions of two hesperiid genera, Apostictopterus and Barca (Lepidoptera: Hesperiidae), remain ambiguous. We sequenced and annotated the two mitogenomes of Apostictopterus fuliginosus and Barca bicolor and inferred the phylogenetic positions of the two genera within the Hesperiidae based on the available mitogenomes. The lengths of the two circular mitogenomes of A. fuliginosus and B. bicolor are 15,417 and 15,574 base pairs (bp), respectively. These two mitogenomes show similar AT skew, GC skew, codon usage and nucleotide bias of AT: the GC skew of the two species is negative, and the AT skew of A. fuliginosus is negative, while the AT skew of B. bicolor is slightly positive. The largest intergenic spacer is located at the same position between trnQ and ND2 in A. fuliginosus (73 bp) and B. bicolor (72 bp). Thirteen protein-coding genes (PCGs) start with ATN codons except for COI, which starts with CGA. The control regions of both mitogenomes possess a long tandem repeat, which is 30 bp long in A. fuliginosus, and 18 bp in B. bicolor. Bayesian inference and maximum likelihood methods were employed to infer the phylogenetic relationships, which suggested that A. fuliginosus and B. bicolor belong in the subfamily Hesperiinae.

The complete mitochondrial genome sequence of Dodona eugenes (Lepidoptera: Riodinidae).

Project description:The complete mitochondrial genome (mitogenome) sequence of Dodona eugenes (Lepidoptera: Riodinidae) was determined and analyzed. The mitogenome is 15,680 bp in length with consisting of 13 protein-coding genes (PCGs), 22 transfer RNA (tRNA) genes, two ribosomal RNA genes (rrnL and rrnS), and one AT-rich region. The gene content, orientation, and order are identical to that of the majority of other lepidopteran insects. The D. eugenes mitogenome includes a cox1 gene with an atypical CGA(R) start codon and three genes (cox1, nad5, and nad4) exhibiting incomplete stop codons. All tRNAs have a typical secondary cloverleaf structure, except for trnS1 which lacks the dihydrouridine (DHU) arm. The 825-bp long AT-rich region is the longest among sequenced riodinids, which range from 349 to 423 bp. The conclusion of phylogenetic analysis highly supported the monophyly of Riodinidae, which is standing as the sister of the family Lycaenidae.

Complete mitochondrial genomes of three skippers in the tribe Aeromachini (Lepidoptera: Hesperiidae: Hesperiinae) and their phylogenetic implications.

Project description:The mitochondrial genome is now widely used in the study of phylogenetics and molecular evolution due to its maternal inheritance, fast evolutionary rate, and highly conserved gene content. To explore the phylogenetic relationships of the tribe Aeromachini within the subfamily Hesperiinae at the mitochondrial genomic level, we sequenced and annotated the complete mitogenomes of 3 skippers: Ampittia virgata, Halpe nephele, and Onryza maga (new mitogenomes for 2 genera) with a total length of 15,333 bp, 15,291 bp, and 15,381 bp, respectively. The mitogenomes all contain 13 protein-coding genes (PCGs), 22 transfer RNAs (tRNAs), 2 ribosomal RNAs (rRNAs), and a noncoding A + T-rich region and are consistent with other lepidopterans in gene order and type. In addition, we reconstructed the phylogenetic trees of Hesperiinae using maximum likelihood (ML) and Bayesian inference (BI) methods based on mitogenomic data. Results show that the tribe Aeromachini in this study robustly constitute a monophyletic group in the subfamily Hesperiinae, with the relationships Coeliadinae + (Euschemoninae + (Pyrginae + ((Eudaminae + Tagiadinae) + (Heteropterinae + ((Trapezitinae + Barcinae) + Hesperiinae))))). Moreover, our study supports the view that Apostictopterus fuliginosus and Barca bicolor should be placed out of the subfamily Hesperiinae.
