'Aggregation-Induced Emission' Active Mono-Cyclometalated Iridium(III) Complex Mediated Efficient Vapor-Phase Detection of Dichloromethane.

ABSTRACT: Selective vapor-phase detection of dichloromethane (DCM) is a challenge, it being a well-known hazardous volatile organic solvent in trace amounts. With this in mind, we have developed an 'Aggregation-induced Emission' (AIE) active mono-cyclometalated iridium(III)-based (M1) probe molecule, which detects DCM sensitively and selectively in vapor phase with a response time <30 s. It reveals a turn-on emission (non-emissive to intense yellow) on exposing DCM vapor directly to the solid M1. The recorded detection limit is 4.9 ppm for DCM vapor with pristine M1. The mechanism of DCM detection was explored. Moreover, the detection of DCM vapor by M1 was extended with a low-cost filter paper as the substrate. The DCM is weakly bound with the probe and can be removed with a mild treatment, so, notably, the probe can be reused.

Oncosis-inducing cyclometalated iridium(iii) complexes.

Project description:Oncosis is a non-apoptotic form of programmed cell death (PCD), which differs from apoptosis in both morphological changes and inner pathways, and might hold the key to defeating a major obstacle in cancer therapy - drug-resistance, which is often a result of the intrinsic apoptosis resistance of tumours. However, despite the fact that the term "oncosis" was coined and used much earlier than apoptosis, little effort has been made to discover new drugs which can initiate this form of cell death, in comparison to drugs inducing apoptosis or any other type of PCD. So herein, we present the synthesis of a series of mitochondria-targeting cyclometalated Ir(iii) complexes, which activated the oncosis-specific protein porimin and calpain in cisplatin-resistant cell line A549R, and determined their cytotoxicity against a wide range of drug-resistant cancer types. To the best of our knowledge, these complexes are the very first metallo-components to induce oncosis in drug-resistant cancer cells.

Solid-state mechanochemistry for the rapid and efficient synthesis of tris-cyclometalated iridium(iii) complexes.

Project description:Tris-cyclometalated iridium(iii) complexes have received widespread attention as attractive prospective materials for e.g., organic light-emitting diodes (OLEDs), photoredox catalysts, and bioimaging probes. However, their preparation usually requires prolonged reaction times, significant amounts of high-boiling solvents, multistep synthesis, and inert-gas-line techniques. Unfortunately, these requirements represent major drawbacks from both a production-cost and an environmental perspective. Herein, we show that a two-step mechanochemical protocol using ball milling enables the rapid and efficient synthesis of various tris-cyclometalated iridium(iii) complexes from relatively cheap iridium(iii) chloride hydrate without the use of significant amounts of organic solvent in air. Notably, a direct one-pot procedure is also demonstrated. The present solid-state approach can be expected to inspire the development of cost-effective and timely production methods for these valuable iridium-based complexes, as well as the discovery of new phosphorescent materials, sensors, and catalysts.

Project description:The first structure report of trichlorido[4'-(p-tolyl)-2,2':6',2''-terpyridine]iridium(III) dimethyl sulfoxide solvate, [IrCl(3)(C(22)H(17)N(3))] x C(2)H(6)OS, (I), is presented, along with a higher-symmetry setting of previously reported bis[4'-(p-tolyl)-2,2':6',2''-terpyridine]iridium(III) tris(hexafluoridophosphate) acetonitrile disolvate, [Ir(C(22)H(17)N(3))(2)](PF(6))(3) x 2 C(2)H(3)N, (II) [Yoshikawa, Yamabe, Kanehisa, Kai, Takashima & Tsukahara (2007). Eur. J. Inorg. Chem. pp. 1911-1919]. For (I), the data were collected with synchrotron radiation and the dimethyl sulfoxide solvent molecule is disordered over three positions, one of which is an inversion center. The previously reported structure of (II) is presented in the more appropriate C2/c space group. The iridium complex and one PF(6)(-) anion lie on twofold axes in this structure, making half of the molecule unique.

Ester-Modified Cyclometalated Iridium(III) Complexes as Mitochondria-Targeting Anticancer Agents.

Project description:Organometallic iridium complexes are potent anticancer candidates which act through different mechanisms from cisplatin-based chemotherapy regimens. Here, ten phosphorescent cyclometalated iridium(III) complexes containing 2,2'-bipyridine-4,4'-dicarboxylic acid and its diester derivatives as ligands are designed and synthesized. The modification by ester group, which can be hydrolysed by esterase, facilitates the adjustment of drug-like properties. The quantum yields and emission lifetimes are influenced by variation of the ester substituents on the Ir(III) complexes. The cytotoxicity of these Ir(III) complexes is correlated with the length of their ester groups. Among them, 4a and 4b are found to be highly active against a panel of cancer cells screened, including cisplatin-resistant cancer cells. Mechanism studies in vitro indicate that they undergo hydrolysis of ester bonds, accumulate in mitochondria, and induce a series of cell-death related events mediated by mitochondria. Furthermore, 4a and 4b can induce pro-death autophagy and apoptosis simultaneously. Our study indicates that ester modification is a simple and feasible strategy to enhance the anticancer potency of Ir(III) complexes.

NIR-II Emission from Cyclometalated Dinuclear Pt(III) Complexes.

Project description:Half-lantern Pt(II) dinuclear complexes [{Pt(C∧Npz)(μ-S∧NR)}2] (HC∧Npz = 1-naphthalen-2-yl-1H-pyrazole; R = H, HS∧N: 2-mercaptopyrimidine 1; R = CF3, HS∧NF: 4-(trifluoromethyl)-2-mercaptopyrimidine 2) were selectively obtained as single isomers with the C∧N groups in an anti-arrangement and rather short metallophilic interactions (dPt-Pt = 2.8684(2) Å for 2). They reacted with haloforms in the air and sunlight to obtain the corresponding oxidized diplatinum(III) derivatives [{Pt(C∧Npz)(μ-S∧NR)X}2] (X = Cl (1-Cl), Br (1-Br), I (1-I, 2-I)). The single-crystal X-ray structures exhibit Pt-Pt distances typical for the existence of a metal-metal bond, which evidence fairly well the influence of the axial ligand (X). The reactions of 1 and 2 with CHI3 in the dark afforded mixtures of [IPt(C∧Npz)(μ-S∧N)2Pt(C∧Npz)CHI2] and 1-I or 2-I, with the former being the major species under an Ar atmosphere, while the reactions of 1 with CHBr3 and CHCl3 need light to occur. These Pt2(III,III) complexes display low-energy absorptions and emissions that strongly depend on the axial ligand. In the solid state, they show a broad NIR emission ranging from 985 to 1070 nm at RT that suffers a hypsochromic shift when cooling down to 77 K. The photoemissive behavior of the dinuclear Pt(II) and Pt(III) systems is disclosed with the aid of density functional theory calculations.

Janus-type emission from a cyclometalated iron(III) complex.

Project description:Although iron is a dream candidate to substitute noble metals in photoactive complexes, realization of emissive and photoactive iron compounds is demanding due to the fast deactivation of their charge-transfer states. Emissive iron compounds are scarce and dual emission has not been observed before. Here we report the FeIII complex [Fe(ImP)2][PF6] (HImP = 1,1'-(1,3-phenylene)bis(3-methyl-1-imidazol-2-ylidene)), showing a Janus-type dual emission from ligand-to-metal charge transfer (LMCT)- and metal-to-ligand charge transfer (MLCT)-dominated states. This behaviour is achieved by a ligand design that combines four N-heterocyclic carbenes with two cyclometalating aryl units. The low-lying π* levels of the cyclometalating units lead to energetically accessible MLCT states that cannot evolve into LMCT states. With a lifetime of 4.6 ns, the strongly reducing and oxidizing MLCT-dominated state can initiate electron transfer reactions, which could constitute a basis for future applications of iron in photoredox catalysis.

Inhibition of autophagic flux by cyclometalated iridium(iii) complexes through anion transportation.

Project description:Synthetic anion transporters that can interfere with the intracellular pH homeostasis are gaining increasing attention for tumor therapy, however, the biological mechanism of anion transporters remains to be explored. In this work, two phosphorescent cyclometalated Ir(iii) complexes containing 2-phenylpyridine (ppy) as the cyclometalated ligand, and 2,2'-biimidazole (H2biim, Ir1) or 2-(1H-imidazol-2-yl)pyridine (Hpyim, Ir2) as the ancillary ligands have been synthesized and characterized. Due to the protonation and deprotonation process of the N-H groups on H2biim and Hpyim, Ir1 and Ir2 display pH-dependent phosphorescence and can specifically image lysosomes. Both Ir1 and Ir2 can act as anion transporters mainly through the anion exchange mechanism with higher potency observed for Ir1. Mechanism investigation shows that Ir1 and Ir2 can induce caspase-independent cell death through reactive oxygen species (ROS) elevation. As Ir1 and Ir2 can alkalinize lysosomes through anion disturbance, they can inhibit autophagic flux. Our work provides a novel anticancer mechanism of metal complexes, which gives insights into the innovative structure-based design of new metallo-anticancer agents.

Targeting cancer cell metabolism with mitochondria-immobilized phosphorescent cyclometalated iridium(iii) complexes.

Project description:Cancer cell metabolism is reprogrammed to sustain the high metabolic demands of cell proliferation. Recently, emerging studies have shown that mitochondrial metabolism is a potential target for cancer therapy. Herein, four mitochondria-targeted phosphorescent cyclometalated iridium(iii) complexes have been designed and synthesized. Complexes 2 and 4, containing reactive chloromethyl groups for mitochondrial fixation, show much higher cytotoxicity than complexes 1 and 3 without mitochondria-immobilization properties against the cancer cells screened. Further studies show that complexes 2 and 4 induce caspase-dependent apoptosis through mitochondrial damage, cellular ATP depletion, mitochondrial respiration inhibition and reactive oxygen species (ROS) elevation. The phosphorescence of complexes 2 and 4 can be utilized to monitor the perinuclear clustering of mitochondria in real time, which provides a reliable and convenient method for in situ monitoring of the therapeutic effect and gives hints for the investigation of anticancer mechanisms. Genome-wide transcriptional analysis shows that complex 2 exerts its anticancer activity through metabolism repression and multiple cell death signalling pathways. Our work provides a strategy for the construction of highly effective anticancer agents targeting mitochondrial metabolism through rational modification of phosphorescent iridium complexes.

Cyclometalated iridium(iii) complexes as lysosome-targeted photodynamic anticancer and real-time tracking agents.

Project description:Stimuli-activatable photosensitizers (PSs) are highly desirable for photodynamic therapy (PDT) to selectively demolish tumor cells. On the other hand, lysosomes are emerging as attractive anticancer targets. Herein, four cyclometalated iridium(iii)-?-carboline complexes with pH-responsive singlet oxygen (1O2) production and lysosome-specific imaging properties have been designed and synthesized. Upon visible light (425 nm) irradiation, they show highly selective phototoxicities against cancer cells. Notably, complex 2 ([Ir(N^C)2(N^N)](PF6) in which N^C = 2-phenylpyridine and N^N = 1-(2-benzimidazolyl)-?-carboline) displays a remarkably high phototoxicity index (PI = IC50 in the dark/IC50 in light) of >833 against human lung carcinoma A549 cells. Further studies show that 2-mediated PDT induces caspase-dependent apoptosis through lysosomal damage. The pH-responsive phosphorescence of complex 2 can be utilized to monitor the lysosomal integrity upon PDT, which provides a reliable and convenient method for in situ monitoring of therapeutic effect and real-time assessment of treatment outcome. Our work provides a strategy for the construction of highly effective multifunctional subcellular targeted photodynamic anticancer agents through rational structural modification of phosphorescent metal complexes.
