The effect of layer thickness and immobilization chemistry on the detection of CRP in LSPR assays.

ABSTRACT: The immobilization of a capture molecule represents a crucial step for effective usage of gold nanoparticles in localized surface plasmon resonance (LSPR)-based bioanalytics. Depending on the immobilization method used, the resulting capture layer is of varying thickness. Thus, the target binding event takes place at different distances to the gold surface. Using the example of a C-reactive protein immunoassay, different immobilization methods were tested and investigated with regard to their resulting target signal strength. The dependency of the target signal on the distance to the gold surface was investigated utilizing polyelectrolyte bilayers of different thickness. It could be experimentally demonstrated how much the LSPR-shift triggered by a binding event on the gold nanoparticles decreases with increasing distance to the gold surface. Thus, the sensitivity of an LSPR assay is influenced by the choice of immobilization chemistry.

Effect of Layer Thickness in Layered Aluminum Matrix Syntactic Foam.

Project description:This work experimentally investigates the effect of layered structure on the static and impact response of a new layered syntactic foam developed for impact energy absorption. The layered syntactic foam had the same density of 1.6 g/cm3 and the same components of 50% large spheres (L) and 50% small spheres (S) with different structures from two layers to five layers. The impact response and energy absorption were investigated by drop-weight impact tests. Under static loading, more layers led to higher yield stress and lower energy absorption. There were three types of progressive failures of layered syntactic form under impact loading. The failure propagation was examined and found to be dependent on the layer number and impact energy. Interestingly, layered syntactic foam absorbed more energy than both of its components in terms of ductility. The ductility of layered syntactic foam decreased with the increase in layer number. The peak stress of layered syntactic foam increased with the increase in layer number. Two-layered syntactic foam LS had the highest ductility under 60 J/g impact, as well as an energy absorption of 35 J/g, compared to other layered syntactic foams. Specifically, its component L had a ductility under 70 J/g and an energy absorption of 25 J/g, while component S had a ductility under 10 J/g and an energy absorption of 10 J/g.

Protein immobilization techniques for microfluidic assays.

Project description:Microfluidic systems have shown unequivocal performance improvements over conventional bench-top assays across a range of performance metrics. For example, specific advances have been made in reagent consumption, throughput, integration of multiple assay steps, assay automation, and multiplexing capability. For heterogeneous systems, controlled immobilization of reactants is essential for reliable, sensitive detection of analytes. In most cases, protein immobilization densities are maximized, while native activity and conformation are maintained. Immobilization methods and chemistries vary significantly depending on immobilization surface, protein properties, and specific assay goals. In this review, we present trade-offs considerations for common immobilization surface materials. We overview immobilization methods and chemistries, and discuss studies exemplar of key approaches-here with a specific emphasis on immunoassays and enzymatic reactors. Recent "smart immobilization" methods including the use of light, electrochemical, thermal, and chemical stimuli to attach and detach proteins on demand with precise spatial control are highlighted. Spatially encoded protein immobilization using DNA hybridization for multiplexed assays and reversible protein immobilization surfaces for repeatable assay are introduced as immobilization methods. We also describe multifunctional surface coatings that can perform tasks that were, until recently, relegated to multiple functional coatings. We consider the microfluidics literature from 1997 to present and close with a perspective on future approaches to protein immobilization.

Thickness-Dependent NIR LSPR of Curved Ag/TiS<sub>2</sub> Bilayer Film.

Project description:We demonstrated that the localized surface plasmon resonance (LSPR) features of Ag/TiS2 nanostructures were dependent on the sublayer thickness. The Ag/TiS2 bilayer film was obtained by the self-assembly method and magnetron sputtering. The thickness was controlled by changing the sputtering time when the sputtering powers were the same. When the Ag thickness decreased from 50 nm to 5 nm, the LSPR was tuned from the visible region to the Near Infrared (NIR) region. When the TiS2 thickness decreased from 60 nm to 2 nm, the LSPR shifted from the IR to NIR region. Analysis showed the thickness changes of Ag and TiS2 resulted in the changed carrier density, which led to the thickness-dependent shift of the LSPR.

Effect of intravitreal ranibizumab and aflibercept injections on retinal nerve fiber layer thickness.

Project description:The purpose is to evaluate the effects of multiple intravitreal ranibizumab (IVR) and aflibercept (IVA) injections on peripapillary retinal nerve fiber layer (RNFL) thickness in patients with exudative age-related macular degeneration (AMD). This retrospective, observational, consecutive case series study enrolled patients newly diagnosed with monocular exudative AMD from January 2014 to October 2019 who were administered IVR or IVA injections. Normal fellow eyes were included as controls. Medical records and spectral domain optical coherence tomography results were reviewed at baseline and at 3, 6, and 12 months after injection. No statistically significant differences in peripapillary RNFL thickness and intraocular pressure were observed between the treated and fellow eyes in the two groups. The global RNFL thicknesses for the treated eyes decreased significantly after 12 months compared with baseline, but no significant difference was observed in any of the six examined sectors (temporal, superior temporal, superior nasal, nasal, inferior nasal, and inferior temporal). At 12 months, the central macular thickness of the treated eyes decreased significantly. Multiple IVR and IVA injections are apparently safe considering peripapillary RNFL damage in patients with exudative AMD. The decreased RNFL thickness of the global sector was presumably due to anatomical improvement of macular lesions.

Photothermal Effect in Plasmonic Nanotip for LSPR Sensing.

Project description:The influence of heat generation on the conventional process of LSPR based sensing has not been explored thus far. Therefore, a need exists to draw attention toward the heat generation issue during LSPR sensing as it may affect the refractive index of the analyte, leading to incorrect sensory conclusions. This manuscript addresses the connection between the photo-thermal effect and LSPR. We numerically analyzed the heat performance of a gold cladded nanotip. The numerical results predict a change in the micro-scale temperature in the microenvironment near the nanotip. These numerical results predict a temperature increase of more than 20 K near the apex of the nanotip, which depends on numerous factors including the input optical power and the diameter of the fiber. We analytically show that this change in the temperature influences a change in the refractive index of the microenvironment in the vicinity of the nanotip. In accordance with our numerical and analytical findings, we experimentally show an LSPR shift induced by a change in the input power of the source. We believe that our work will bring the importance of temperature dependence in nanotip based LSPR sensing to the fore.

LSPR Biosensing Approach for the Detection of Microtubule Nucleation.

Project description:Microtubules are dynamic protein filaments that are involved in a number of cellular processes. Here, we report the development of a novel localized surface plasmon resonance (LSPR) biosensing approach for investigating one aspect of microtubule dynamics that is not well understood, namely, nucleation. Using a modified Mie theory with radially variable refractive index, we construct a theoretical model to describe the optical response of gold nanoparticles when microtubules form around them. The model predicts that the extinction maximum wavelength is sensitive to a change in the local refractive index induced by microtubule nucleation within a few tens of nanometers from the nanoparticle surface, but insensitive to a change in the refractive index outside this region caused by microtubule elongation. As a proof of concept to demonstrate that LSPR can be used for detecting microtubule nucleation experimentally, we induce spontaneous microtubule formation around gold nanoparticles by immobilizing tubulin subunits on the nanoparticles. We find that, consistent with the theoretical model, there is a redshift in the extinction maximum wavelength upon the formation of short microtubules around the nanoparticles, but no significant change in maximum wavelength when the microtubules are elongated. We also perform kinetic experiments and demonstrate that the maximum wavelength is sensitive to the microtubule nuclei assembly even when microtubules are too small to be detected from an optical density measurement.

Thickness-Dependent Permeance of Molecular Layer-By-Layer Polyamide Membranes.

Project description:We present the thickness-dependent permeance of highly cross-linked polyamide (PA) membranes formed by a molecular layer-by-layer (mLbL) deposition process. The deposition allows for the synthesis of extremely smooth, uniform PA films of tunable thickness, which is counter to the less controlled interfacial polymerization process used commercially. The ability to control and measure the membrane thickness allows us to elucidate the relationships among network structure, transport properties, and separation performance. In this work, a series of large-area mLbL PA membranes is prepared with thickness ranging from less than 5 nm to greater than 100 nm, which can be transferred defect-free via a film floating technique onto a macroporous support layer and challenged with salt solutions. A critical thickness of 15 nm is identified for efficient desalination, and water permeance is described using a multi-layer solution diffusion model that allows for the extraction of material properties relevant to transport. Finally, the model demonstrates the existence of two distinct layers in the mLbL films, one layer comprised of a (5 to 10) nm graded or less cross-linked layer at the surface and a more densely cross-linked layer in the interior of the film. This graded layer appears inherent to the mLbL deposition process and is observed at all film thicknesses.

Silver nanotriangle array based LSPR sensor for rapid coronavirus detection.

Project description:A rapid, portable, and cost-effective method to detect the infection of SARS-CoV-2 is fundamental toward mitigating the current COVID-19 pandemic. Herein, a human angiotensin-converting enzyme 2 protein (ACE2) functionalized silver nanotriangle (AgNT) array localized surface plasmon resonance (LSPR) sensor is developed for rapid coronavirus detection, which is validated by SARS-CoV-2 spike RBD protein and CoV NL63 virus with high sensitivity and specificity. A linear shift of the LSPR wavelength versus the logarithm of the concentration of the spike RBD protein and CoV NL63 is observed. The limits of detection for the spike RBD protein, CoV NL63 in buffer and untreated saliva are determined to be 0.83 pM, 391 PFU/mL, and 625 PFU/mL, respectively, while the detection time is found to be less than 20 min. Thus, the AgNT array optical sensor could serve as a potential rapid point-of-care COVID-19 diagnostic platform.

LSPR-Based Aptasensor for Rapid Urinary Detection of NT-proBNP.

Project description:N-terminal pro-brain natriuretic peptide (NT-proBNP) is a myocardial stress biomarker that can be found in serum or plasma, saliva, and urine in the context of cardiovascular disease. In this study, we developed a rapid (~25 min) and straightforward localized surface plasmon resonance (LSPR)-based assay for detecting NT-proBNP in urine. The assay employs citrate-capped gold nanoparticles (AuNPs) and an aptamer specific for NT-proBNP, which initially interacts with NT-proBNP. The remaining unbound aptamer then interacts with the AuNPs, and the addition of NaCl induces the aggregation of the unprotected AuNPs, resulting in a decrease in absorbance at the LSPR band (A521) and an increase in absorbance at 750 nm (A750). The concentration of NT-proBNP showed a linear correlation with the aggregation ratio (A521/A750), and the assay demonstrated a limit of detection (LOD) of 0.303 µg·L-1 and a detection range of 0.566-8 µg·L-1. However, the presence of sulfur-containing proteins in saliva and fetal bovine serum hindered the detection of NT-proBNP in these biofluids. Nevertheless, the assay successfully detected NT-proBNP in diluted urine with an LOD of 0.417 µg·L-1 and a detection range of 0.589-6 µg·L-1. The observed values in urine samples from preterm infants with cardiovascular disease fell within this range, indicating the potential clinical relevance of the assay. The recovery percentages ranged from 92.3 to 116.3%. Overall, our findings suggest that the LSPR-based assay for NT-proBNP detection in urine can be a valuable tool for the diagnosis and treatment of cardiovascular disease.

Reliability of Intra-Retinal Layer Thickness Estimates.

Project description:PurposeMeasurement of intra-retinal layer thickness using optical coherence tomography (OCT) has become increasingly prominent in multiple sclerosis (MS) research. Nevertheless, the approaches used for determining the mean layer thicknesses vary greatly. Insufficient data exist on the reliability of different thickness estimates, which is crucial for their application in clinical studies. This study addresses this lack by evaluating the repeatability of different thickness estimates.MethodsStudies that used intra-retinal layer segmentation of macular OCT scans in patients with MS were retrieved from PubMed. To investigate the repeatability of previously applied layer estimation approaches, we generated datasets of repeating measurements of 15 healthy subjects and 13 multiple sclerosis patients using two OCT devices (Cirrus HD-OCT and Spectralis SD-OCT). We calculated each thickness estimate in each repeated session and analyzed repeatability using intra-class correlation coefficients and coefficients of repeatability.ResultsWe identified 27 articles, eleven of them used the Spectralis SD-OCT, nine Cirrus HD-OCT, two studies used both devices and two studies applied RTVue-100. Topcon OCT-1000, Stratus OCT and a research device were used in one study each. In the studies that used the Spectralis, ten different thickness estimates were identified, while thickness estimates of the Cirrus OCT were based on two different scan settings. In the simulation dataset, thickness estimates averaging larger areas showed an excellent repeatability for all retinal layers except the outer plexiform layer (OPL).ConclusionsGiven the good reliability, the thickness estimate of the 6mm-diameter area around the fovea should be favored when OCT is used in clinical research. Assessment of the OPL was weak in general and needs further investigation before OPL thickness can be used as a reliable parameter.
