Project description:Aerosols dispersed and transmitted through the air (e.g., particulate matter pollution and bioaerosols) are ubiquitous and one of the leading causes of adverse health effects and disease transmission. A variety of sampling methods (e.g., filters, cyclones, and impactors) have been developed to assess personal exposures. However, a gap still remains in the accessibility and ease-of-use of these technologies for people without experience or training in collecting airborne samples. Additionally, wet scrubbers (large non-portable industrial systems) utilize liquid sprays to remove aerosols from the air; the goal is to "scrub" (i.e., clean) the exhaust of industrial smokestacks, not collect the aerosols for analysis. Inspired by wet scrubbers, we developed a device fundamentally different from existing portable air samplers by using aerosolized microdroplets to capture aerosols in personal spaces (e.g., homes, offices, and schools). Our aerosol-sampling device is the size of a small teapot, can be operated without specialized training, and features a winding flow path in a supersaturated relative humidity environment, enabling droplet growth. The integrated open mesofluidic channels shuttle coalesced droplets to a collection chamber for subsequent sample analysis. Here, we present the experimental demonstration of aerosol capture in water droplets. An iterative study optimized the non-linear flow manipulating baffles and enabled an 83% retention of the aerosolized microdroplets in the confined volume of our device. As a proof-of-concept for aerosol capture into a liquid medium, 0.5-3 μm model particles were used to evaluate aerosol capture efficiency. Finally, we demonstrate that the device can capture and keep a bioaerosol (bacteriophage MS2) viable for downstream analysis.

Project description:Among the current CO2 capture technologies, membrane gas separation has many inherent advantages over other conventional techniques. However, fabricating gas separation membranes with both high CO2 permeance and high CO2/N2 selectivity, especially under wet conditions, is a challenge. In this study, sub-20-nm thick, layered graphene oxide (GO)-based hollow fiber membranes with grafted, brush-like CO2-philic agent alternating between GO layers are prepared by a facile coating process for highly efficient CO2/N2 separation under wet conditions. Piperazine, as an effective CO2-philic agent, is introduced as a carrier-brush into the GO nanochannels with chemical bonding. The membrane exhibits excellent separation performance under simulated flue gas conditions with CO2 permeance of 1,020 GPU and CO2/N2 selectivity as high as 680, demonstrating its potential for CO2 capture from flue gas. We expect this GO-based membrane structure combined with the facile coating process to facilitate the development of ultrathin GO-based membranes for CO2 capture.

Project description:We describe a straightforward and scalable fabrication of diamine-appended metal-organic framework (MOF)/polymer composite hollow fiber sorbent modules for CO2 capture from dilute streams, such as flue gas from natural gas combined cycle (NGCC) power plants. A specific Mg-MOF, Mg2(dobpdc) (dobpdc4- = 4,4'-dioxidobiphenyl-3,3'-dicarboxylate), incorporated into poly(ether sulfone) (PES) is directly spun through a conventional "dry-jet, wet-quench" method. After phase separation, a cyclic diamine 2-(aminomethyl)piperidine (2-ampd) is infused into the MOF within the polymer matrix during postspinning solvent exchange. The MOF hollow fibers from direct spinning contain as high as 70% MOF in the total fibers with 98% of the pure MOF uptake. The resulting fibers exhibit a step isotherm and a "shock-wave-shock" breakthrough profile consistent with pure 2-ampd-Mg2(dobpdc). This work demonstrates a practical method for fabricating 2-ampd-Mg2(dobpdc) fiber sorbents that display the MOF's high CO2 adsorption capacity while lowering the pressure drop during operation.

Project description:The CO2 separation from flue gas based on membrane technology has drawn great attention in the last few decades. In this work, polyetherimide (PEI) hollow fibers were fabricated by using a dry-jet-wet spinning technique. Subsequently, the composite hollow fiber membranes were prepared by dip coating of polydimethylsiloxane (PDMS) selective layer on the outer surface of PEI hollow fibers. The hollow fibers spun from various spinning conditions were fully characterized. The influence of hollow fiber substrates on the CO2/N2 separation performance of PDMS/PEI composite membranes was estimated by gas permeance and ideal selectivity. The prepared composite membrane where the hollow fiber substrate was spun from 20 wt% of dope solution, 12 mL/min of bore fluid (water) flow rate exhibited the highest ideal selectivity equal to 21.3 with CO2 permeance of 59 GPU. It was found that the dope concentration, bore fluid flow rate and bore fluid composition affect the porous structure, surface morphology and dimension of hollow fibers. The bore fluid composition significantly influenced the gas permeance and ideal selectivity of the PDMS/PEI composite membrane. The prepared PDMS/PEI composite membranes possess comparable CO2/N2 separation performance to literature ones.

Project description:Hollow-core anti-resonant fiber technology has made a rapid progress in low loss broadband transmission, enabled by its much reduced light-material overlap. This unique characteristic has driven emerging of new applications spanning from extreme wavelength generation to beam delivery. The successful demonstrations appear to suggest progression of the technology toward device level development and all-fiberized systems. We investigate this opportunity and report an in-fiber interferometer built in a dual hollow-core anti-resonant fiber. By placing multiple air cores in a single fiber, coherently interacting transverse modes are excited, which becomes a basis of an interferometer. We use this hollow core based inherent supermodal interaction to demonstrate highly sensitive in-fiber interferometer. Unique combination of the air guidance and the supermodal interaction offers robust, simple yet highly sensitive interferometer with suppressed temperature cross-talk that has been an enduring problem in fiber strain sensing applications. The in-fiber interferometer is further investigated as a sensing element for pressure measurement based on an interferometric phase change upon external strain. The interferometer features 39.3 nm/MPa of ultrahigh sensitivity with 0.14 KPa/°C of negligible gas pressure temperature crosstalk. The performance, which is much improved from prior fiber sensors, testifies advances of hollow core fiber technology toward a device level.

Project description:There has been a resurgence of interest in aerosolization of antibiotics for treatment of patients with severe pneumonia caused by multidrug-resistant pathogens. A combination formulation of amikacin-fosfomycin is currently undergoing clinical testing although the exposure-response relationships of these drugs have not been fully characterized. The aim of this study was to describe the individual and combined antibacterial effects of simulated epithelial lining fluid exposures of aerosolized amikacin and fosfomycin against resistant clinical isolates of Pseudomonas aeruginosa (MICs of 16 mg/liter and 64 mg/liter) and Klebsiella pneumoniae (MICs of 2 mg/liter and 64 mg/liter) using a dynamic hollow-fiber infection model over 7 days. Targeted peak concentrations of 300 mg/liter amikacin and/or 1,200 mg/liter fosfomycin as a 12-hourly dosing regimens were used. Quantitative cultures were performed to describe changes in concentrations of the total and resistant bacterial populations. The targeted starting inoculum was 108 CFU/ml for both strains. We observed that neither amikacin nor fosfomycin monotherapy was bactericidal against P. aeruginosa while both were associated with rapid amplification of resistant P. aeruginosa strains (about 108 to 109 CFU/ml within 24 to 48 h). For K. pneumoniae, amikacin but not fosfomycin was bactericidal. When both drugs were combined, a rapid killing was observed for P. aeruginosa and K. pneumoniae (6-log kill within 24 h). Furthermore, the combination of amikacin and fosfomycin effectively suppressed growth of resistant strains of P. aeruginosa and K. pneumoniae In conclusion, the combination of amikacin and fosfomycin was effective at maximizing bacterial killing and suppressing emergence of resistance against these clinical isolates.

Project description:Kidney-on-a-chip devices may revolutionize the discovery of new therapies. However, fabricating a 3D glomerulus remains a challenge, due to a requirement for a microscale soft material with complex topography to support cell culture in a native configuration. Here, we describe the use of microfluidic spinning to recapitulate complex concave and convex topographies over multiple length scales, required for biofabrication of a biomimetic 3D glomerulus. We produced a microfluidic extruded topographic hollow fiber (h-FIBER), consisting of a vessel-like perfusable tubular channel for endothelial cell cultivation, and a glomerulus-like knot with microconvex topography on its surface for podocyte cultivation. Meter long h-FIBERs were produced in microfluidics within minutes, followed by chemically induced inflation for generation of topographical cues on the 3D scaffold surface. The h-FIBERs were assembled into a hot-embossed plastic 96-well plate. Long-term perfusion, podocyte barrier formation, endothelialization, and permeability tests were easily performed by a standard pipetting technique on the platform. Following long-term culture (1 month), a functional filtration barrier, measured by the transfer of albumin from the blood vessel side to the ultrafiltrate side, suggested the establishment of an engineered glomerulus.

Project description:Plasma wakefield accelerators have been used to accelerate electron and positron particle beams with gradients that are orders of magnitude larger than those achieved in conventional accelerators. In addition to being accelerated by the plasma wakefield, the beam particles also experience strong transverse forces that may disrupt the beam quality. Hollow plasma channels have been proposed as a technique for generating accelerating fields without transverse forces. Here we demonstrate a method for creating an extended hollow plasma channel and measure the wakefields created by an ultrarelativistic positron beam as it propagates through the channel. The plasma channel is created by directing a high-intensity laser pulse with a spatially modulated profile into lithium vapour, which results in an annular region of ionization. A peak decelerating field of 230?MeV?m(-1) is inferred from changes in the beam energy spectrum, in good agreement with theory and particle-in-cell simulations.

Project description:Bioprinting emerges as a powerful flexible approach for tissue engineering with prospective capability to produce tissue on demand, including biomimetic hollow-core fiber structures. In spite of significance for tissue engineering, hollow-core structures proved difficult to fabricate, with the existing methods limited to multistage, time-consuming, and cumbersome procedures. Here, we report a versatile cell-friendly photopolymerization approach that enables single-step prototyping of hollow-core as well as solid-core hydrogel fibers initially loaded with living cells. This approach was implemented by extruding cell-laden hyaluronic acid glycidyl methacrylate hydrogel directly into aqueous solution containing free radicals generated by continuous blue light photoexcitation of the flavin mononucleotide/triethanolamine photoinitiator. Diffusion of free radicals from the solution to the extruded structure initiated cross-linking of the hydrogel, progressing from the structure surface inwards. Thus, the cross-linked wall is formed and its thickness is limited by penetration of free radicals in the hydrogel volume. After developing in water, the hollow-core fiber is formed with centimeter range of lengths. Amazingly, HaCaT cells embedded in the hydrogel successfully go through the fabrication procedure. The broad size ranges have been demonstrated: from solid core to 6% wall thickness of the outer diameter, which was variable from sub-millimeter to 6 mm, and Young's modulus ∼1.6 ± 0.4 MPa. This new proof-of-concept fibers photofabrication approach opens lucrative opportunities for facile three-dimensional fabrication of hollow-core biostructures with controllable geometry.

Project description:The HIV-1 glycoprotein spike (gp120) is typically the first viral antigen that cells encounter before initiating immune responses, and is often the sole target in vaccine designs. Thus, characterizing the presence of cellular antigens on the surfaces of HIV particles may help identify new antiviral targets or impact targeting of gp120. Despite the importance of characterizing proteins on the virion surface, current techniques available for this purpose do not support high-throughput analysis of viruses, and typically only offer a semi-quantitative assessment of virus-associated proteins. Traditional bulk techniques often assess averages of viral preparations, which may mask subtle but important differences in viral subsets. On the other hand, microscopy techniques, which provide detail on individual virions, are difficult to use in a high-throughput manner and have low levels of sensitivity for antigen detection. Flow cytometry is a technique that traditionally has been used for rapid, high-sensitivity characterization of single cells, with limited use in detecting viruses, since the small size of viral particles hinders their detection. Herein, we report the detection and surface antigen characterization of HIV-1 pseudovirus particles by light scattering and fluorescence with flow cytometry, termed flow virometry for its specific application to viruses. We quantified three cellular proteins (integrin α4β7, CD14, and CD162/PSGL-1) in the viral envelope by directly staining virion-containing cell supernatants without the requirement of additional processing steps to distinguish virus particles or specific virus purification techniques. We also show that two antigens can be simultaneously detected on the surface of individual HIV virions, probing for the tetraspanin marker, CD81, in addition to α4β7, CD14, and CD162/PSGL-1. This study demonstrates new advances in calibrated flow virometry as a tool to provide sensitive, high-throughput characterization of the viral envelope in a more efficient, quantitative manner than previously reported techniques.
