Molecular Detection of Nosema spp. in Honey in Bulgaria.

ABSTRACT: Environmental DNA (eDNA) analysis is related to screening genetic material of various organisms in environmental samples. Honey represents a natural source of exogenous DNA, which allows for the detection of different honey bee pathogens and parasites. In the present study, we extracted DNA from 20 honey samples from different regions in Bulgaria and tested for the presence of DNA of the ectoparasitic mite Varroa destructor, as well as Nosema apis and Nosema ceranae. Only Nosema ceranae was detected, showing up in 30% of all samples, which confirms the widespread prevalence of this pathogen. All positive samples were found in plain regions of the country, while this pathogen was not detected in mountainous parts. None of the samples gave positive amplifications for the Nosema apis and Varroa mite. The obtained results from this study confirm previous observations that eDNA contained in honey is a potent source for effective biomonitoring of actual diseases in the honey bee.

Cold Ambient Temperature Promotes Nosema spp. Intensity in Honey Bees (Apis mellifera).

Project description:Interactions between parasites and environmental factors have been implicated in the loss of managed Western honey bee (=HB, Apis mellifera) colonies. Although laboratory data suggest that cold temperature may limit the spread of Nosema ceranae, an invasive species and now ubiquitous endoparasite of Western HBs, the impact of weather conditions on the distribution of this microsporidian in the field is poorly understood. Here, we conducted a survey for Nosema spp. using 18 Swiss apiaries (four colonies per apiary) over a period of up to 18 months. Samples consisting of 60 workers were collected monthly from each colony to estimate Nosema spp. intensity, i.e., the number of spores in positive samples using microscopy. Ambient apiary temperature was measured daily to estimate the proportion of days enabling HB flight (>10 °C at midday). The results show that Nosema spp. intensities were negatively correlated with the proportion of days enabling HB flight, thereby suggesting a significant and unexpected positive impact of cold ambient temperature on intensities, probably via regulation of defecation opportunities for infected hosts.

Geographical distribution and molecular detection of Nosema ceranae from indigenous honey bees of Saudi Arabia.

Project description:The aim of the study was to detect the infection level of honey bees with Nosema apis and/or Nosema ceranae using microscopic and molecular analysis from indigenous honeybee race of eight Saudi Arabian geographical regions. A detailed survey was conducted and fifty apiaries were chosen at random from these locations. Infection level was determined both by microscope and Multiplex-PCR and data were analyzed using bioinformatics tools and phylogenetic analysis. Result showed that N. ceranae was the only species infecting indigenous honeybee colonies in Saudi Arabia. As determined by microscope, Nosema spores were found to be in 20.59% of total samples colonies, while 58% of the samples evaluated by PCR were found to be positive for N. ceranae, with the highest prevalence in Al-Bahah, a tropical wet and dry climatic region, whereas low prevalence was found in the regions with hot arid climate. Honeybees from all eight locations surveyed were positive for N. ceranae. This is the first report about the N. ceranae detection, contamination level and distribution pattern in Saudi Arabia.

Nosema Disease of European Honey Bees.

Project description:Nosematosis is currently a frequently discussed honey bee disease caused by two types of Microsporidia: Nosema apis and Nosema ceranae. Nosematosis as an intestinal disease caused by these species is one of the main factors associated with the weakening and loss of hives, with none of the stressors acting in isolation and all having an important synergistic or additive effect on the occurrence of parasitic infection. The most important factors are exposure to pesticides and nutritional stress, both worsening the immune response. Honey bees Apis mellifera become more susceptible to parasites and subsequently the disease manifests itself. Choosing the right laboratory diagnostics is important to determine the prevalence of both species. Our review summarizes the most commonly used methodologies, especially polymerase chain reaction (PCR), which is a reliable method for detecting nosematosis, as well as for distinguishing between the two species causing the disease.

Nosema spp. infection and its negative effects on honey bees (Apis mellifera iberiensis) at the colony level.

Project description:Nosemosis caused by the microsporidia Nosema apis and Nosema ceranae are among the most common pathologies affecting adult honey bees. N. apis infection has been associated with a reduced lifespan of infected bees and increased winter mortality, and its negative impact on colony strength and productivity has been described in several studies. By contrast, when the effects of nosemosis type C, caused by N. ceranae infection, have been analysed at the colony level, these studies have largely focused on collapse as a response to infection without addressing the potential sub-clinical effects on colony strength and productivity. Given the spread and prevalence of N. ceranae worldwide, we set out here to characterize the sub-clinical and clinical signs of N. ceranae infection on colony strength and productivity. We evaluated the evolution of 50 honey bee colonies naturally infected by Nosema (mainly N. ceranae) over a one year period. Under our experimental conditions, N. ceranae infection was highly pathogenic for honey bee colonies, producing significant reductions in colony size, brood rearing and honey production. These deleterious effects at the colony level may affect beekeeping profitability and have serious consequences on pollination. Further research is necessary to identify possible treatments or beekeeping techniques that will limit the rapid spread of this dangerous emerging disease.

Project description:We report on the identification of two new Francisella-like endosymbionts (FLEs) found in three different tick species from Bulgaria. The FLEs were characterized by 16S rRNA and tul4 gene sequencing and seem to lack the molecular marker RD1. These two new taxa seem to be facultative secondary endosymbionts of ticks.

The first report of the prevalence of Nosema ceranae in Bulgaria.

Project description:Nosema apis and Nosema ceranae are the two main microsporidian parasites causing nosematosis in the honey bee Apis mellifera. The aim of the present study is to investigate the presence of Nosema apis and Nosema ceranae in the area of Bulgaria. The 16S (SSU) rDNA gene region was chosen for analysis. A duplex PCR assay was performed on 108 honey bee samples from three different parts of the country (South, North and West Bulgaria). The results showed that the samples from the northern part of the country were with the highest prevalence (77.2%) for Nosema ceranae while those from the mountainous parts (the Rodopa Mountains, South Bulgaria) were with the lowest rate (13.9%). Infection with Nosema apis alone and co-infection N. apis/N. ceranae were not detected in any samples. These findings suggest that Nosema ceranae is the dominant species in the Bulgarian honey bee. It is not known when the introduction of Nosema ceranae in Bulgaria has occurred, but as in the rest of the world, this species has become the dominant one in Bulgarian Apis mellifera. In conclusion, this is the first report for molecular detection of Nosema infection of honey bee in Bulgaria. The results showed that N. ceranae is the main Nosema species in Bulgaria.

Proteomics of anatomical sections of the gut of Nosema-infected Western honey bee (Apis mellifera) reveals different early responses to Nosema spp. isolates

Project description:We aim to evaluate the effects of four Nosema spores’ isolates, (i) and (ii) N. ceranae isolated from A. mellifera hosts from two different geographical origins, (iii) N. ceranae from A. cerana host and (iv) N. apis from A. mellifera, on the A. mellifera on gut proteomics at the early stage of infection. To dissect the molecular mechanism responsible of the susceptibility of A. mellifera to Nosema, we investigated by high-resolution proteomics (LC-ESI-MS/MS) and differential label-free quantification of proteins (LFQ) the molecular cross-talk existing between different species and isolates of N. apis and N. ceranae, and the targetted gut tissue of A. mellifera. To reach the objectives of this study, we performed a bottom-up proteomic analysis on the different anatomical sections of the gut tissue (esophagus, crop, midgut, ileum and rectum) at an early stage of the exposition to Nosema spores (4 days). Then, we focused on the midgut, the region targeted by Nosema sposres for germination and, as we found out, the second region with the highest load of Nosema proteins, after the rectum, to perform differential quantitative proteomic analyses and acquire series of up- and down-regulated proteins. We discussed the different pathways observed to be impacted by different Nosema species and isolates with a main focus on the deregulated metabolic and response to stimuli processes.

Nosema ceranae escapes fumagillin control in honey bees.

Project description:Fumagillin is the only antibiotic approved for control of nosema disease in honey bees and has been extensively used in United States apiculture for more than 50 years for control of Nosema apis. It is toxic to mammals and must be applied seasonally and with caution to avoid residues in honey. Fumagillin degrades or is diluted in hives over the foraging season, exposing bees and the microsporidia to declining concentrations of the drug. We showed that spore production by Nosema ceranae, an emerging microsporidian pathogen in honey bees, increased in response to declining fumagillin concentrations, up to 100% higher than that of infected bees that have not been exposed to fumagillin. N. apis spore production was also higher, although not significantly so. Fumagillin inhibits the enzyme methionine aminopeptidase2 (MetAP2) in eukaryotic cells and interferes with protein modifications necessary for normal cell function. We sequenced the MetAP2 gene for apid Nosema species and determined that, although susceptibility to fumagillin differs among species, there are no apparent differences in fumagillin binding sites. Protein assays of uninfected bees showed that fumagillin altered structural and metabolic proteins in honey bee midgut tissues at concentrations that do not suppress microsporidia reproduction. The microsporidia, particularly N. ceranae, are apparently released from the suppressive effects of fumagillin at concentrations that continue to impact honey bee physiology. The current application protocol for fumagillin may exacerbate N. ceranae infection rather than suppress it.

Characterizing the Impact of Commercial Pollen Substitute Diets on the Level of Nosema spp. in Honey Bees (Apis mellifera L.).

Project description:Western honey bee (Apis mellifera L.) populations face declines commonly attributed to pesticide, pathogen, and parasite stress. One way beekeepers combat these stressors is by providing supplemental protein diets to honey bee colonies to ensure adequate colony nutrition. However Nosema spp., a microsporidian parasite of the honey bee, is thought to be associated closely with a colony's nutritional intake, thus possibly negating any benefit the bees otherwise would have received from a nutritional supplement. Through three objectives, we examined how adult bees' consumption of wildflower pollen or commercial pollen substitute diets affected Nosema levels in the bees' midguts. For our first objective, we investigated how method of inoculation with Nosema affects infection levels in inoculated bees. Bees were infected with spores of Nosema four days after emergence. On day 15, bees were collected from the cages and Nosema spores were quantified. We found that inoculation through the pollen diet resulted in the highest Nosema levels in inoculated bees. In our second and third objectives, we provided the test diets to caged, newly emerged bees for a period of 15 days. Bees consuming pollen and a sucrose solution had more Nosema in their midguts than did bees consuming the sucrose solution alone (control). The overall volume of diet consumed by the bees did not correlate with the level of Nosema in their midguts. The level of Nosema was higher in bees fed certain commercial pollen substitute diets than in bees fed wildflower pollen. Our study illustrates how providing nutritional supplements to adult honey bees can impact the intensity of Nosema in their midguts.
