Molecular and morphological evidence for a new species of Pogostemon (Lamiaceae) from Hainan Island, China.

ABSTRACT: Pogostemonhainanensis, a new species of Lamiaceae from Hainan Island, China, is described. The phylogenetic position of the new species within Pogostemon was investigated based on analyses of the nuclear ribosomal internal transcribed spacer (nrITS) and five plastid markers (viz. matK, psbA-trnH, rbcL, rsp16, trnL-F). The results show that P.hainanensis is supported to be a member of subgenus Pogostemon and is sister to P.parviflorus, a species widely distributed from Eastern Himalaya, through the Indo-China peninsula to China. Morphologically, the new species can be distinguished from all the other taxa of subgenus Pogostemon in having long petioles usually 4.5‒11.5 cm in length, and the calyx teeth 2/3 to subequal as long as the calyx tube. The new species differs from P.parviflorus further by its obviously double serrate leaf margin, spikes of inflorescence usually 2.5-8.0 cm long, calyx 4‒5 mm long and corolla 6-7 mm long.

Morphological and Molecular Identification of Hard Ticks in Hainan Island, China.

Project description:Ticks are small, blood-sucking arachnids, known vectors of various diseases, and found throughout the world. They are distributed basically in almost all regions of China. At present, there is not much information regarding tick species on Hainan Island. They were subjected to morphological identification and imaging on an individual basis. Molecular phylogenetic analyses, based on cox1 and 16S rRNA genes, were utilized to identify the species and determine their approximate phylogenetic origin and genetic diversity. The genomic DNA of tick species was extracted, and cytochrome oxidase subunit 1 (cox1) and 16S ribosomal RNA (rRNA) genes were amplified and sequenced. The identification of five tick species, namely Rhipicephalus microplus, Rhipicephalus sanguineus, Rhipicephalus haemaphysaloides, Haemaphysalis cornigera and Haemaphysalis mageshimaensis, was carried out by morphological analysis. When employing the cox1 and 16S rRNA phylogenetic tree, all isolates of R. microplus from Hainan Island were classified as clade A and B, respectively. R. sanguineus was recognized as a member of the tropical lineage by phylogenetic analysis on the cox1 and 16S rRNA genes. Three phylogenetic groups of R. haemaphysaloides were recognized and found to be related closely to strains from China. H. cornigera and H. mageshimaensis formed one phylogenetic group, presumably from tick strains prevalent in Japan and China. The haplotype network analysis indicated that R. microplus is classed into 26 and 6 haplotypes, which correspond to cox1 and 16S rRNA gene assemblages, respectively. In addition, four cox1 haplotypes were detected in R. sanguineus. This is the first evidence that suggests genetic diversity, host range and geographical distribution of hard ticks in Hainan Island, China.

Protura from Hainan Island, China: new species, checklist and distribution.

Project description:More than 1500 proturan specimens from Hainan Island are systematically studied. An annotated list of all species of Protura from Hainan Island is provided and their geographical distribution is discussed. The genus Paracondeellum is reported from Hainan Island for the first time, and Paracondeellum paradisum sp. nov. is described. The type species Paracondeellum dukouense (Tang & Yin, 1988) is redescribed based on syntype, and the lectotype and paralectotype are designated. The characters of the genus Paracondeellum are redefined, and the two known species are compared in detail. The Protura fauna of Hainan Island is mainly composed of species from the Oriental region, with 91% of the species belonging to the families Berberentulidae and Eosentomidae.

Project description:Patania clava sp. n. is described from the Diaoluo Mountain of Hainan Island, China. The new species is distinguished from its most similar congener, Patania iopasalis (Walker, 1859), by the following features: wingspan 33.0-35.0 mm (vs. 21.0-30.0 mm), ventral cilia of the male antenna as long as the diameter of flagellomere (vs. 1/4), the thick finger-like gnathos (vs. the short broad sheet-like), a long thick needle-like cornutus stretching out from (vs. embedded in) a cluster of spicular cornuti near apex. Images of adult and genitalia of the new species are provided.

<i>Rhamnella brachycarpa</i> (Rhamnaceae), a new species from Hainan Island, China.

Project description:Rhamnella brachycarpa Z. Qiang Lu & Y. Shuai Sun, a new evergreen woody species from Hainan Island, is described and illustrated. The specimens of this new species have previously been identified and placed under R. rubrinervis (H. Lév.) Rehder, with which it shares evergreen leaves, erect and climbing habits and axillary flowering branches with bracteole leaves. However, the specimens from three distinct Hainan populations significantly differ from those of R. rubrinervis from other regions with smaller length to width ratios of leaves, fruit and seeds, smaller sizes of fruit and seeds and mucronate seed apices. Principal Component Analysis of the closely related taxa, based on multiple morphological characters, further recognised two separated groups. One of them comprises R. tonkinensis and R. rubrinervis, the other merely includes all individuals from these distinct Hainan populations. Therefore, R. brachycarpa, based on these distinct Hainan populations, is here erected as a new species, distinctly different from its published relatives.

Oreocharis jasminina (Gesneriaceae), a new species from mountain tops of Hainan Island, South China.

Project description:A new species of Gesneriaceae, Oreocharis jasminina S.J.Ling, F.Wen & M.X. Ren from Hainan Island, south China, is highlighted and described. The new species is distinguished by its actinomorphic corolla, narrow floral tube and ovate anthers hidden in the floral tube. The new species also showed clear geographic and altitudinal isolation from the three currently-recognised Oreocharis species on the Island. Molecular phylogenetic analysis, based on nuclear ITS1/2 and plastid trnL-trnF sequences, supported the delimitation of the new species, which forms a single lineage with all the other Oreocharis species from Hainan Island. The roles of geographic and floral isolation in the evolution of the new species and its affinities are discussed.

A new species of the genus <i>Micryletta</i> (Anura, Microhylidae) from Hainan Island, China.

Project description:We report on a new amphibian species of the genus Micryletta from Hainan Island, China, based on morphological and molecular analyses. The new species, Micryletta immaculata sp. nov., is diagnosed by a combination of the following morphological characters: medium-sized within genus (SVL 23.3-24.8 mm in males, n=3; 27.7-30.1 mm in females, n=2); dorsum bronze brown to reddish brown in life; dark brown spots and stripes on dorsum and flank absent; flanks largely pigmented with silver white; throat in adult males dark brown; undersides without dark patterns; outer metatarsal tubercle absent; supratympanic fold distinct; webbing between toes basal and poorly developed; tibiotarsal articulation adpressed limb reaching level of tympanum. The new species is divergent from all other congeners based on 16S rRNA gene sequences (3.0%-7.7%). Data on the natural history and male advertisement calls of the new species are provided. Following the IUCN Red List Categories and Criteria, we propose the new species to be listed as Vulnerable B1ab (iii).

(?), Q. jianfengsp. n. (??), Q. yinisp. n. (?), Q. wuzhisp. n. (??). Together with the type species of Qiongthela, Q. baishensis Xu, 2015, five species are found and described from Hainan, China, and seven species are now known in the genus.

Project description:The primitively segmented spider genus Qiongthela Xu & Kuntner, 2015 consists of seven species that are distributed in Hainan Island, China and southern Vietnam. Of the seven species, five are known from Hainan Island. In this study, four more Qiongthela species collected from Hainan Island are diagnosed and described as new to science based on morphological characters: Q. baoting sp. nov. (??), Q. qiongzhong sp. nov. (??), Q. sanya sp. nov. (??), Q. yinggezui sp. nov. (??). To facilitate future identification, the GenBank accession codes of the DNA barcode gene, cytochrome c oxidase subunit I (COI), for all the type specimens are also provided.
