ABSTRACT: Outdoor breathing trials with simulated avalanche snow are fundamental for the research of the gas exchange under avalanche snow, which supports the development of the international resuscitation guidelines. However, these studies have to face numerous problems, including unstable weather and variable snow properties. This pilot study examines a mineral material perlite as a potential snow model for studies of ventilation and gas exchange parameters. Thirteen male subjects underwent three breathing phases-into snow, wet perlite and dry perlite. The resulting trends of gas exchange parameters in all tested materials were similar and when there was a significant difference observed, the trends in the parameters for high density snow used in the study lay in between the trends in dry and wet perlite. These findings, together with its stability and accessibility year-round, make perlite a potential avalanche snow model material. Perlite seems suitable especially for simulation and preparation of breathing trials assessing gas exchange under avalanche snow, and potentially for testing of new avalanche safety equipment before their validation in real snow.The study was registered in ClinicalTrials.gov on January 22, 2018; the registration number is NCT03413878.

Project description:The world glaciers and areas of persistent summer snowpack are being lost due to warming temperatures. For cold-adapted species, habitat features may offer opportunities for cooling during summer heat yet the loss of snow and ice may compromise derived thermoregulatory benefits. Herein we offer insights about habitat selection for snow and the extent to which other behavioral adjustments reduce thermal debt among high elevation mammals. Specifically, we concentrate on respiration in mountain goats (Oreamnos americanus), a species whose native distribution is currently tied to northern mountain ranges of North America, where large patches of persistent summer snow are declining, and which became extinct during geologically warmer epochs. To examine sensitivity to possible thermal stressors and use of summer snow cover, we tracked marked and unmarked mountain goats in Glacier National Park, Montana, USA, to test hypotheses about selection for cold microclimates including shade and snow during periods of relatively high temperature. To understand functional responses of habitat choices, we measured microhabitat temperatures and a component of goat physiology-breaths per minute-as an index for metabolic expenditure. Individuals 1) selected areas closer to snow on warmer summer days, and 2) on snow had a 15% mean reduction in respiration when accounting for other factors, which suggests remnant snow plays an important role in mediating effects of air temperature. The use of shade was not as an important variable in models explaining respiration. Despite the loss of 85% of glaciers in in Glacier National Park, summer's remnant snow patches are an important reservoir by which animals reduce heat stress and potential hyperthermia. Our findings, when contextualized with behavioral strategies deployed by other high elevation mammalian taxa help frame how ambient temperatures may be modulated, and they offer a direct way by which to assess susceptibility to increasing heat in cold-adapted species.

Project description:Diffuse optical imaging (DOI) provides in vivo quantification of tissue chromophores such as oxy- and deoxyhemoglobin (HbO2 and HHb, respectively). These parameters have been shown to be useful for predicting neoadjuvant treatment response in breast cancer patients. However, most DOI devices designed for the breast are nonportable, making frequent longitudinal monitoring during treatment a challenge. Furthermore, hemodynamics related to the respiratory cycle are currently unexplored in the breast and may have prognostic value. To design, fabricate, and validate a high optode-density wearable continuous wave diffuse optical probe for the monitoring of breathing hemodynamics in breast tissue. The probe has a rigid-flex design with 16 dual-wavelength sources and 16 detectors. Performance was characterized on tissue-simulating phantoms, and validation was performed through flow phantom and cuff occlusion measurements. The breasts of N = 4 healthy volunteers were measured while performing a breathing protocol. The probe has 512 unique source-detector (S-D) pairs that span S-D separations of 10 to 54 mm. It exhibited good performance characteristics: μa drift of 0.34%/h, μa precision of 0.063%, and mean SNR ≥ 24 dB up to 41 mm S-D separation. Absorption contrast was detected in flow phantoms at depths exceeding 28 mm. A cuff occlusion measurement confirmed the ability of the probe to track expected hemodynamics in vivo. Breast measurements on healthy volunteers during paced breathing revealed median signal-to-motion artifact ratios ranging from 8.1 to 8.7 dB. Median ΔHbO2 and ΔHHb amplitudes ranged from 0.39 to 0.67 μM and 0.08 to 0.12 μM, respectively. Median oxygen saturations at the respiratory rate ranged from 82% to 87%. A wearable diffuse optical probe has been designed and fabricated for the measurement of breast tissue hemodynamics. This device is capable of quantifying breathing-related hemodynamics in healthy breast tissue.
