Prevalence and Antimicrobial Resistance of Escherichia coli, Salmonella and Vibrio Derived from Farm-Raised Red Hybrid Tilapia (Oreochromis spp.) and Asian Sea Bass (Lates calcarifer, Bloch 1970) on the West Coast of Peninsular Malaysia.

ABSTRACT: Antibiotics are widely used in intensive fish farming, which in turn increases the emergence of antimicrobial-resistant (AMR) bacteria in the aquatic environment. The current study investigates the prevalence and determines the antimicrobial susceptibility of E. coli, Salmonella, and Vibrio in farmed fishes on the west coast of Peninsular Malaysia. Over a period of 12 months, 32 aquaculture farms from the Malaysian states of Selangor, Negeri Sembilan, Melaka, and Perak were sampled. Both E. coli and Salmonella were highly resistant to erythromycin, ampicillin, tetracycline, and trimethoprim, while Vibrio was highly resistant to ampicillin and streptomycin. Resistance to the antibiotics listed as the highest priority and critically important for human therapy, such as colistin in E. coli (18.1%) and Salmonella (20%) in fish, is a growing public health concern. The multi-drug resistance (MDR) levels of E. coli and Salmonella in tilapia were 46.5% and 77.8%, respectively. Meanwhile, the MDR levels of E. coli, Salmonella, V. parahaemolyticus, V. vulnificus and V. cholerae in Asian seabass were 34%, 100%, 21.6%, 8.3% and 16.7%, respectively. Our findings provide much-needed information on AMR in aquaculture settings that can be used to tailor better strategies for the use of antibiotics in aquaculture production at the local and regional levels.

Project description:This study aims to estimate the economic loss due to vibriosis in the production of Asian seabass in floating net-cages on the east coast of Peninsular Malaysia. Asian seabass has contributed significantly to Malaysia's economic activities and food security. However, its production can be hindered by the occurrence of diseases, such as vibriosis, causing severe economic losses to farmers. A questionnaire-based survey was conducted on 14 small-scale monoculture Asian seabass net-cage farms. Using a stochastic bioeconomic model and inputs from the survey, existing literature, and expert opinion, the economic losses were determined. Moreover, this model considered the prevalence of Vibrio spp. at a farm on the east coast and the risk posed by its infection from hatcheries. The results showed that 71.09% of Asian seabass simulated in the stochastic model survived. The mortality rate due to vibriosis and other causes was at 16.23 and 12.68%, respectively. The risk posed by Vibrio spp. infection from hatcheries contributed to 2.77% of the increase in Asian seabass mortality. The stochastic model estimated that the total cost of producing a tail of Asian seabass was €2.69 per kilogram. The economic loss of vibriosis was estimated at €0.19 per tail per kilogram, which represents 7.06% of the total production cost of Asian seabass per kilogram. An increase in the prevalence of clinical vibriosis and vibriosis case fatality rate at 42 and 100%, respectively, will lead to an increase in the cost of grow-out Asian seabass by €0.29 per tail from the default value. An increase in pellet price per kilogram by €1.38 and feed conversion ratio pellet by 0.96 will consequently increase the cost of grow-out Asian seabass by €2.29 per tail and €0.82 per tail, respectively. We find that the occurrence of Vibrio spp. infection at the hatchery level can contribute to an increased risk in the mortality of Asian seabass during the grow-out phase. Hence, we also need to focus on the control and prevention of vibriosis infection from hatcheries.

Project description:BackgroundThe global crisis of antibiotic resistance increases the demand for the new promising alternative drugs such as antimicrobial peptides (AMPs). Accordingly, we have described a new, previously unrecognized effective AMP, named dicentracin-like, from Asian sea bass and characterized its antimicrobial activity by comparison with moronecidin.Methodology/ resultsGene expression analysis demonstrated the expression of dicentracin-like peptide in tissues of the immune system such as the skin and the head kidney, which is an important endocrine and lymphoid organ. Moronecidin and dicentracin-like exhibited a higher antibacterial activity against gram-positive bacteria relative to gram-negative ones, while both peptides showed a greater binding ability to gram-negative bacteria compared to gram-positive ones. This contradiction between antibacterial activity and binding affinity may be related to the outer membrane from gram-negative bacteria. Compared with moronecidin, dicentracin-like peptide showed more potent binding ability to all gram-positive and gram-negative bacteria. In addition, dicentracin-like peptide exhibited a high antibacterial activity against the investigated microorganisms, except against Staphylococcus aureus. A direct relationship was found between the binding affinity/cationicity and the antibiofilm activity of the peptides wherein, an elevation in pH corresponded to a decrease in their antibiofilm property. Time-kill kinetics analysis against clinical Acinetobacter baumannii isolate indicated that bactericidal effect of dicentracin-like and moronecidin at inhibitory concentration (1XMIC) was observed after 4 and 6 hours, respectively, while bactericidal effect of both AMPs at concentration of 2XMIC was observed after 2 hours. Dicentracin-like peptide showed higher inhibitory activity at subinhibitory concentration (1/2XMIC), relative to moronecidin. Compared with moronecidin, dicentracin-like peptide possessed greater binding affinity to bacteria at high salt concentration, as well as at alkaline pH; In addition, dicentracin-like exhibited a higher antibiofilm activity in comparison to moronecidin even at alkaline pH. Hemolytic analysis against human RBC revealed that hemolytic activity of moronecidin was more potent than that of dicentracin-like, which is consistent with its greater non-polar face hydrophobicity.ConclusionsIn the present study, In Silico comparative sequence analysis and antimicrobial characterization led to identify a new, previously unrecognized antimicrobial function for named dicentracin-like peptide by comparison with moronecidin, representing a possible template for designing new effective AMPs and improving known ones.

Project description:BackgroundProbiotics have recently been applied in aquaculture as eco-friendly alternatives to antibiotics to improve fish health, simultaneously with the increase of production parameters. The present study aimed to investigate the functional potential of lactic acid bacteria (LAB) isolated from the gut of Tilapia (Oreochromis niloticus) originating from the aquaculture farm of Oceanologic Research Center in Ivory Coast.ResultsTwelve LAB strains were identified by 16 S rDNA gene sequence homology analysis belonging to two genera Pediococcus (P. acidilactici and P. pentosaceus) and Lactobacillus (L. plantarum) with a predominance of P. acidilactici. Several aspects including functional, storage, and safety characteristics were taken into consideration in the selection process of the native LAB isolates as potential probiotics. All LAB isolates showed high antagonistic activity against bacterial pathogens like Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa, Proteus mirabilis, and Staphylococcus aureus. In addition, the LAB isolates exhibited different degrees of cell surface hydrophobicity in the presence of hexane, xylene, and chloroform as solvents and a good ability to form biofilm. The strong antioxidant activity expressed through the DPPH scavenging capacity of LAB intact cells and their cell-free supernatants was detected. LAB strains survived between 34.18% and 49.9% when exposed to low pH (1.5) and pepsin for 3 h. In presence of 0.3% bile salts, the growth rate ranged from 0.92 to 21.46%. Antibiotic susceptibility pattern of LAB isolates showed sensitivity or intermediate resistance to amoxicillin, cephalothin, chloramphenicol, imipenem, kanamycin, penicillin, rifampicin, streptomycin, tetracycline and resistance to oxacillin, gentamicin, and ciprofloxacin. No significant difference in antibiotic susceptibility pattern was observed between P. acidilactici and P. pentosaceus strains. The non-hemolytic activity was detected. Following the analysis of the enzyme profile, the ability of LAB isolates to produce either lipase or β-galactosidase or both enzymes was highlighted. Furthermore, the efficacy of cryoprotective agents was proved to be isolate-dependent, with LAB isolates having a high affinity for D-sorbitol and sucrose.ConclusionThe explored LAB strains inhibited the growth of pathogens and survived after exposure to simulated gastrointestinal tract conditions. The safety and preservative properties are desirable attributes of these new probiotic strains hence recommended for future food and feed applications.
