Light-Induced Dynamic Holography.

ABSTRACT: Holographic photomobile polymers (H-PMP) are a novel class of photomobile materials in which holograms can be optically recorded. They can be used in a large variety of applications, including optical switches and color selectors. In this work, we show one of the most important properties of the photomobile film, which is the photophobicity of the unpolymerized parts of the photomobile mixture. In order to investigate this property, we recorded a transmission phase grating on an H-PMP film, and used a different experimental technique to measure the diffraction efficiency, surface tension, and mixture properties. The results allowed for a better understanding of the mechanism of the light-controlled bending observed in these compounds.

Three-dimensional supercritical resolved light-induced magnetic holography.

Project description:In the era of big data, there exists a growing gap between data generated and storage capacity using two-dimensional (2D) magnetic storage technologies (for example, hard disk drives), because they have reached their performance saturation. 3D volumetric all-optical magnetic holography is emerging rapidly as a promising road map to realizing high-density capacity for its fast magnetization control and subwavelength magnetization volume. However, most of the reported light-induced magnetization confronts the problems of impurely longitudinal magnetization, diffraction-limited spot, and uncontrollable magnetization reversal. To overcome these challenges, we propose a novel 3D light-induced magnetic holography based on the conceptual supercritical design with multibeam combination in the 4π microscopic system. We theoretically demonstrate a 3D deep super-resolved [Formula: see text] purely longitudinal magnetization spot by focusing six coherent circularly polarized beams with two opposing high numerical aperture objectives, which allows 3D magnetic holography with a volumetric storage density of up to 1872 terabit per cubic inches. The number and locations of the super-resolved magnetization spots are controllable, and thus, desired magnetization arrays in 3D volume can be produced with properly designed phase filters. Moreover, flexible magnetization reversals are also demonstrated in multifocal arrays by using different illuminations with opposite light helicity. In addition to data storage, this magnetic holography may find applications in information security, such as identity verification for a credit card with magnetic stripe.

Incoherent color holography lattice light-sheet for subcellular imaging of dynamic structures.

Project description:The purpose of the article is to explore the need and advantages of using the incoherent color holography lattice light-sheet (ICHLLS) to provide multiwavelength quantitative monitoring of 3D cellular dynamics in live tissue to further understand complex functions of cells and cellular compartments. We have explored the use of incoherent color holography lattice light-sheet to investigate colocalization of fluorescent markers in live cells in intact tissue. Neuronal structures provide an attractive target for incoherent color holography lattice light-sheet. The cells show a complex architecture in 3D space in which signaling both between cells and within subcellular structures requires colocalization of proteins and lipids to function. During activity and over long periods it is important in understanding these signaling functions in Parkinson's, Alzheimer's and motoneuron diseases within live cells in intact tissue. As a proof of concept this article recalls the key aspects in lattice light-sheet imaging and provides a description of the incoherent detection system configuration to actively control dual diffractive lenses phase-shifting at multiple excitation wavelengths sequentially, and per each z-galvo scanning level, with extended field-of-view. The incoherent color holography lattice light-sheet system will allow simultaneous recording of multidimensional object waves that contain intensity in 3D space, phase, and wavelength information. We measure colocalization of fluorescence indicators introduced into live cells in intact neural tissue.

Quantum holography with undetected light.

Project description:Holography exploits the interference of a light field reflected/transmitted from an object with a reference beam to obtain a reconstruction of the spatial shape of the object. Classical holography techniques have been very successful in diverse areas such as microscopy, manufacturing technology, and basic science. However, detection constraints for wavelengths outside the visible range restrict the applications for imaging and sensing in general. For overcoming these detection limitations, we implement phase-shifting holography with nonclassical states of light, where we exploit quantum interference between two-photon probability amplitudes in a nonlinear interferometer. We demonstrate that it allows retrieving the spatial shape (amplitude and phase) of the photons transmitted/reflected from the object and thus obtaining an image of the object despite those photons are never detected. Moreover, there is no need to use a well-characterized reference beam, since the two-photon scheme already makes use of one of the photons as reference for holography.

Dynamic complex opto-magnetic holography.

Project description:Despite recent significant progress in real-time, large-area computer-generated holography, its memory requirements and computational loads will be hard to tackle for several decades to come with the current paradigm based on a priori calculations and bit-plane writing to a spatial light modulator. Here we experimentally demonstrate a holistic approach to serial computation and repeatable writing of computer-generated dynamic holograms without Fourier transform, using minimal amounts of computer memory. We use the ultrafast opto-magnetic recording of holographic patterns in a ferrimagnetic film with femtosecond laser pulses, driven by the on-the-fly hardware computation of a single holographic point. The intensity-threshold nature of the magnetic medium allows sub-diffraction-limited, point-by-point toggling of arbitrarily localized magnetic spots on the sample, according to the proposed circular detour-phase encoding, providing complex modulation and symmetrical suppression of upper diffractive orders and conjugated terms in holographically reconstructed 3-D images.

Precise Nanosizing with High Dynamic Range Holography.

Project description:Optical sensing is one of the key enablers of modern diagnostics. Especially label-free imaging modalities hold great promise as they eliminate labeling procedures prior to analysis. However, scattering signals of nanometric particles scale with their volume square. This unfavorable scaling makes it extremely difficult to quantitatively characterize intrinsically heterogeneous clinical samples, such as extracellular vesicles, as their signal variation easily exceeds the dynamic range of currently available cameras. Here, we introduce off-axis k-space holography that circumvents this limitation. By imaging the back-focal plane of our microscope, we project the scattering signal of all particles onto all camera pixels, thus dramatically boosting the achievable dynamic range to up to 110 dB. We validate our platform by detecting and quantitatively sizing metallic and dielectric particles over a 200 × 200 μm field of view and demonstrate that independently performed signal calibrations allow correctly sizing particles made from different materials. Finally, we present quantitative size distributions of extracellular vesicle samples.

Comparative oncology chemosensitivity assay for personalized medicine using low-coherence digital holography of dynamic light scattering from cancer biopsies.

Project description:Nearly half of cancer patients who receive standard-of-care treatments fail to respond to their first-line chemotherapy, demonstrating the pressing need for improved methods to select personalized cancer therapies. Low-coherence digital holography has the potential to fill this need by performing dynamic contrast OCT on living cancer biopsies treated ex vivo with anti-cancer therapeutics. Fluctuation spectroscopy of dynamic light scattering under conditions of holographic phase stability captures ultra-low Doppler frequency shifts down to 10 mHz caused by light scattering from intracellular motions. In the comparative preclinical/clinical trials presented here, a two-species (human and canine) and two-cancer (esophageal carcinoma and B-cell lymphoma) analysis of spectral phenotypes identifies a set of drug response characteristics that span species and cancer type. Spatial heterogeneity across a centimeter-scale patient biopsy sample is assessed by measuring multiple millimeter-scale sub-samples. Improved predictive performance is achieved for chemoresistance profiling by identifying red-shifted sub-samples that may indicate impaired metabolism and removing them from the prediction analysis. These results show potential for using biodynamic imaging for personalized selection of cancer therapy.

Intensity interferometry for holography with quantum and classical light.

Project description:As first demonstrated by Hanbury Brown and Twiss, it is possible to observe interference between independent light sources by measuring correlations in their intensities rather than their amplitudes. In this work, we apply this concept of intensity interferometry to holography. We combine a signal beam with a reference and measure their intensity cross-correlations using a time-tagging single-photon camera. These correlations reveal an interference pattern from which we reconstruct the signal wavefront in both intensity and phase. We demonstrate the principle with classical and quantum light, including a single photon. Since the signal and reference do not need to be phase-stable nor from the same light source, this technique can be used to generate holograms of self-luminous or remote objects using a local reference, thus opening the door to new holography applications.

Dynamic Tomographic Phase Microscopy by Double Six-Pack Holography.

Project description:Three-dimensional (3D) optical imaging of rapidly moving biological cells is difficult to achieve as such samples cannot be scanned over time. Here, we present a dynamic scan-free optical tomography approach for stain-free 3D imaging of biological cells using our new double six-pack tomography technique, whereby 12 off-axis holograms are captured in a single camera exposure without sacrificing resolution or field of view. The proposed system illuminates the sample from 12 angles simultaneously, and 3D refractive index (RI) tomograms are reconstructed from each recorded video frame of the dynamic sample. The technique is verified experimentally by recording flowing silica beads, 3 μm in diameter, with the resulting tomogram RI accuracy being 98.5%. A live swimming sperm cell is also imaged, and dynamic 3D imaging results for both beads and sperm cell are presented. The proposed technique represents a 12-fold increase in dynamic holographic data for tomography.

Addressable metasurfaces for dynamic holography and optical information encryption.

Project description:Metasurfaces enable manipulation of light propagation at an unprecedented level, benefitting from a number of merits unavailable to conventional optical elements, such as ultracompactness, precise phase and polarization control at deep subwavelength scale, and multifunctionalities. Recent progress in this field has witnessed a plethora of functional metasurfaces, ranging from lenses and vortex beam generation to holography. However, research endeavors have been mainly devoted to static devices, exploiting only a glimpse of opportunities that metasurfaces can offer. We demonstrate a dynamic metasurface platform, which allows independent manipulation of addressable subwavelength pixels at visible frequencies through controlled chemical reactions. In particular, we create dynamic metasurface holograms for advanced optical information processing and encryption. Plasmonic nanorods tailored to exhibit hierarchical reaction kinetics upon hydrogenation/dehydrogenation constitute addressable pixels in multiplexed metasurfaces. The helicity of light, hydrogen, oxygen, and reaction duration serve as multiple keys to encrypt the metasurfaces. One single metasurface can be deciphered into manifold messages with customized keys, featuring a compact data storage scheme as well as a high level of information security. Our work suggests a novel route to protect and transmit classified data, where highly restricted access of information is imposed.

Magnesium-Based Metasurfaces for Dual-Function Switching between Dynamic Holography and Dynamic Color Display.

Project description:Metasurface-based color display and holography have greatly advanced the state of the art display technologies. To further enrich the metasurface functionalities, recently a lot of research endeavors have been made to combine these two display functions within a single device. However, so far such metasurfaces have remained static and lack tunability once the devices are fabricated. In this work, we demonstrate a dynamic dual-function metasurface device at visible frequencies. It allows for switching between dynamic holography and dynamic color display, taking advantage of the reversible phase transition of magnesium through hydrogenation and dehydrogenation. Spatially arranged stepwise nanocavity pixels are employed to accurately control the amplitude and phase of light, enabling the generation of high-quality color prints and holograms. Our work represents a paradigm toward compact and multifunctional optical elements for future display technologies.
