Investigation of avian rotavirus infections in broiler chicks from commercial flocks with different performance efficiency indexes.

ABSTRACT: The aim of this study was to investigate and compare the frequency of occurrence of avian rotavirus (AvRV) in poultry flocks according to its Performance Efficiency Index (PEI) scores. A total of 256 individual intestinal content samples of small sized-chicks (runts) with clinical signs of Runting Stunting Syndrome (RSS) and 24 clinically healthy chicks (control) were collected from twelve flocks in southern Brazil with different PEI scores: good (n = 4, PEI mean = 365); moderate (n = 4, PEI mean = 342) or poor (n = 4, PEI mean = 319). Silver-stained polyacrylamide gel electrophoresis (ss-PAGE) was used to detect and identify the AvRV species followed by RT-PCR and sequencing of the partial VP6 gene for species confirmation. AvRV was detected in 83% (10/12) of the flocks and 23.4% (60/256) of the chicks. The electrophoretic migration patterns of viral dsRNA segments were compatible with AvRV species A (AvRV- A), D (AvRV-D) and F (AvRV-F) in 9 (15%), 18 (30%), and 33 (55%) of the positive chicks fecal samples, respectively. The AvRV species identified by ss-PAGE were confirmed by RT-PCR and partial sequence analysis of the VP6 gene. The AvRV detection rate was statistically higher (p = 0.007) in chicks from flocks with poor PEI when compared to those with good PEI. The occurrence of AvRV-D and AvRV-F was statistically higher in 7 to 9 days old chicks, while AvRV-A was detected only in 13 to 14 days old animals.

Project description:BackgroundAvian leukosis virus Subgroup-J (ALV-J) is a rapidly oncogenic evolving retrovirus infecting a variety of avian species; causing severe economic losses to the local poultry industry.MethodsTo investigate ALV-J, a total of 117 blood samples and 57 tissue specimens of different organs were collected for virological, and pathological identification, serological examinations, molecular characterization, and sequencing analysis. To the best of our knowledge, this is the first detailed report recorded in broiler flocks in Egypt. The present study targets the prevalence of a viral tumor disease circulating in broiler flocks in the El-Sharqia, El-Dakahliya, and Al-Qalyubiyya Egyptian governorates from 2021 to 2023 using different diagnostic techniques besides ALV-J gp85 genetic diversity determination.ResultWe first isolated ALV-J on chicken embryo rough cell culture; showing aggregation, rounding, and degeneration. Concerning egg inoculation, embryonic death, stunting, and curling were observed. Only 79 serum samples were positive for ALV-J (67.52%) based on the ELISA test. Histopathological investigation showed tumors consist of uniform masses, usually well-differentiated myelocytes, lymphoid cells, or both in the liver, spleen, and kidneys. Immunohistochemical examination showed that the myelocytomatosis-positive signals were in the spleen, liver, and kidney. The PCR assay of ALV-J gp85 confirmed 545 base pairs with only 43 positive samples (75.4%). Two positive samples were sequenced and submitted to the Genbank with accession numbers (OR509852-OR509853). Phylogenetic analysis based on the gp85 gene showed that the ALV-J Dakahlia-2 isolate is genetically related to ALV-EGY/YA 2021.3, ALV-EGY/YA 2021.4, ALV-EGY/YA 2021.14, and ALV-EGY/YA 2021.9 with amino acid identity percentage 96%, 97%; 96%, 96%; respectively. Furthermore, ALV-J Sharqia-1 isolate is highly genetically correlated to ALV-EGY/YA 2021.14, and ALV-EGY/YA 2021.9, ALV-J isolate QL1, ALV-J isolate QL4, ALV-J isolate QL3, ALV-EGY/YA 2021.4 with amino acid identity percentage 97%, 97%; 98%, 97%, 97%, 95%; respectively.ConclusionsThis study confirmed that ALV-J infection had still been prevalent in broilers in Egypt, and the genetic characteristics of the isolates are diverse.

Project description:Salmonella Heidelberg (SH) is responsible for economic losses in poultry farming and food infections in humans and is a serious public health problem. Recently, there has been an increase in the frequency of isolation of this serotype in batches of broilers raised in Brazil. It is necessary to find new ways to help control this pathogen. The present study aimed to evaluate the effect of diets containing the compound Original XPC, which is a prebiotic-like fermented compound (PFC), and/or Sangrovit, which is a sanguinarine-based phytobiotic (SAN), on SH infection in broiler chicks. For this purpose, SH colonization in the cecum and its invasion into the spleen and liver were evaluated, as were the histopathological changes caused in these organs. The lowest cecal SH counts were observed in birds that ingested SAN, followed by those fed PFC (P < 0.05), with no added effect when the two bioproducts were used together (SAN + PFC). The mean SH and liver spleen counts did not differ between groups (P > 0.05). In general, birds from all groups challenged with SH showed similar macroscopic changes, such as hemorrhagic areas, hepatomegaly, and splenomegaly, such changes being more intense in the infected control group. The microscopic changes observed in the liver included hepatocyte congestion, heterophil infiltration in the sinusoid capillaries, areas of necrosis, and mononuclear inflammation. In the cecum, heterophilic infiltrate and thickening of the lamina propria were observed. In the ileum, the most common changes were congestion and thickening of the lamina propria and atrophy of the villi and crypts. The microscopic changes were less intense in the supplemented birds than the infected control group, and those supplemented with SAN developed the least changes. As ideal conditions for histomorphometric parameters of the ileum, the villus:crypt ratio in birds should be high, the villi should be long, and the crypts should be shallow. In the present study, higher mean heights and villus areas were observed in uninfected control and SAN group birds, and the crypt depth was lower in birds in the negative control group. The lowest villus:crypt ratio was observed in the birds of the infected control group. Although additional studies are needed, the preliminary results of the current investigation indicated that the addition of bioproducts, especially SAN, to the diet of birds helped to control SH infection, reducing its count in the cecum and improving overall and intestinal health.
