Coding of chromatic spatial contrast by macaque V1 neurons.

ABSTRACT: Color perception relies on comparisons between adjacent lights, but how the brain performs these comparisons is poorly understood. To elucidate the underlying mechanisms, we recorded spiking responses of individual V1 neurons in macaque monkeys to pairs of stimuli within the classical receptive field (RF). We estimated the spatial-chromatic RF of each neuron and then presented customized colored edges using a closed-loop technique. We found that many double-opponent (DO) cells, which have spatially and chromatically opponent RFs, responded to chromatic contrast as a weighted sum, akin to how other V1 neurons responded to luminance contrast. Yet other neurons integrated chromatic signals nonlinearly, confirming that linear signal integration is not an obligate property of V1 neurons. The functional similarity of cone-opponent DO cells and cone non-opponent simple cells suggests that these two groups may share a common underlying circuitry, promotes the construction of image-computable models for full-color image representation, and sheds new light on V1 complex cells.

Comparison of spatial summation properties of neurons in macaque V1 and V2.

Project description:In visual cortex, responses to stimulation of the receptive field (RF) are modulated by simultaneous stimulation of the RF surround. The mechanisms for surround modulation remain unidentified. We previously proposed that in the primary visual cortex (V1), near surround modulation is mediated by geniculocortical and horizontal connections and far surround modulation by interareal feedback connections. To understand spatial integration in the secondary visual cortex (V2) and its underlying circuitry, we have characterized spatial summation in different V2 layers and stripe compartments and compared it to that in V1. We used grating stimuli in circular and annular apertures of different sizes to estimate the extent and sensitivity of RF and surround components in V1 and V2. V2 RFs and surrounds were twice as large as those in V1. As in V1, V2 RFs doubled in size when measured at low contrast. In both V1 and V2, surrounds were about fivefold the size of the RF and the far surround could exceed 12.5 degrees in radius, averaging 5.5 degrees in V1 and 9.2 degrees in V2. The strength of surround suppression was similar in both areas. Thus although differing in spatial scale, the interactions among RF components are similar in V1 and V2, suggesting similar underlying mechanisms. As in V1, the extent of V2 horizontal connections matches that of the RF center, but is much smaller than the largest far surrounds, which likely derive from interareal feedback. In V2, we found no laminar or stripe differences in size and magnitude of surround suppression, suggesting conservation across stripes of the basic circuit for surround modulation.

Contrast response in a comprehensive network model of macaque V1.

Project description:The response to contrast is one of the most important functions of the macaque primary visual cortex, V1, but up to now there has not been an adequate theory for it. To fill this gap in our understanding of cortical function, we built and analyzed a new large-scale, biologically constrained model of the input layer, 4Cα, of macaque V1. We called the new model CSY2. We challenged CSY2 with a three-parameter family of visual stimuli that varied in contrast, orientation, and spatial frequency. CSY2 accurately simulated experimental data and made many new predictions. It accounted for 1) the shapes of firing-rate-versus-contrast functions, 2) orientation and spatial frequency tuning versus contrast, and 3) the approximate contrast-invariance of cortical activity maps. Post-analysis revealed that the mechanisms that were needed to produce the successful simulations of contrast response included strong recurrent excitation and inhibition that find dynamic equilibria across the cortical surface, dynamic feedback between L6 and L4, and synaptic dynamics like inhibitory synaptic depression.

Chromatic detection from cone photoreceptors to V1 neurons to behavior in rhesus monkeys.

Project description:Chromatic sensitivity cannot exceed limits set by noise in the cone photoreceptors. To determine how close neurophysiological and psychophysical chromatic sensitivity come to these limits, we developed a parameter-free model of stimulus encoding in the cone outer segments, and we compared the sensitivity of the model to the psychophysical sensitivity of monkeys performing a detection task and to the sensitivity of individual V1 neurons. Modeled cones had a temporal impulse response and a noise power spectrum that were derived from in vitro recordings of macaque cones, and V1 recordings were made during performance of the detection task. The sensitivity of the simulated cone mosaic, the V1 neurons, and the monkeys were tightly yoked for low-spatiotemporal-frequency isoluminant modulations, indicating high-fidelity signal transmission for this class of stimuli. Under the conditions of our experiments and the assumptions for our model, the signal-to-noise ratio for these stimuli dropped by a factor of ?3 between the cones and perception. Populations of weakly correlated V1 neurons narrowly exceeded the monkeys' chromatic sensitivity but fell well short of the cones' chromatic sensitivity, suggesting that most of the behavior-limiting noise lies between the cone outer segments and the output of V1. The sensitivity gap between the cones and behavior for achromatic stimuli was larger than for chromatic stimuli, indicating greater postreceptoral noise. The cone mosaic model provides a means to compare visual sensitivity across disparate stimuli and to identify sources of noise that limit visual sensitivity.

The orientation selectivity of color-responsive neurons in macaque V1.

Project description:Form has a strong influence on color perception. We investigated the neural basis of the form-color link in macaque primary visual cortex (V1) by studying orientation selectivity of single V1 cells for pure color patterns. Neurons that responded to color were classified, based on cone inputs and spatial selectivity, into chromatically single-opponent and double-opponent groups. Single-opponent cells responded well to color but weakly to luminance contrast; they were not orientation selective for color patterns. Most double-opponent cells were orientation selective to pure color stimuli as well as to achromatic patterns. We also found non-opponent cells that responded weakly or not at all to pure color; most were orientation selective for luminance patterns. Double-opponent and non-opponent cells' orientation selectivities were not contrast invariant; selectivity usually increased with contrast. Double-opponent cells were approximately equally orientation selective for luminance and equiluminant color stimuli when stimuli were matched in average cone contrast. V1 double-opponent cells could be the neural basis of the influence of form on color perception. The combined activities of single- and double-opponent cells in V1 are needed for the full repertoire of color perception.

Functional interactions among neurons within single columns of macaque V1.

Project description:Recent developments in high-density neurophysiological tools now make it possible to record from hundreds of single neurons within local, highly interconnected neural networks. Among the many advantages of such recordings is that they dramatically increase the quantity of identifiable, functional interactions between neurons thereby providing an unprecedented view of local circuits. Using high-density, Neuropixels recordings from single neocortical columns of primary visual cortex in nonhuman primates, we identified 1000s of functionally interacting neuronal pairs using established crosscorrelation approaches. Our results reveal clear and systematic variations in the synchrony and strength of functional interactions within single cortical columns. Despite neurons residing within the same column, both measures of interactions depended heavily on the vertical distance separating neuronal pairs, as well as on the similarity of stimulus tuning. In addition, we leveraged the statistical power afforded by the large numbers of functionally interacting pairs to categorize interactions between neurons based on their crosscorrelation functions. These analyses identified distinct, putative classes of functional interactions within the full population. These classes of functional interactions were corroborated by their unique distributions across defined laminar compartments and were consistent with known properties of V1 cortical circuitry, such as the lead-lag relationship between simple and complex cells. Our results provide a clear proof-of-principle for the use of high-density neurophysiological recordings to assess circuit-level interactions within local neuronal networks.

A quadratic model captures the human V1 response to variations in chromatic direction and contrast.

Project description:An important goal for vision science is to develop quantitative models of the representation of visual signals at post-receptoral sites. To this end, we develop the quadratic color model (QCM) and examine its ability to account for the BOLD fMRI response in human V1 to spatially uniform, temporal chromatic modulations that systematically vary in chromatic direction and contrast. We find that the QCM explains the same, cross-validated variance as a conventional general linear model, with far fewer free parameters. The QCM generalizes to allow prediction of V1 responses to a large range of modulations. We replicate the results for each subject and find good agreement across both replications and subjects. We find that within the LM cone contrast plane, V1 is most sensitive to L-M contrast modulations and least sensitive to L+M contrast modulations. Within V1, we observe little to no change in chromatic sensitivity as a function of eccentricity.

Spatiotemporal functional organization of excitatory synaptic inputs onto macaque V1 neurons.

Project description:The integration of synaptic inputs onto dendrites provides the basis for neuronal computation. Whereas recent studies have begun to outline the spatial organization of synaptic inputs on individual neurons, the underlying principles related to the specific neural functions are not well understood. Here we perform two-photon dendritic imaging with a genetically-encoded glutamate sensor in awake monkeys, and map the excitatory synaptic inputs on dendrites of individual V1 superficial layer neurons with high spatial and temporal resolution. We find a functional integration and trade-off between orientation-selective and color-selective inputs in basal dendrites of individual V1 neurons. Synaptic inputs on dendrites are spatially clustered by stimulus feature, but functionally scattered in multidimensional feature space, providing a potential substrate of local feature integration on dendritic branches. Furthermore, apical dendrite inputs have larger receptive fields and longer response latencies than basal dendrite inputs, suggesting a dominant role for apical dendrites in integrating feedback in visual information processing.

Perceptual learning improves contrast sensitivity of V1 neurons in cats.

Project description:Perceptual learning has been documented in adult humans over a wide range of tasks. Although the often-observed specificity of learning is generally interpreted as evidence for training-induced plasticity in early cortical areas, physiological evidence for training-induced changes in early visual cortical areas is modest, despite reports of learning-induced changes of cortical activities in fMRI studies. To reveal the physiological bases of perceptual learning, we combined psychophysical measurements with extracellular single-unit recording under anesthetized preparations and examined the effects of training in grating orientation identification on both perceptual and neuronal contrast sensitivity functions of cats.We have found that training significantly improved perceptual contrast sensitivity of the cats to gratings with spatial frequencies near the "trained" spatial frequency, with stronger effects in the trained eye. Consistent with behavioral assessments, the mean contrast sensitivity of neurons recorded from V1 of the trained cats was significantly higher than that of neurons recorded from the untrained cats. Furthermore, in the trained cats, the contrast sensitivity of V1 neurons responding preferentially to stimuli presented via the trained eyes was significantly greater than that of neurons responding preferentially to stimuli presented via the "untrained" eyes. The effect was confined to the trained spatial frequencies. In both trained and untrained cats, the neuronal contrast sensitivity functions derived from the contrast sensitivity of the individual neurons were highly correlated with behaviorally determined perceptual contrast sensitivity functions.We suggest that training-induced neuronal contrast gain in area V1 underlies behaviorally determined perceptual contrast sensitivity improvements.

Attention alters spatial integration in macaque V1 in an eccentricity-dependent manner.

Project description:Attention can selectively enhance neuronal responses and exclude external noise, but the neuronal computations that underlie these effects remain unknown. At the neuronal level, noise exclusion might result in altered spatial integration properties. We tested this proposal by recording neuronal activity and length tuning in neurons of the primary visual cortex of the macaque when attention was directed toward or away from stimuli presented in each neuron's classical receptive field. For cells with central-parafoveal receptive fields, attention reduced spatial integration, as demonstrated by a reduction in preferred stimulus length and in the size of the spatial summation area. Conversely, in cells that represented more peripheral locations, attention increased spatial integration by increasing the cell's summation area. This previously unknown dichotomy between central and peripheral vision could support accurate analysis of attended foveal objects and target selection for impending eye movements to peripheral objects.

Contrast-dependence of surround suppression in Macaque V1: experimental testing of a recurrent network model.

Project description:Neuronal responses in primary visual cortex (V1) to optimally oriented high-contrast stimuli in the receptive field (RF) center are suppressed by stimuli in the RF surround, but can be facilitated when the RF center is stimulated at low contrast. The neural circuits and mechanisms for surround modulation are still unknown. We previously proposed that topdown feedback connections mediate suppression from the "far" surround, while "near' surround suppression is mediated primarily by horizontal connections. We implemented this idea in a recurrent network model of V1. A model assumption needed to account for the contrast-dependent sign of surround modulation is a response asymmetry between excitation and inhibition; accordingly, inhibition, but not excitation, is silent for weak visual inputs to the RF center, and surround stimulation can evoke facilitation. A prediction stemming from this same assumption is that surround suppression is weaker for low than for high contrast stimuli in the RF center. Previous studies are inconsistent with this prediction. Using single unit recordings in macaque V1, we confirm this model's prediction. Model simulations demonstrate that our results can be reconciled with those from previous studies. We also performed a systematic comparison of the experimentally measured surround suppression strength with predictions of the model operated in different parameter regimes. We find that the original model, with strong horizontal and no feedback excitation of local inhibitory neurons, can only partially account quantitatively for the experimentally measured suppression. Strong direct feedback excitation of V1 inhibitory neurons is necessary to account for the experimentally measured surround suppression strength.
