Reduced neural activity but improved coding in rodent higher-order visual cortex during locomotion.

ABSTRACT: Running profoundly alters stimulus-response properties in mouse primary visual cortex (V1), but its effect in higher-order visual cortex is under-explored. Here we systematically investigate how visual responses vary with locomotive state across six visual areas and three cortical layers using a massive dataset from the Allen Brain Institute. Although previous work has shown running speed to be positively correlated with neural activity in V1, here we show that the sign of correlations between speed and neural activity varies across extra-striate cortex, and is even negative in anterior extra-striate cortex. Nevertheless, across all visual cortices, neural responses can be decoded more accurately during running than during stationary periods. We show that this effect is not attributable to changes in population activity structure, and propose that it instead arises from an increase in reliability of single-neuron responses during locomotion.

Higher order visual areas enhance stimulus responsiveness in mouse primary visual cortex.

Project description:Over the past few years, the various areas that surround the primary visual cortex (V1) in the mouse have been associated with many functions, ranging from higher order visual processing to decision-making. Recently, some studies have shown that higher order visual areas influence the activity of the primary visual cortex, refining its processing capabilities. Here, we studied how in vivo optogenetic inactivation of two higher order visual areas with different functional properties affects responses evoked by moving bars in the primary visual cortex. In contrast with the prevailing view, our results demonstrate that distinct higher order visual areas similarly modulate early visual processing. In particular, these areas enhance stimulus responsiveness in the primary visual cortex, by more strongly amplifying weaker compared with stronger sensory-evoked responses (for instance specifically amplifying responses to stimuli not moving along the direction preferred by individual neurons) and by facilitating responses to stimuli entering the receptive field of single neurons. Such enhancement, however, comes at the expense of orientation and direction selectivity, which increased when the selected higher order visual areas were inactivated. Thus, feedback from higher order visual areas selectively amplifies weak sensory-evoked V1 responses, which may enable more robust processing of visual stimuli.

Performance-optimized hierarchical models predict neural responses in higher visual cortex.

Project description:The ventral visual stream underlies key human visual object recognition abilities. However, neural encoding in the higher areas of the ventral stream remains poorly understood. Here, we describe a modeling approach that yields a quantitatively accurate model of inferior temporal (IT) cortex, the highest ventral cortical area. Using high-throughput computational techniques, we discovered that, within a class of biologically plausible hierarchical neural network models, there is a strong correlation between a model's categorization performance and its ability to predict individual IT neural unit response data. To pursue this idea, we then identified a high-performing neural network that matches human performance on a range of recognition tasks. Critically, even though we did not constrain this model to match neural data, its top output layer turns out to be highly predictive of IT spiking responses to complex naturalistic images at both the single site and population levels. Moreover, the model's intermediate layers are highly predictive of neural responses in the V4 cortex, a midlevel visual area that provides the dominant cortical input to IT. These results show that performance optimization--applied in a biologically appropriate model class--can be used to build quantitative predictive models of neural processing.

Degradation of neural representations in higher visual cortex by sleep deprivation.

Project description:A night of total sleep deprivation (TSD) impairs selective attention and is accompanied by attenuated activation within ventral visual cortex (VVC). However, finer details of how TSD compromises selectivity of visual processing remain unclear. Drawing from prior work in cognitive aging, we predicted that TSD would result in dedifferentiation of neural responses for faces and houses within the VVC. Instead, we found preservation of category selectivity. This was observed both in voxels highly selective for each category, and also across multiple voxels evaluated using MVPA. Based on prior findings of impaired attentional modulation following TSD, we also predicted reduced biasing of neural representations towards the attended category when participants viewed ambiguous face/house images. When participants were well rested, attention to houses (or faces) caused activation patterns to more closely resemble those elicited by isolated house (face) images than face (house) images. During TSD, attention to faces enhanced neural similarity to both target (face) and distractor (house) representations, signifying reduced suppression of irrelevant information. Degraded sensory processing reflected in reduced VVC activation following TSD, thus appears to be a result of impaired top-down modulation of sensory representations instead of degraded selectivity of maximally category sensitive voxels, or the dedifferentiation of neural activation patterns.

Integration of visual motion and locomotion in mouse visual cortex.

Project description:Successful navigation through the world requires accurate estimation of one's own speed. To derive this estimate, animals integrate visual speed gauged from optic flow and run speed gauged from proprioceptive and locomotor systems. The primary visual cortex (V1) carries signals related to visual speed, and its responses are also affected by run speed. To study how V1 combines these signals during navigation, we recorded from mice that traversed a virtual environment. Nearly half of the V1 neurons were reliably driven by combinations of visual speed and run speed. These neurons performed a weighted sum of the two speeds. The weights were diverse across neurons, and typically positive. As a population, V1 neurons predicted a linear combination of visual and run speeds better than either visual or run speeds alone. These data indicate that V1 in the mouse participates in a multimodal processing system that integrates visual motion and locomotion during navigation.

Distinct recruitment of feedforward and recurrent pathways across higher-order areas of mouse visual cortex.

Project description:Cortical visual processing transforms features of the external world into increasingly complex and specialized neuronal representations. These transformations arise in part through target-specific routing of information; however, within-area computations may also contribute to area-specific function. Here, we sought to determine whether higher order visual cortical areas lateromedial (LM), anterolateral (AL), posteromedial (PM), and anteromedial (AM) have specialized anatomical and physiological properties by using a combination of whole-cell recordings and optogenetic stimulation of primary visual cortex (V1) axons in vitro. We discovered area-specific differences in the strength of recruitment of interneurons through feedforward and recurrent pathways, as well as differences in cell-intrinsic properties and interneuron densities. These differences were most striking when comparing across medial and lateral areas, suggesting that these areas have distinct profiles for net excitability and integration of V1 inputs. Thus, cortical areas are not defined simply by the information they receive but also by area-specific circuit properties that enable specialized filtering of these inputs.

Locomotion controls spatial integration in mouse visual cortex.

Project description:Growing evidence indicates that responses in sensory cortex are modulated by factors beyond direct sensory stimulation. In primary visual cortex (V1), for instance, responses increase with locomotion. Here we show that this increase is accompanied by a profound change in spatial integration. We recorded from V1 neurons in head-fixed mice placed on a spherical treadmill. We characterized spatial integration and found that the responses of most neurons were suppressed by large stimuli. As in primates, this surround suppression increased with stimulus contrast. These effects were captured by a divisive normalization model, where the numerator originates from a central region driving the neuron and the denominator originates from a larger suppressive field. We then studied the effects of locomotion and found that it markedly reduced surround suppression, allowing V1 neurons to integrate over larger regions of visual space. Locomotion had two main effects: it increased spontaneous activity, and it weakened the suppressive signals mediating normalization, relative to the driving signals. We conclude that a fundamental aspect of visual processing, spatial integration, is controlled by an apparently unrelated factor, locomotion. This control might operate through the mechanisms that are in place to deliver surround suppression.

Higher-order neural processing tunes motion neurons to visual ecology in three species of hawkmoths.

Project description:To sample information optimally, sensory systems must adapt to the ecological demands of each animal species. These adaptations can occur peripherally, in the anatomical structures of sensory organs and their receptors; and centrally, as higher-order neural processing in the brain. While a rich body of investigations has focused on peripheral adaptations, our understanding is sparse when it comes to central mechanisms. We quantified how peripheral adaptations in the eyes, and central adaptations in the wide-field motion vision system, set the trade-off between resolution and sensitivity in three species of hawkmoths active at very different light levels: nocturnal Deilephila elpenor, crepuscular Manduca sexta, and diurnal Macroglossum stellatarum. Using optical measurements and physiological recordings from the photoreceptors and wide-field motion neurons in the lobula complex, we demonstrate that all three species use spatial and temporal summation to improve visual performance in dim light. The diurnal Macroglossum relies least on summation, but can only see at brighter intensities. Manduca, with large sensitive eyes, relies less on neural summation than the smaller eyed Deilephila, but both species attain similar visual performance at nocturnal light levels. Our results reveal how the visual systems of these three hawkmoth species are intimately matched to their visual ecologies.

Neural Coding for Instruction-Based Task Sets in Human Frontoparietal and Visual Cortex.

Project description:Task preparation has traditionally been thought to rely upon persistent representations of instructions that permit their execution after delays. Accumulating evidence suggests, however, that accurate retention of task knowledge can be insufficient for successful performance. Here, we hypothesized that instructed facts would be organized into a task set; a temporary coding scheme that proactively tunes sensorimotor pathways according to instructions to enable highly efficient "reflex-like" performance. We devised a paradigm requiring either implementation or memorization of novel stimulus-response mapping instructions, and used multivoxel pattern analysis of neuroimaging data to compare neural coding of instructions during the pretarget phase. Although participants could retain instructions under both demands, we observed striking differences in their representation. To-be-memorized instructions could only be decoded from mid-occipital and posterior parietal cortices, consistent with previous work on visual short-term memory storage. In contrast, to-be-implemented instructions could also be decoded from frontoparietal "multiple-demand" regions, and dedicated visual areas, implicated in processing instructed stimuli. Neural specificity in the latter moreover correlated with performance speed only when instructions were prepared, likely reflecting the preconfiguration of instructed decision circuits. Together, these data illuminate how the brain proactively optimizes performance, and help dissociate neural mechanisms supporting task control and short-term memory storage.

The neural origin for asymmetric coding of surface color in the primate visual cortex.

Project description:The coding privilege of end-spectral hues (red and blue) in the early visual cortex has been reported in primates. However, the origin of such bias remains unclear. Here, we provide a complete picture of the end-spectral bias in visual system by measuring fMRI signals and spiking activities in macaques. The correlated end-spectral biases between the LGN and V1 suggest a subcortical source for asymmetric coding. Along the ventral pathway from V1 to V4, red bias against green peaked in V1 and then declined, whereas blue bias against yellow showed an increasing trend. The feedforward and recurrent modifications of end-spectral bias were further revealed by dynamic causal modeling analysis. Moreover, we found that the strongest end-spectral bias in V1 was in layer 4C[Formula: see text]. Our results suggest that end-spectral bias already exists in the LGN and is transmitted to V1 mainly through the parvocellular pathway, then embellished by cortical processing.

Running modulates primate and rodent visual cortex differently.

Project description:When mice run, activity in their primary visual cortex (V1) is strongly modulated. This observation has altered conceptions of a brain region assumed to be a passive image processor. Extensive work has followed to dissect the circuits and functions of running-correlated modulation. However, it remains unclear whether visual processing in primates might similarly change during locomotion. We therefore measured V1 activity in marmosets while they viewed stimuli on a treadmill. In contrast to mouse, running-correlated modulations of marmoset V1 were small and tended to be slightly suppressive. Population-level analyses revealed trial-to-trial fluctuations of shared gain across V1 in both species, but while strongly correlated with running in mice, gain modulations were smaller and more often negatively correlated with running in marmosets. Thus, population-wide fluctuations of V1 may reflect a common feature of mammalian visual cortical function, but important quantitative differences point to distinct consequences for the relation between vision and action in primates versus rodents.
