Ionic liquid stationary phase coating optimization for semi-packed microfabricated columns.

ABSTRACT: This work highlights the effect of the stationary phase coating process on the separation efficiency of gas chromatography microcolumns. The stationary phase coating quality was characterized by three different bis(trifluoromethylsulfonyl)imide (NTf₂) anion based ionic liquids. The ionic liquids containing NTf₂ anion are used for gas chromatography due to their high temperature stability. In this work, the chemical and physical approaches of column deactivation as well as the temperature treatment were evaluated by separating a mixture of 20 organic components and saturated alkanes. The results show that higher oven temperature treatment provides higher efficiency while losing a bit of peak symmetry. The thermal treated 1-butylpyridinum bis(trifluoromethylsulfonyl) imide [BPY][NTf₂] stationary phase at 240°C demonstrated as high as 8300 plates per meter for naphthalene. This was a 5-fold increase in separation efficiency in comparison to those of the columns treated at 200°C. Albeit being within acceptable ranges, the peak tailing degraded from 1.17 to 1.46 for naphthalene when the processing temperature for coating increased. Both chemical and physical deactivation process increased separation efficiencies and peak resolution.

Project description:Protein glycosylation is a common post-translational modification that modulates biological processes such as the immune response and protein trafficking. Altered glycosylation profiles are associated with cancer and inflammatory diseases, as well as impacting the efficacy of therapeutic monoclonal antibodies. Consisting of oligosaccharides attached to asparagine residues, enzymatically released N-linked glycans are analytically challenging due to the diversity of isomeric structures that exist. A commonly used technique for quantitative N-glycan analysis is liquid chromatography-mass spectrometry (LC-MS), which performs glycan separation and characterization. Although many reversed and normal stationary phases have been utilized for the separation of N-glycans, porous graphitic carbon (PGC) chromatography has become desirable because of its higher resolving capability, but is difficult to implement in a robust and reproducible manner. Herein, we demonstrate the analytical properties of a 15 cm fused silica capillary (75 µm i.d., 360 µm o.d.) packed in-house with Hypercarb PGC (3 µm) coupled to an Agilent 6550 Q-TOF mass spectrometer for N-glycan analysis in positive ion mode. In repeatability and intermediate precision measurements conducted on released N-glycans from a glycoprotein standard mixture, the majority of N-glycans reported low coefficients of variation with respect to retention times (≤4.2%) and peak areas (≤14.4%). N-glycans released from complex samples were also examined by PGC LC-MS. A total of 120 N-glycan structural and compositional isomers were obtained from formalin-fixed paraffin-embedded ovarian cancer tissue sections. Finally, a comparison between early- and late-stage formalin-fixed paraffin-embedded ovarian cancer tissues revealed qualitative changes in the α2,3- and α2,6-sialic acid linkage of a fucosylated bi-antennary complex N-glycan. Although the α2,3-linkage was predominant in late-stage ovarian cancer, the alternate α2,6-linkage was more prevalent in early-stage ovarian cancer. This study establishes the utility of in-house packed PGC columns for the robust and reproducible LC-MS analysis of N-glycans.

Project description:In vivo multi-electrode arrays (MEAs) can sense electrical signals from a small set of neurons or modulate neural activity through micro-stimulation. Electrode's geometric surface area (GSA) and impedance are important for both unit recording and neural stimulation. Smaller GSA is preferred due to enhanced selectivity of neural signal, but it tends to increase electrode impedance. Higher impedance leads to increased electrical noise and signal loss in single unit neural recording. It also yields a smaller charge injection window for safe neural stimulation. To address these issues, poly (3, 4-ethylenedioxythiophene) - ionic liquid (PEDOT-IL) conducting polymers were electrochemically polymerized on the surface of the microelectrodes. The PEDOT-IL coating reduced the electrode impedance modulus by over 35 times at 1 kHz. It also exhibited compelling nanostructure in surface morphology and significant impedance reduction in other physiologically relevant range (100Hz-1000Hz). PEDOT-IL coated electrodes exhibited a Charge Storage Capacity (CSC) that was about 20 times larger than that of bare electrodes. The neural recording performance of PEDOT-IL coated electrodes was also compared with uncoated electrodes and PEDOT-poly (styrenesulfonate) (PSS) coated electrodes in rat barrel cortex (SI). Spontaneous neural activity and sensory evoked neural response were utilized for characterizing the electrode performance. The PEDOT-IL electrodes exhibited a higher unit yield and signal-to-noise ratio (SNR) in vivo. The local field potential recording was benefited from the low impedance PEDOT-IL coating in noise and artifact reduction as well. Moreover, cell culture on PEDOT-IL coating demonstrated that the material is safe for neural tissue and reduces astrocyte fouling. Taken together, PEDOT-IL coating has the potential to benefit neural recording and stimulation electrodes, especially when integrated with novel small GSA electrode arrays designed for high recording density, minimal insertion damage and alleviated tissue reaction.
