TDDFT-ECD and DFT-NMR studies of thaigranatins A-E and granatumin L isolated from Xylocarpus granatum.

ABSTRACT: TDDFT-ECD calculations were utilized to explain the mirror image or different ECD spectra of previously reported homochiral natural products thaigranatins A-E and granatumin L, the simple comparison of which would result in a wrong stereochemical conclusion. The configurational assignment was confirmed independently and geometrical parameters of the chromophores governing the ECD spectra were identified in the structurally related natural products by analyzing the ECD spectra and geometries of the low-energy computed conformers obtained by different methods. Different conformations of the furan-2-yl-δ-lactone subunit were found responsible for the mirror image ECD spectra of the homochiral thaigranatins C-E. Two DFT ¹³C NMR chemical shift calculation methods and DP4+ analysis were performed on the C-6 epimers of thaigranatin D, which together with the ECD calculation, could determine the absolute configuration of C-6 as (R).

Glucopyranosylidene-Spiro-Thiazolinones: Synthetic Studies and Determination of Absolute Configuration by TDDFT-ECD Calculations.

Project description:Reactions of O-peracylated C-(1-bromo-β-d-glucopyranosyl)formamides with thioamides furnished the corresponding glucopyranosylidene-spiro-thiazolin-4-one. While O-debenzoylations under a variety of conditions resulted in decomposition, during O-deacetylations the addition of MeOH to the thiazolinone moiety was observed, and with EtOH and water similar adducts were isolated or detected. The structure and stereochemistry of the new compounds were established by means of NMR and electronic circular dichroism (ECD) data supported by time-dependent density functional theory ECD (TDDFT-ECD) calculations. TDDFT-ECD calculations could efficiently distinguish the proposed epimeric products having different absolute configuration in the spiro heterocyclic ring.

Novel and Neuroprotective Tetranortriterpenoids from Chinese Mangrove Xylocarpus granatum Koenig.

Project description:Eight new tetranortriterpenoids (1-8) were isolated from the twigs and leaves of the Chinese mangrove plant Xylocarpus granatum, together with four related known ones (9-12). The structures of new compounds were elucidated by detailed spectroscopic analysis. The absolute configuration of 9-epixylogranatin A (1) was determined by time-dependent density functional theory-electronic circular dichroism (TDDFT-ECD) calculations of the solution conformers. Xylogranatumin A (2) represents the first example of the 9, 10-seco limonoid with an unprecedented oxygen-bridged B ring (2,7-dioxabicyclo[2.2.1]-heptane). All the isolates were evaluated for the in vitro neuroprotective activity, both compounds 11 and 12 displayed moderate effects against H2O2-induced neurotoxicity in PC12 cells at the concentration of 10 μM, with an increase in cell viability of 12.0% and 11.6%, respectively.

Structure Revision of Formyl Phloroglucinol Meroterpenoids: A Unified Approach Using NMR Fingerprinting and DFT NMR and ECD Analyses.

Project description:NMR fingerprints are valuable tools for analyzing complex natural product mixtures and identifying incorrectly assigned structures in the literature. Our diagnostic NMR fingerprints for formyl phloroglucinol meroterpenoids revealed discrepancies in the structures reported for eucalyprobusal C (1a) and eucalypcamal K (2a). NMR fingerprinting PCA analyses identified 1a as an oxepine-diformyl phloroglucinol and 2a as an oxepine 3-acyl-1-formyl phloroglucinol, contrary to their initial assignments as pyrano-diformyl and pyrano 3-acyl-1-formyl phloroglucinols, respectively. Extensive reinterpretation of their reported one- and two-dimensional NMR data, coupled with GIAO DFT-calculated 1H and 13C NMR chemical shift and DP4+ analyses, supported the unequivocal reassignment of eucalyprobusal C to 1b and eucalypcamal K to 2b. The absolute configurations of the revised oxepine-containing phloroglucinol meroterpenoids were confirmed via the reinterpretation of their reported ROESY and NOESY NMR data, along with comparative TDDFT-calculated and experimental ECD spectra.

Chamigrane Sesquiterpenes from a Basidiomycetous Endophytic Fungus XG8D Associated with Thai Mangrove Xylocarpus granatum.

Project description:Six new chamigrane sesquiterpenes, merulinols A‒F (1‒6), and four known metabolites (7‒10) were isolated from the culture of the basidiomycetous fungus XG8D, a mangrove-derived endophyte. Their structures were elucidated mainly by 1D and 2D NMR, while the structures of 1 and 2 were further confirmed by single-crystal X-ray diffraction analysis. The in vitro cytotoxicity of all compounds was evaluated against three human cancer cell lines, MCF-7, Hep-G2, and KATO-3. Compounds 3 and 4 selectively displayed cytotoxicity against KATO-3 cells with IC50 values of 35.0 and 25.3 μM, respectively.

Evaluation of Indonesian mangrove Xylocarpus granatum leaves ethyl acetate extract as potential anticancer drug.

Project description:Local Xylocarpus granatum leaves were extracted by ethyl acetate solvent and characterized by TLC fingerprinting and 2D 1H NMR spectroscopy to contain phenolic compounds as well as several organic and amino acids as metabolic byproducts, such as succinic acid and acetic acid. Traces of flavonoids and other non-categorized phenolic compounds exhibited intermediate antioxidant activity (antioxidant IC50 84.93 ppm) as well as anticancer activity against HeLa, T47D, and HT-29 cell lines; which the latter being most effective against HT-29 with Fraction 5 contained the strongest activity (anticancer IC50 23.12 ppm). Extracts also behaved as a natural growth factor and nonlethal towards brine shrimps as well as human adipose-derived stem cell hADSC due to antioxidative properties. A stability test was performed to examine how storage conditions factored in bioactivity and phytochemical structure. Extracts were compared with several studies about X. granatum leaves extracts to evaluate how ethnogeography and ecosystem factored on biologically active compounds. Further research on anticancer or antioxidant mechanism on cancer cells is needed to determine whether the extract is suitable as a candidate for an anticancer drug.

DFT/TDDFT study of lanthanide(III) mono- and bisporphyrin complexes.

Project description:The electronic structure, molecular structure, and electronic spectra of lanthanide(III) mono- and bisporphyrin complexes are investigated using a DFT/TDDFT method. These complexes include YbP(acac), YbP(2), [YbP(2)](+), YbHP(2), and [YbP(2)](-) (where P = porphine and acac = acetylacetonate). To shed some light on the origin of the out-of-plane displacement of Yb in YbP(acac), unligated model systems, namely, planar D(4h) and distorted C(4nu) YbP, were calculated. For comparison, the calculations were also extended to include the C and [Ce(IV)P(2) ](+) systems. Even without an axial ligand, the lanthanide atom lies considerably above the porphyrin plane; the distortion of the YbP molecular structure from a planar D(4h) to the nonplanar C(4nu) symmetry leads to a considerable energy lowering. The axial ligand makes the metal out-of-plane displacement even larger, and it also changes the redox properties of the lanthanide monoporphyrin. The ground-state configurations of YbP(2) and YbHP(2) were determined by considering several possible low-lying states. YbP(2) is confirmed to be a single-hole radical. The special redox properties of the bisporphyrin complexes can well be accounted for by the calculated ionization potentials and electron affinities. The TDDFT results provide a clear description of the UV-vis and near-IR absorption spectra of the various lanthanide porphyrins.

Designing Thiadiazoloquinoxaline-Based Conjugated Polymers for Efficient Organic Photovoltaics: A DFT/TDDFT Study.

Project description:Clean and renewable energy development is becoming frontier research for future energy resources, as renewable energy offers sustainable and environmentally friendly alternatives to non-renewable sources such as fossil fuels. Among various renewable energy sources, tremendous progress has been made in converting solar energy to electric energy by developing efficient organic photovoltaics. Organic photovoltaic materials comprising conjugated polymers (CP) with narrow optical energy gaps are promising candidates for developing sustainable sources due to their potentially lower manufacturing costs. Organic semiconductor materials with a high electron affinity are required for many optoelectronic applications. We have designed a series of organic semiconductors comprised of cyclopentadithiophene as a donor and thiadiazoloquinoxaline (TQ) as an acceptor, varying the π-conjugation and TQ-derivatives. We have employed density functional theory (DFT) and time-dependent DFT (TDDFT) to evaluate the designed CP's optoelectronic properties, such as optical energy gap, dipole moment, and absorption spectra. Our DFT/TDDFT result shows that the energy gap of CPs is lowered and redshifted in the absorption spectra if there is no insertion of conjugation units such as thiophene and selenophene between donor and acceptor. In addition, selenophene shows relatively better redshift behavior compared to thiophene. Our work also provides rational insight into designing donor/acceptor-based CPs for organic solar cells.

Designing Potential Donor Materials Based on DRCN5T with Halogen Substitutions: A DFT/TDDFT Study.

Project description:Experimental researchers have found that the organic solar cell (OSC) based on DRCN5T (an oligothiophene) possesses excellent power conversion efficiency (PCE) of 10.1%. However, to date, there have been few studies about halogenation of DRCN5T, and its effects on photovoltaic properties of halogenated DRCN5T are still not clear. In the present work, we first perform benchmark calculations and effectively reproduce experimental results. Then, eight halogenated DRCN5T molecules are designed and investigated theoretically by using density functional theory (DFT) and time-dependent DFT. The dipole moments, frontier molecular orbital energies, absorption spectra, exciton binding energy (Eb), singlet-triplet energy gap (ΔEST), and electrostatic potential (ESP) of these molecules, and the estimated open circuit voltages (VOCs) of the OSCs with PC71BM as acceptor are presented. We find that (1) generally, halogen substitutions would increase VOC; (2) Eb rises with more fluorine substitutions, but for Cl and Br substitutions, Eb increases firstly and then drops; (3) ΔEST keeps increasing with more halogen substitutions; (4) except for Br substitutions, the averaged ESP arises along with more halogen substitutions; (5) the absorption strength of UV-Vis spectra of DRCN5T2F, DRCN5T4F, DRCN5T6F, and DRCN5T2Cl in the visible region is enhanced with respect to DRCN5T. Based on these results, overall, DRCN5T2Cl, DRCN5T4F, and DRCN5T6F may be promising donors.
