Solvent-controlled synthesis of multicolor photoluminescent carbon dots for bioimaging.

ABSTRACT: Multicolor fluorescent carbon dots (CDs) have potential applications in multichannel detection and multicolor imaging. In this study, multicolor fluorescent CDs were synthesized by changing the solvent type and adjusting the reactant ratio. The four prepared CDs emitted bright and stable blue (B-), green (G-), yellow (Y-), and red (R-) fluorescence under a single UV light (λ _ex = 365 nm). The photoluminescence (PL) emission wavelengths changed from 445 nm for B-CDs to 620 nm for R-CDs, and therefore covered the entire visible spectrum. The absolute quantum yields for the B-, G-, Y-, and R-CDs were 27.3%, 31.1%, 22.9%, and 8.8%, respectively. Characterization of the CDs showed that the differences among the optical features of the four prepared CDs arise from the differences among the surface states and nitrogen-derived structures in the carbon core. The four prepared CDs all showed low toxicity and steady PL, and therefore have potential applications in both in vitro and in vivo imaging.

Novel synthesis of fibronectin derived photoluminescent carbon dots for bioimaging applications.

Project description:Fibronectin (FN) derived from human plasma has been used for the first time as the carbon precursor in the top-down, microwave-assisted hydrothermal synthesis of nitrogen doped carbon dots (CDs). FN is a large glycoprotein primarily known for its roles in cell adhesion and cell growth. Due to these properties FN can be over expressed in the extracellular matrix (ECM) of some cancers allowing FN to be used as an indicator for the detection of cancerous cells over non-cancerous cells. These FN derived CDs display violet photoluminescence with UV excitation and appear to possess similar functional groups on their surface to their carbon precursor (-COOH and -NH2). This is believed to be due to the self-passivation of the CDs' nitrogen-containing surface functional groups during the heating process. These CDs were then used to stain MCF-7 and MDA-231 breast cancer cells and were observed to interact primarily with the cell membrane rather than intercalating into the cell like the many other types of CDs. This led to the hypothesis that the CDs are selectively binding to the FN overexpressed within the cancer cells' ECM via amide linkages. This is in agreement with the EDX and FTIR spectra of the FN CDs which indicate the presence of -COOH and nitrogen containing surface groups like -NH3. The inherent selectivity of the CDs combined with their ability to photoluminesce enables their use as a fluorophore for bioimaging applications.

Preparation of Multicolor Photoluminescent Carbon Dots by Tuning Surface States.

Project description:The achievements of multicolor photoluminescent (PL)-emissive carbon dots (CDs), particularly red to near infrared (NIR), are critical for their applications in optoelectronic devices and bioimaging, but it still faces great challenges to date. In this study, PL emission red-shifts were observed when tartaric acid (TA) was added into m-phenylenediamine (mPD) or o-phenylenediamine (oPD) solutions as carbon sources to prepare CDs, i.e., from blue to green for mPD and from yellow-green to red for oPD. Morphology and structure analyses revealed that the increased surface oxidation and carboxylation were responsible for the red-shifts of emission, indicating that TA played a key role in tuning the surface state of CDs. These factors could be employed as effective strategies to adjust PL emissions of CDs. Consequently, multicolor PL CDs (i.e., blue-, green-, yellow-green- and red-emissive CDs) can be facilely prepared using mPD and oPD in the absence and presence of TA. Particularly, the obtained red-emissive CDs showed a high PL quantum yield up to 22.0% and an emission covering red to NIR regions, demonstrating great potentials in optoelectronic devices and bioimaging. Moreover, multicolor phosphors were further prepared by mixing corresponding CDs with polyvinylpyrrolidone (PVP), among which the blue, green, and red ones could serve as three primary color phosphors for fabricating multicolor and white light-emitting diodes (LEDs). The white LED was measured to show a Commission Internationale de L'Eclairage (CIE) 1931 chromaticity coordinate of (0.34, 0.32), a high color rendering index (CRI) of 89, and a correlated color temperature (CCT) of 5850 K, representing one of the best performances of white LEDs based on CDs.

Multicolor Photoluminescent Carbon Dots a La Carte for Biomedical Applications.

Project description:Dual-emission fluorescence probes that provide high sensitivity are key for biomedical diagnostic applications. Nontoxic carbon dots (CDs) are an emerging alternative to traditional fluorescent probes; however, robust and reproducible synthetic strategies are still needed to access materials with controlled emission profiles and improved fluorescence quantum yields (FQYs). Herein, we report a practical and general synthetic strategy to access dual-emission CDs with FQYs as high as 0.67 and green/blue, yellow/blue, or red/blue excitation-dependent emission profiles using common starting materials such as citric acid, cysteine, and co-dopants to bias the synthetic pathway. Structural and physicochemical analysis using nuclear magnetic resonance, absorbance and fluorescence spectroscopy, Fourier-transform infrared spectroscopy, and X-ray photoelectron spectroscopy in addition to transmission electron and atomic force microscopy (TEM and AFM) is used to elucidate the material's composition which is responsible for the unique observed photoluminescence properties. Moreover, the utility of the probes is demonstrated in the clinical setting by the synthesis of green/blue emitting antibody-CD conjugates which are used for the immunohistochemical staining of human brain tissues of glioblastoma patients, showing detection under two different emission channels.

Synthesis of Multicolor Carbon Dots Based on Solvent Control and Its Application in the Detection of Crystal Violet.

Project description:The adjustment of the emitting wavelength of carbon dots (CDs) is usually realized by changing the raw materials, reaction temperature, or time. This paper reported the effective synthesis of multicolor photoluminescent CDs only by changing the solvent in a one-step solvothermal method, with 1,2,4,5-tetraaminobenzene as both the novel carbon source and nitrogen source. The emission wavelengths of the as-prepared CDs ranged from 527 to 605 nm, with quantum yields (QYs) reaching 10.0% to 47.6%, and it was successfully employed as fluorescence ink. The prepared red-emitting CDs (R-CDs, λem = 605 nm) and yellow-emitting CDs (Y-CDs, λem = 543 nm) were compared through multiple characterization methods, and their luminescence mechanism was studied. It was discovered that the large particle size, the existence of graphite Ns, and oxygen-containing functional groups are beneficial to the formation of long wavelength-emitting CDs. Y-CDs responded to crystal violet, and its fluorescence could be quenched. This phenomenon was thus employed to develop a detection method for crystal violet with a linear range from 0.1 to 11 µM and a detection limit of 20 nM.

Direct Synthesis of Multicolor Fluorescent Hollow Carbon Spheres Encapsulating Enriched Carbon Dots.

Project description:Multicolor fluorescent hollow carbon spheres (HCSs) are fabricated by an easy one-step route of in situ pyrolysis process with the use of natural scales and collagen powders as the precursor. The gas blow forming mechanism and photoluminescence (PL) emission mechanism of HCSs have been thoroughly discussed and proved that HCSs represent the first examples of three-dimensional multicolor fluorescent nanomaterials based on carbon dots (CDs). The HCSs encapsulate enriched carbon dots with high quantum yields (QYs) of 38%, and thus are applied in inkjet printing and sensitized solar cells. This strategy offers a promising avenue for preparing multicolor fluorescent hollow carbon materials on an industrial scale.

Solvent mediated synthesis of multicolor narrow bandwidth emissive carbon quantum dots and their potential in white light emitting diodes.

Project description:The development of efficient and environmentally sustainable materials for white light-emitting diodes (WLEDs) is of paramount importance in the field of lighting technology. In this study, we present a solvent-modulated synthesis approach for the fabrication of multicolor narrow-bandwidth emissive carbon quantum dots (CQDs) as a promising solution for constructing WLEDs. The synthesis method involves the controlled reaction of organic precursors in different solvent environments, leading to the formation of CQDs with distinct emission wavelengths with a relatively small full width at half maximum, ranging from 28 to 42 nm. Moreover, these synthesized multicolor CQDs demonstrate a remarkably high fluorescence quantum yield of up to 65%, indicating their potential for constructing efficient WLED when incorporated in polymer matrix and coated on the surface of blue light-emitting diode (LED).

Soybean-derived blue photoluminescent carbon dots.

Project description:Biomass-derived carbon dots (CDs) are biocompatible and have potential for a variety of applications, including bioimaging and biosensing. In this work, we use ground soybean residuals to synthesize carbon nanoparticles by hydrothermal carbonization (HTC), annealing at high temperature, and laser ablation (LA) in a NH4OH solution. The carbon nanoparticles synthesized with the HTC process (HTC-CDs) exhibit photoluminescent characteristics with strong blue emission. The annealing of the HTC-processed carbon particles in the range of 250 to 850 °C causes a loss of the photoluminescent characteristics of the CDs without any significant change in the microstructure (amorphous structure) of the carbon particles. The LA processing of the annealed HTC-processed carbon particles introduces nitrogen-containing surface-functional groups and leads to the recovery of the photoluminescent features that are different from those of the HTC-CDs and dependent on the fraction of nitrogen in the surface-functional groups. The photoluminescence of both the HTC-CDs and LA-CDs is largely due to the presence of N-containing surface-functional groups. The quantum yield of the LA-CDs is more constant than that of the HTC-CDs under continuous UV excitation and does not exhibit a significant reduction after 150 min of excitation. The methods used in this work provide a simple and green strategy to introduce N-surface-functional groups to carbon nanoparticles made from biomass and biowaste and to produce stable photoluminescent CDs with excellent water-wettability.

Multicolor polymeric carbon dots: synthesis, separation and polyamide-supported molecular fluorescence.

Project description:Multicolor carbon dots (CDs) have been developed recently and demonstrate great potential in bio-imaging, sensing, and LEDs. However, the fluorescence mechanism of their tunable colors is still under debate, and efficient separation methods are still challenging. Herein, we synthesized multicolor polymeric CDs through solvothermal treatment of citric acid and urea in formamide. Automated reversed-phase column separation was used to achieve fractions with distinct colors, including blue, cyan, green, yellow, orange and red. This work explores the physicochemical properties and fluorescence origins of the red, green, and blue fractions in depth with combined experimental and computational methods. Three dominant fluorescence mechanism hypotheses were evaluated by comparing time-dependent density functional theory and molecular dynamics calculation results to measured characteristics. We find that blue fluorescence likely comes from embedded small molecules trapped in carbonaceous cages, while pyrene analogs are the most likely origin for emission at other wavelengths, especially in the red. Also important, upon interaction with live cells, different CD color fractions are trafficked to different sub-cellular locations. Super-resolution imaging shows that the blue CDs were found in a variety of organelles, such as mitochondria and lysosomes, while the red CDs were primarily localized in lysosomes. These findings significantly advance our understanding of the photoluminescence mechanism of multicolor CDs and help to guide future design and applications of these promising nanomaterials.

Systematic safety evaluation on photoluminescent carbon dots.

Project description:Photoluminescent carbon dots (C-dots) were prepared using the improved nitric acid oxidation method. The C-dots were characterized by tapping-mode atomic force microscopy, and UV-vis absorption spectroscopy. The C-dots were subjected to systematic safety evaluation via acute toxicity, subacute toxicity, and genotoxicity experiments (including mouse bone marrow micronuclear test and Salmonella typhimurium mutagenicity test). The results showed that the C-dots were successfully prepared with good stability, high dispersibility, and water solubility. At all studied C-dot dosages, no significant toxic effect, i.e., no abnormality or lesion, was observed in the organs of the animals. Therefore, the C-dots are non-toxic to mice under any dose and have potential use in fluorescence imaging in vivo, tumor cell tracking, and others.
