Synthesis of uniform Prussian blue nanoparticles by a polyol process using a polyethylene glycol aqueous solution.

ABSTRACT: A polyol process was applied to the synthesis of Prussian blue nanoparticles that have a narrow size distribution. Potassium hexacyanidoferrate(iii) and iron(iii) nitrate aqueous solutions were introduced into a 50% polyethylene glycol (PEG) aqueous solution under magnetic stirring at 50 °C and reacted for 48 h. The shape of the so-obtained particles was cubic with somewhat rounded edges and the mean size was 70 nm. In the formation process, nanoparticles of Prussian green, which is a partially oxidized state of Prussian blue, were firstly generated via reduction of the precursors by PEG molecules. The Prussian green nanoparticles were then reduced subsequently to Prussian blue nanoparticles. Rate constants for both the reduction steps have been estimated using the time evolution of absorbance.

More Efficient Prussian Blue Nanoparticles for an Improved Caesium Decontamination from Aqueous Solutions and Biological Fluids.

Project description:Any release of radioactive cesium-137, due to unintentional accidents in nuclear plants, represents a dangerous threat for human health and the environment. Prussian blue has been widely studied and used as an antidote for humans exposed to acute internal contamination by Cs-137, due to its ability to act as a selective adsorption agent and to its negligible toxicity. In the present work, the synthesis protocol has been revisited avoiding the use of organic solvents to obtain Prussian blue nanoparticles with morphological and textural properties, which positively influence its Cs+ binding capacity compared to a commercially available Prussian blue sample. The reduction of the particle size and the increase in the specific surface area and pore volume values compared to the commercial Prussian blue reference led to a more rapid uptake of caesium in simulated enteric fluid solution (+35% after 1 h of contact). Then, after 24 h of contact, both solids were able to remove >98% of the initial Cs+ content. The Prussian blue nanoparticles showed a weak inhibition of the bacterial luminescence in the aqueous phase and no chronic detrimental toxic effects.

Human Serum Albumin Protein Corona in Prussian Blue Nanoparticles.

Project description:Prussian Blue nanoparticles (PBnps) are now popular in nanomedicine thanks to the FDA approval of PB. Despite the numerous papers suggesting or describing the in vivo use of PBnps, no studies have been carried out on the formation of a protein corona on the PBnp surface and its stabilizing role. In this paper, we studied qualitatively and quantitatively the corona formed by the most abundant protein of blood, human serum albumin (HSA). Cubic PBnps (41 nm side), prepared in citric acid solution at PB concentration 5 × 10-4 M, readily form a protein corona by redissolving ultracentrifuged PBnp pellets in HSA solutions, with CHSA ranging from 0.025 to 7.0 mg/mL. The basic decomposition of PBnp@HSA was studied in phosphate buffer at the physiological pH value of 7.4. Increased stability with respect to uncoated PBnps was observed at all concentrations, but a minimum CHSA value of 3.0 mg/mL was determined to obtain stability identical to that observed at serum-like HSA concentrations (35-50 mg/mL). Using a modified Lowry protocol, the quantity of firmly bound HSA in the protein corona (hard corona) was determined for all the CHSA used in the PBnp@HSA synthesis, finding increasing quantities with increasing CHSA. In particular, an HSA/PBnp number in the 1500-2300 range was found for CHSA 3.0-7.0 mg/mL, largely exceeding the 180 HSA/PBnp value calculated for an HSA monolayer on a PBnp. Finally, the stabilization brought by the HSA corona allowed us to carry out pH-spectrophotometric titrations on PBnp@HSA in the 3.5-9-0 pH range, revealing a pKa value of 6.68 for the water molecules bound to the Fe3+ centers on the PBnp surface, whose deprotonation is responsible for the blue-shift of the PBnp band from 706 nm (acidic solution) to 685 nm (basic solution).

Plasmonic Imaging of Electrochemical Reactions at Individual Prussian Blue Nanoparticles.

Project description:Prussian blue is an iron-cyanide-based pigment steadily becoming a widely used electrochemical sensor in detecting hydrogen peroxide at low concentration levels. Prussian blue nanoparticles (PBNPs) have been extensively studied using traditional ensemble methods, which only provide averaged information. Investigating PBNPs at a single entity level is paramount for correlating the electrochemical activities to particle structures and will shed light on the major factors governing the catalyst activity of these nanoparticles. Here we report on using plasmonic electrochemical microscopy (PEM) to study the electrochemistry of PBNPs at the individual nanoparticle level. First, two types of PBNPs were synthesized; type I synthesized with double precursors method and type II synthesized with polyvinylpyrrolidone (PVP) assisted single precursor method. Second, both PBNPs types were compared on their electrochemical reduction to form Prussian white, and the effect from the different particle structures was investigated. Type I PBNPs provided better PEM sensitivity and were used to study the catalytic reduction of hydrogen peroxide. Progressively decreasing plasmonic signals with respect to increasing hydrogen peroxide concentration were observed, demonstrating the capability of sensing hydrogen peroxide at a single nanoparticle level utilizing this optical imaging technique.

In situ IR spectroscopy during oxidation process of cobalt Prussian blue analogues.

Project description:Cobalt Prussian blue analogues (Co-PBA; NaxCo[Fe(CN)6]y), consisting of cyano-bridged transition metal network, -Fe-CN-Co-NC-Fe-, are promising cathode materials for Na-ion secondary batteries. In the oxidation process, oxidization of Fe and/or Co are compensated by Na+ deintercalation. Here, we investigated the oxidization process of three Co-PBAs by means of in situ infrared absorption (IR) spectroscopy. With use of an empirical rule of the frequencies of the CN- stretching mode in ferrocyanide ([FeII(CN)6]4-) and ferricyanide ([FeIII(CN)6]3-), the oxidation processes of Co-PBAs were determined against the Fe concentration (y) and temperature (T). We will discuss the interrelation between the oxidation processes and Fe concentration (y).

Thallium Labeled Citrate-Coated Prussian Blue Nanoparticles as Potential Imaging Agent.

Project description:BackgroundThe aim of this study was to develop and characterize a nanoparticle-based image-contrast platform which is biocompatible, chemically stable, and accessible for radiolabeling with 201Tl. We explored whether this nanoparticle enhanced the T1 signal which might make it an MRI contrast agent as well.MethodsThe physical properties of citrate-coated Prussian blue nanoparticles (PBNPs) (iron(II);iron(III);octadecacyanide) doped with 201Tl isotope were characterized with atomic force microscopy, dynamic light scattering, and zeta potential measurement. PBNP biodistribution was determined by using SPECT and MRI following intravenous administration into C57BL6 mice. Activity concentrations (MBq/cm3) were calculated from the SPECT scans for each dedicated volume of interest (VOI) of liver, kidneys, salivary glands, heart, lungs, and brain.ResultsPBNP accumulation peaked at 2 hours after injection predominantly in the kidneys and the liver followed by a gradual decrease in activity in later time points.ConclusionWe synthetized, characterized, and radiolabeled a Prussian blue-based nanoparticle platform for contrast material applications. Its in vivo radiochemical stability and biodistribution open up the way for further diagnostic applications.

Manganese-containing Prussian blue nanoparticles for imaging of pediatric brain tumors.

Project description:Pediatric brain tumors (PBTs) are a leading cause of death in children. For an improved prognosis in patients with PBTs, there is a critical need to develop molecularly-specific imaging agents to monitor disease progression and response to treatment. In this paper, we describe manganese-containing Prussian blue nanoparticles as agents for molecular magnetic resonance imaging (MRI) and fluorescence-based imaging of PBTs. Our core-shell nanoparticles consist of a core lattice structure that incorporates and retains paramagnetic Mn(2+) ions, and generates MRI contrast (both negative and positive). The biofunctionalized shell is comprised of fluorescent avidin, which serves the dual purpose of enabling fluorescence imaging and functioning as a platform for the attachment of biotinylated ligands that target PBTs. The surfaces of our nanoparticles are modified with biotinylated antibodies targeting neuron-glial antigen 2 or biotinylated transferrin. Both neuron-glial antigen 2 and the transferrin receptor are protein markers overexpressed in PBTs. We describe the synthesis, biofunctionalization, and characterization of these multimodal nanoparticles. Further, we demonstrate the MRI and fluorescence imaging capabilities of manganese-containing Prussian blue nanoparticles in vitro. Finally, we demonstrate the potential of these nanoparticles as PBT imaging agents by measuring their organ and brain biodistribution in an orthotopic mouse model of PBTs using ex vivo fluorescence imaging.

Polyethylene Glycol-Modified Layered Double Hydroxides: Synthesis, Characterization, and Study on Adsorption Characteristics for Removal of Acid Orange II from Aqueous Solution.

Project description:The present study aimed to improve the adsorption characteristics of the pristine layered double hydroxide (LDH) by physicochemical modification using polyethylene glycol (PEG400), a nontoxic hydrophilic polymer. With this objective, LDH was synthesized and modified with different concentrations of PEG400. The PEG-modified LDHs (LDH/PEGs) were characterized using X-ray diffraction, thermogravimetric analysis, Brunauer-Emmett-Teller surface area and porosity measurement, scanning electron microscopy, transmission electron microscopy, Fourier transform infrared spectroscopy, and zeta potential measurements. The adsorption properties of the pristine LDH (PLDH) and the LDH/PEGs were studied for the removal of Acid Orange II from water, and the results were compared. The PLDH treated with 15% PEG solution showed ∼30% increase in adsorption capacity as compared to the PLDH. The adsorption isotherm data were analyzed using Langmuir, Freundlich, and Temkin isotherm models. The values of thermodynamic parameters such as ΔS and ΔH showed the spontaneous and endothermic nature of the adsorption process. The adsorption kinetics data for both PLDH and the LDH/PEG adsorbents presented a good fit to the pseudo-second-order kinetic model.

Photochemical transformations of thiolated polyethylene glycol coatings on gold nanoparticles.

Project description:Photochemical reactions can cause significant transformations of manufactured nanomaterials in sunlit environments. While transformations of inorganic nanoparticles (NPs) have been investigated extensively, less attention has been focused on the direct impact of aqueous photochemical reactions on adsorbed organic macromolecules that form the NP corona and strongly influence the surface interactions and reactivity that affect NP transport, fate, and toxicity. Here, we assess the transformations of methoxy polyethylene glycol thiol (mPEGSH) coatings on gold NPs (AuNPs) under controlled UV irradiation. A decrease in the adsorbed layer thickness of polymer was observed within 24 h of UV irradiation, resulting in increased susceptibility of the transformed NPs to aggregation. Surface chemistry analyses, including X-ray photoelectron spectroscopy (XPS), showed loss of the ether groups but persistence of reduced S on the AuNP surface, indicative of a chain scission mechanism yielding different NP surface properties from that of either the initial PEGylated AuNP or the citrate-stabilized AuNP prior to coating with mPEGSH. The transformation of the chemisorbed polymer was compared to that of dissolved mPEGSH in the presence and absence of the Au NPs, as evaluated by liquid chromatography - mass spectrometry (LC-MS). In contrast to the NP-adsorbed coating, the primary observed transformation of the dissolved mPEGSH was thiol oxidation to disulfides without extensive chain scission. This study demonstrates that transformations of adsorbed macromolecular coatings must be considered to accurately predict NP attachment behavior, and hence transport, in environmental systems. Because the corona transformation was not predictable from that of the dissolved polymer, direct NP surface characterization is required to discern the fundamental reactions involved in the photochemical transformation of coatings after sorption to the NP surface.

Management of polyethylene glycol solution aspiration using bronchoscopic lavage in a dog.

Project description:Polyethylene glycol lavage solutions are used for colonic preparation in dogs and are considered relatively safe. Aspiration is an uncommon but potentially devastating complication of polyethylene glycol administration. Full recovery is possible and often rapid in people treated with bronchoalveolar lavage. A healthy 2-year-old male Beagle used in an endoscopy teaching laboratory aspirated a small amount of polyethylene glycol lavage solution. Although initially appearing unaffected, the dog quickly became hypoxemic. Bronchoscopy was used to lavage the lungs and aspirate tracheal/pulmonary fluid 5 times over the course of 45 minutes. The dog completely recovered. This report presents the successful treatment of polyethylene glycol aspiration in a dog. Although the seriousness of aspiration might not be immediately evident, bronchoscopy and lavage should be pursued because of the rapidly progressive nature of polyethylene glycol-induced pulmonary edema.

Stable cycling of Prussian blue/Zn battery in a nonflammable aqueous/organic hybrid electrolyte.

Project description:Although rechargeable aqueous batteries are attracting increasing attention in recent years due to high safety, low cost, high power density and environmental friendliness, the aqueous batteries suffer from limited cycle life due to a narrow electrochemical window of the aqueous electrolytes, severe side reaction and instability of electrode materials in aqueous electrolytes. In this work, we propose a hybrid aqueous electrolyte with a mixed solvent of water and acetonitrile (ACN), which exhibits a wide electrochemical window, high ionic conductivity, and nonflammability. An aqueous battery with an iron hexacyanoferrate (FeHCF) cathode, Zn anode and H2O/ACN hybrid electrolyte shows a high capacity of 69.1 mA h g-1 at 10C (89.5% relative to that at 1C) and an extremely long cycle life with 51.4% capacity retention after 19 000 cycles at 10C. The excellent cycling performance of the aqueous FeHCF/Zn batteries can be attributed to the reduced water activity and extended electrochemical window because of the strong hydrogen-bonding interaction between ACN and H2O. Besides, the large particle size and good crystallization of FeHCF can inhibit its dissolution in the aqueous electrolyte which further improves cycling performance. This work will shed light on the design of safe aqueous batteries for applications in large-scale energy storage.
